This study investigates the use of pretreatment and enzymatic hydrolysis side streams and conversion to lignocellulose nanofibers. We used a steam-exploded and partial enzymatic hydrolyzed hardwood pulp and an organosolv pretreated softwood pulp to prepare lignocellulose nanofibers (LCNF) via microfluidization. The energies applied on fibrillation were estimated to examine the energy consumption levels of LCNF production. The energy consumptions of the fibrillation processes of the hardwood LCNF production and the softwood LCNF production were about 7040-14080 kWh/ton and 4640 kWh/ton on a dry material basis, respectively. The morphology and dimension of developed hardwood and softwood LCNFs and the stability and rheological behavior of their suspensions were investigated and are discussed.

John Clare observed and described the natural world with an unsurpassed accuracy and intimacy. But his landscapes also bore the memories of life and labour. Like Wordsworth, he sought to create textual objects in transmissible forms, to deliver their reported worlds – expansive, dynamic, somehow inhabitable – to distant readers, drawing them into sympathetic intercommunion with a complex living scene. His intimate descriptive poetry reveals the tangible qualities of light and sound, and the material basis of the apparently abstract concept of time. Memory and imagination are understood to inhabit bodily spaces, provoking ‘real transport’: an observer lost in – and to – the moment. From his place and time, Clare felt solidarity with isolated birds, alienation from labour, estrangement from human communities. Publications such as annuals often showcased formulaic reflections on nature and on memory; Clare exploited textual duplicability, his meditative descriptive poetry spanning the history and futurity of an observed scene.

Two forms or rather perspectives of observations appear alongside practice theories: The first perspective can be called the „theatre perspective“: practice here is observed as a regular, spatiotemporally ordered, socially structured, and therefore recognizable historical form of „practical doings and sayings“, in which participants are understood as mere carriers of practices and their bodies as the raw material for processes of formation. In the other perspective, understood as the perspective of the participants themselves, practices come into view as ongoing, conflictual, and contingent accomplishments, in which participants occur as intelligently collaborating contributors with so called „lived bodies“. These bodies are affectable, sites of experience, and media of a sensitivity that allow an embodied self to orientate itself (with)in a practice. This paper proposes a methodological mediation of both perspectives by taking into account both a sociological analysis of discipline, formation, or adjustment, and the reflexive sensing in action, which can be modeled phenomenologically. Thus, a „lived-body-in-accomplishment“ comes into view that serves the material basis of subjectivation procceses, i. e. the (self-)formation of a constitutionally conditioned (political) agency.

BackgroundThe degree of immune infiltration in tumors, especially CD8+ T cells, greatly impacts patient disease course and response to interventional immunotherapy. Hence, enhancement of TIL prevalence is a preferred clinical endpoint, one that may be achieved via administration of agents that normalize the tumor vasculature (VN) leading to improved immune cell recruitment and/or that induce the development of local tertiary lymphoid structures (TLS) within the tumor microenvironment (TME).MethodsLow-dose STING agonist ADU S-100 (5 μg/mouse) was delivered intratumorally to established s.c. B16.F10 melanomas on days 10, 14 and 17 post-tumor inoculation under an IACUC-approved protocol. Treated and control, untreated tumors were isolated at various time points to assess transcriptional changes associated with VN and TLS formation via qPCR, with corollary immune cell composition changes determined using flow cytometry and immunofluorescence microscopy. In vitro assays were performed on CD11c+ BMDCs treated with 2.5 μg/mL ADU S-100 (vs PBS control) and associated transcriptional changes analyzed via qPCR or profiled using DNA microarrays. For TCRβ-CDR3 analyses, CDR3 was sequenced from gDNA isolated from enzymatically digested tumors and splenocytes.ResultsWe report that activation of STING within the TME leads to slowed melanoma growth in association with increased production of angiostatic factors including Tnfsf15 (Vegi), Cxcl10 and Angpt1, and TLS inducing factors including Ccl19, Ccl21, Lta, Ltb and Tnfsf14 (Light). Therapeutic responses from intratumoral STING activation were characterized by increased vascular normalization (VN), enhanced tumor infiltration by CD8+ T cells and CD11c+ DCs and local TLS neo-genesis, all of which were dependent on host expression of STING. Consistent with a central role for DC in TLS formation, ex vivo ADU S-100-activated mCD11c+ DCs also exhibited upregulated expression of TLS promoting factors including lymphotoxin-α (LTA), IL-36, inflammatory chemokines and type I interferons. TLS formation was associated with the development of a therapeutic TIL TCR repertoire enriched in T cell clonotypes uniquely detected within the tumor but not the peripheral circulation in support or local T cell cross-priming within the TME.ConclusionsOur data support the premise that i.t. delivery of STING agonist promotes a pro-inflammatory TME in support of VN and TLS formation, leading to the local expansion of unique TIL repertoire in association with superior anti-melanoma efficacy.

Protein is an important component of all the cells and tissues of the human body and is the material basis of life. Its content, sequence, and spatial structure have a great impact on proteomics and human biology. It can reflect the important information of normal or pathophysiological processes and promote the development of new diagnoses and treatment methods. However, the current techniques of proteomics for protein analysis are limited by chemical modifications, large sample sizes, or cumbersome operations. Solving this problem requires overcoming huge challenges. Nanopore single molecule detection technology overcomes this shortcoming. As a new sensing technology, it has the advantages of no labeling, high sensitivity, fast detection speed, real-time monitoring, and simple operation. It is widely used in gene sequencing, detection of peptides and proteins, markers and microorganisms, and other biomolecules and metal ions. Therefore, based on the advantages of novel nanopore single-molecule detection technology, its application to protein sequence detection and structure recognition has also been proposed and developed. In this paper, the application of nanopore single-molecule detection technology in protein detection in recent years is reviewed, and its development prospect is investigated.
