Anthocyanins belong to the group of flavonoid compounds broadly distributed in plant species responsible for attractive colors. In black rice (Oryza sativa L.), they are present in the stems, leaves, stigmas, and caryopsis. However, there is still no scientific evidence supporting the existence of compartmentalization and trafficking of anthocyanin inside the cells. In the current study, we took advantage of autofluorescence with anthocyanin’s unique excitation/emission properties to elucidate the subcellular localization of anthocyanin and report on the in planta characterization of anthocyanin prevacuolar vesicles (APV) and anthocyanic vacuolar inclusion (AVI) structure. Protoplasts were isolated from the stigma of black and brown rice and imaging using a confocal microscope. Our result showed the fluorescence displaying magenta color in purple stigma and no fluorescence in white stigma when excitation was provided by a helium–neon 552 nm and emission long pass 610–670 nm laser. The fluorescence was distributed throughout the cell, mainly in the central vacuole. Fluorescent images revealed two pools of anthocyanin inside the cells. The diffuse pools were largely found inside the vacuole lumen, while the body structures could be observed mostly inside the cytoplasm (APV) and slightly inside the vacuole (AVI) with different shapes, sizes, and color intensity. Based on their sizes, AVI could be grouped into small (Ф < 0.5 um), middle (Ф between 0.5 and 1 um), and large size (Ф > 1 um). Together, these results provided evidence about the sequestration and trafficking of anthocyanin from the cytoplasm to the central vacuole and the existence of different transport mechanisms of anthocyanin. Our results suggest that stigma cells are an excellent system for in vivo studying of anthocyanin in rice and provide a good foundation for understanding anthocyanin metabolism in plants, sequestration, and trafficking in black rice.

Field experiments were conducted to investigate methods of controlling red rice (Oryza sativaL. ♯ ORYSA) in drill-seeded rice (O. sativa). Treatments included the rice cultivar ‘Mars', coated with calcium peroxide (CaO2) at 40% (w/w) and a crop protectant, R-33865 (O,O-diethyl-O-phenyl phosphorothioate) at 0.5 and 1% (v/w). Molinate (S-ethyl hexahydro-1H-azepine-1-carbothioate) at 6.7 kg ai/ha was applied preplant incorporated (ppi). The land was flooded (2.5 to 5 cm deep) after seeding with rice (100 kg/ha, 2.5 cm deep), and the water was maintained throughout the growing season. CaO2, with or without molinate, increased rice grain yield 50% and increased rice culm density fivefold above untreated rice. Molinate applied ppi controlled 96% of the red rice. Rice seed coated with only CaO2or with CaO2plus R-33865 at 0.5%, each combined with ppi molinate, produced 5690 and 6030 kg/ha of grain, respectively. These high yields were associated with red rice control by molinate and good stands of rice provided by O2supplied by CaO2. R-33865 applied to rice seed at 1% (v/w) injured rice by reducing rice culm densities 41%, compared with rice without protectant.

A series of on-farm fi eld experiments were conducted in two locations in Padang, Koto Panjang and Koto Tingga, West Sumatera, Indonesia in 2015. The purpose of the experiment was to establish the formula of organic fertilizer derived from Tithonia supplied with micro nutrients, Zn and Mn, on rice. The experiments were conducted using completely-randomized block design with six treatments and three replications. The treatments were P = Tithonia Organic Fertilizer Plus (TOFP) + 3.0 kg Mn.ha-1, Q = TOFP + 3.0 kg Mn.ha-1+ 3.0 kg Zn.ha-1, R = TOFP + 4.5 kg Mn.ha-1 + 6 kg Zn.ha-1, S= TOFP + 4.5 kg Mn.ha-1+ 9 kg Zn.ha-1, T = TOFP only, U = 100% chemical fertilizer only. Treatment with micro nutrients as addition to TOFP (TOFP+ 3.0 kg Mn.ha-1 and TOFP+ 3.0 kg Mn.ha-1+3.0 kg Zn.ha-1) increased rice grain yield by 80 g per clump.

Rice (Oryza sativa L.) is one of the important cereal crop grown extensively in Gujarat. It suffers from grain discoloration fungal disease causing heavy losses at maturity stage of rice. A field experiment was laid out with the single and new combinations fungicides against rice grain discoloration disease under field conditions during kharif-2013-14, 2014-15 and 2015-16. Six fungicides at various concentrations were screened against rice grain discoloration disease. The trifloxystrobin 25% + tebuconazole 50% (0.03 %) @ 0.4 g l-1 and propiconazole 25 EC (0.025 %) @ 1.0 ml l-1 was found effective against rice grain discoloration and recorded significantly higher healthy grains.

Crops and stored grains are susceptible to pathogens that represent a threat to our health. The study presented herein compares the normal surface and endophytic fungal communities present on white and brown rice grains. One hundred grains of each rice variety was analyzed to determine their fungal contaminants and endophytes. Fungi were inoculated on SDA media, and purified in PDA media; morphological characterization was performed followed by amplification of the ITS region using PCR for all fungal isolates. Statistical analysis indicated significant differences between medium brown rice compared to white rice for surface and endophytic communities (p-value £ 0.05). In addition, a higher fungal diversity was found on brown rice grains compared to white rice. This variation may be due to differences in the processing methods used for each rice grain type. BLAST analysis revealed the presence of toxigenic strains of Aspergillus flavus, A.oryzae, Penicillium verrucosum, and P. viridicatum. The study of fungal growth in rice grains can contribute to the minimization of mycotoxin production by its prevention and control; therefore, decreasing crop contamination and human exposure to their metabolites. Strength properties of grains have a significant impact on the energy demand of grinding mills. This paper presents the results of tests of strength and energy needed the for destruction of rice grains. The research aim was to experimentally determine mechanical and processing properties of the rice grains. The research problem was formulated in the form of questions: (1) what force and energy are needed to induce a rupture of rice grain of the Oryza sativa L. of long-grain variety? (2) what is the relationship between grain size and strength parameters and the energy of grinding rice grain of the species Oryza sativa L. long-grain variety? In order to find the answer to the problems posed, a static compression test of rice grains was done. The results indicate that the average forces needed to crush rice grain are 174.99 kg m·s−2, and the average energy is 28.03 mJ. There was no statistically significant relationship between the grain volume calculated based on the volumetric mass density Vρ and the crushing energy, nor between the volume Vρ and other strength properties of rice grains. In the case of Vs, a low negative correlation between strength σmin and a low positive correlation between the power inducing the first crack were found for the grain size related volume. A low negative correlation between the grain thickness a3, stresses σmin and work WFmax was found as well as a low positive correlation between thickness a3 and the force inducing the first crack Fmin.
