Abstract Red algae, belonging to the phylum Rhodophyta, contain an abundance of useful chemicals including bioactive molecules and present opportunities for the production of different products through biorefinery cascades. The rhodophyte Palmaria palmata, commonly termed dulse or dillisk, grows predominantly on the northern coasts of the Atlantic and Pacific Oceans and is a well-known snack food. Due to its abundance, availability and cultivation capacity, P. palmata was selected for study as a potential candidate for a biorefinery process. In addition to studying juice and solid fractions of freshly harvested P. palmata, we have investigated the novel possibility of preserving algal biomass by ensilaging protocols similar to those employed for terrestrial forage crops. In the metabolite partitioning within the solid and liquid fractions following screw-pressing, the majority of the metabolites screened for—water soluble carbohydrates, proteins and amino acids, lipids, pigments, phenolics and antioxidant activity—remained in the solid fraction, though at differing proportions depending on the metabolite, from 70.8% soluble amino acids to 98.2% chlorophyll a and 98.1% total carotenoids. For the ensiling study, screw-pressed P. palmata, with comparative wilted and chopped, and chopped only samples, were ensiled at scale with and without Safesil silage additive. All samples were successfully ensiled after 90 days, with screw-pressing giving lower or equal pH before and after ensiling compared with the other preparations. Of particular note was the effluent volumes generated during ensiling: 26–49% of the fresh weight, containing 16–34% of the silage dry matter. This may be of advantage depending on the final use of the biomass.

Silage inoculants consisting of primarily Lactobacillus plantarum, are widely used to ensure that lactic acid bacteria dominate the fermentation of water soluble carbohydrates (WSC) during the ensilage process. Previous studies have shown that the supplementation of ensiled forage crops with cellulases can also improve the quality of silage through i) increasing the generation of WSC, and therefore ensuring an adequate supply of substrate for L. plantanim; ii) Partial hydrolysis of the plant cell wall increasing the rate of cellulose hydrolysis within the rumen. From the above discussion it is apparent that the use of an L.plantarum strain, with the capacity to hydrolyse cellulose, could be beneficial in the ensiling process. No celluloytic lactic bacterium has been isolated from microbial ecosystems. However, the advent of recombinant DNA technology affords us the possibility of engineering a cellulolytic derivative of L. plantarum. This report describes progress towards this objective.

Thalli of three species of Plagiochasma, collected during different seasons from their natural habitats of Mandi region of Himachal Pradesh were evaluated for some biochemical parameters viz. total water soluble carbohydrates, proteins, free amino acids, total chlorophyll, carotenoid and the activities of amylases, invertase and protease. Highly significantly contents of carbohydrates (70.83±2.54 mg/g fw in P. appendiculatum, 21.26± 0.09 mg/g fw in P. articulatum and 52.75±2.95 mg/g fw in P. intermedium)  and of chlorophyll (0.76±0.005 mg/g fw in P. appendiculatum,  0.69±0.005 mg/g fw in P. articulatum and 1.2±0.006 mg/g fw in P. intermedium ) were observed towards the end of the growing season (January-March period of collection), whereas the content of protein (23.46±0.14 mg/g fw in P. appendiculatum, 23.33±0.71 mg/g fw in P. articulatum and 22.99±0.27 mg/g fw in P. intermedium)  was maximum during winter (October-December) and that of free amino acids (37.48±1.05 mg/g fw in P. appendiculatum, 70.9±0.91 mg/g fw in P. articulatum and 25.13±0.31 mg/g fw in P. intermedium)  in the rainy season (July-September). On the other hand, the activities of enzymes that breakdown the carbohydrates into simple sugars were recorded least towards the end of the favourable period of their growth. The activity of protease was maximum in the rainy season (July-September) and minimum in the winter season (October-December). This study concluded that the seasonal changes in Plagiochasma induced alterations in the biochemical compounds and in the activities of related enzymes that may be responsible for the adaptation of these plants in their natural habitats.

Microbial protein contributes about two-thirds of the amino acids absorbed by ruminants. Information on the proportion of bacterial and protozoal N passing to the duodenum would enhance our understanding of the effect of diet not only on microbial protein synthesis but also on the contribution of bacteria and protozoa to duodenal flow of others metabolites of interest. However, differentiation of the duodenal fractions has proven to be difficult because routine procedures cannot separate microbial protein at the duodenum into bacteria and protozoa (Punia et al., 1992). Molecular techniques that use group-specific rRNA-targeted probes may help overcome these problems. The objective of this experiment was to quantify the flow of protozoal N at the duodenum in steers, fed two silage diets differing in water soluble carbohydrates (WSC) content, by real-time PCR using protozoa specific primers.

In pot trial the biochemical composition and phosphorus use efficiency of birdsfoot trefoil, sainfoin and subterranean clover grown pure and in mixtures with perennial ryegrass in the next ratios were studied in the Institute of Forage Crops, Pleven, Bulgaria: birdsfoot trefoil + perennial ryegrass (50:50%); sainfoin + perennial ryegrass (50:50%); subterranean clover + perennial ryegrass (50:50%); birdsfoot trefoil + subterranean clover + perennial ryegrass (33:33:33%); sainfoin + subterranean clover + perennial ryegrass (33:33:33%). The highest crude protein content was found in the aboveground mass of birdsfoot trefoil (19.17%) and sainfoin (19.30%). The water soluble sugars contents in mixtures was found higher compared to the pure grown legumes. Birdsfoot trefoil showed the highest phosphorus use efficiency for plant biomass accumulation and nodules formation. In mixtures the phosphorus use efficiency was found be higher as compared to the same in pure grown legumes.

The objective was to study the effects of sugar cane molasses addition on the fermentation quality and tastes of alfalfa silage. Fresh alfalfa was ensiled with no additive (Control), 1% molasses (M1), 2% molasses (M2), and 3% molasses (M3) for 206 days. The chemical composition and fermentation characteristics of the alfalfa silages were determined, the microbial communities were described by 16S rRNA sequencing, and the tastes were evaluated using an electronic tongue sensing system. With the amount of added molasses (M), most nutrition (dry matter and crude protein) was preserved and water-soluble carbohydrates (WSC) were sufficiently used to promote the fermentation, resulting in a pH reduction from 5.16 to 4.48. The lactic acid (LA) content and LA/acetic acid (AA) significantly increased, indicating that the fermentation had turned to homofermentation. After ensiling, Enterococcus and Lactobacillus were the dominant genus in all treatments and the undesirable microbes were inhibited, resulting in lower propionic acid (PA), butyric acid (BA), and NH3-N production. In addition, bitterness, astringency, and sourness reflected tastes of alfalfa silage, while umami and sourness changed with the amount of added molasses. Therefore, molasses additive had improved the fermentation quality and tastes of alfalfa silage, and the M3 group obtained the ideal pH value (below 4.5) and the best condition for long-term preservation.

AbstractCyanophycin (multi-l-arginyl-poly-l-aspartic acid; also known as cyanophycin grana peptide [CGP]) is a biopolymer that could be used in various fields, for example, as a potential precursor for the synthesis of polyaspartic acid or for the production of CGP-derived dipeptides. To extend the applications of this polymer, it is therefore of interest to synthesize CGP with different compositions. A recent re-evaluation of the CGP synthesis in C. glutamicum has shown that C. glutamicum is a potentially interesting microorganism for CGP synthesis with a high content of alternative amino acids. This study shows that the amount of alternative amino acids can be increased by using mutants of C. glutamicum with altered amino acid biosynthesis. With the DM1729 mutant, the lysine content in the polymer could be increased up to 33.5 mol%. Furthermore, an ornithine content of up to 12.6 mol% was achieved with ORN2(Pgdh4). How much water-soluble or insoluble CGP is synthesized is strongly related to the used cyanophycin synthetase. CphADh synthesizes soluble CGP exclusively. However, soluble CGP could also be isolated from cells expressing CphA6308Δ1 or CphA6308Δ1_C595S in addition to insoluble CGP in all examined strains. The point mutation in CphA6308Δ1_C595S partially resulted in a higher lysine content. In addition, the CGP content could be increased to 36% of the cell dry weight under optimizing growth conditions in C. glutamicum ATCC13032. All known alternative major amino acids for CGP synthesis (lysine, ornithine, citrulline, and glutamic acid) could be incorporated into CGP in C. glutamicum.
