In this study the effect of N, P and K nutrients on the Cu and Fe content of winter wheat (Triticum aestivum L.) grains was investigated in a long-term fertilization experiment set up in Nagyhörcsök. Samples were also harvested from four experimental stations of the Hungarian national long-term fertilization trials. These are the following: Bicsérd, Iregszemcse, Karcag, and Putnok. Plant samples were collected in 2005 which was very wet. Our results from Nagyhörcsök were compared with the Cu and Fe content of samples which were harvested from control plots of other experimental stations. The Cu and Fe content of grain samples were measured using inductively coupled plasma mass spectrometer (ICP-MS) followed by digestion with HNO3-H2O2 solution. All data were subjected to ANOVA, and when significant differences (P<0.05) were detected, Duncan’s test was performed to allow separation of means. The main conclusions are as follows: Cu and Fe content of wheat grains was higher and higher in every NPK treatments. Samples were harvested from the control plots of Iregszemcse and Bicsérd have higher Cu content than the treated samples from Nagyhörcsök.

We investigated the anther culture (AC) efficiency of thirteen F<sub>4</sub> combinations of winter wheat (Triticum aestivum L.). The genotype dependency was assessed during the induction of the androgenic entities, i.e. embryo-like structures (ELS), regenerated-, green-, albino-, and transplanted plantlets. The number of green plantlets per 100 anthers (GP/100A) varied from 0.36 to 24.74 GP/100A with a mean of 8.31 GP/100A. Albino plantlets (AP) occurred in each combination, ranging from 0.20 to 22.80 AP/100A with an average value of 5.59 AP/100A. Between 25–87.76 doubled haploid (DH) plants per 100 acclimatised plantlets (DH/100ADP), depending on the combination, with a mean of 59.74% were recovered. We have found the highest DH production in the combinations Béres/Midas, Kalász/Tacitus, Béres/Pamier, and Premio/5009. This improves remarkably the choice of basic genetic material in subsequent crossing programmes. These observations emphasise the usability and efficiency of in vitro AC in producing a large number of DH lines for breeding and the applied researches of winter wheat. Although albinism was found in each combination, it was mitigated by the in vitro AC application.

Aim. To analyze the efficiency of using tissues of immature and mature embryos for Agrobacterium-mediated transformation in vitro of new breeding-valuable genotypes of winter wheat (Triticum aestivum L.) for the purpose of their genetic improvement. Methods. Culture of in vitro, extraction and electrophoresis of DNA, PCR analysis. Results. The efficiency of induction of callusogenesis and regeneration of winter wheat shoots was analyzed. The morphogenetic response of callus cultures obtained from different explants under Agrobacterium-mediated transformation in vitro was investigated. Molecular genetic analysis of wheat regenerants for transgenes was performe. Conclusions. The tissues of immature and mature embryos of novel breeding-valuable wheat genotypes are competent explants for Agrobacterium-mediated transformation in vitro. In this case, the tissues of two daily sprouts of mature wheat germ are characterized by higher morphogenetic parameters, which helps to obtain a greater percentage of genetically modified variants.
