SummaryOptical imaging has long been considered a method for histological or microscopic investigations. Over the last 15 years, however, this method was applied for preclinical molecular imaging and, just recently, was also able to show its principal potential for clinical applications (e.g. fluorescence-guided surgery). Reviewing the development and preclinical evaluation of new fluorescent dyes and target-specific dye conjugates, these often show characteristic patterns of their routes of excretion and biodistribution, which could also be interesting for the development and optimization of radiopharmaceuticals. Especially ionic charges show a great influence on biodistribution and netcharge and charge-distribution on a conjugate often determines unspecific binding or background signals in liver, kidney or intestine, and other organs.Learning from fluorescent probe behaviour in vivo and translating this knowledge to radio-pharmaceuticals might be useful to further optimize emerging and existing radiopharmaceuticals with respect to their biodistribution and thereby availability for binding to their targets.

Abstract Growing water demand poses a challenge for supply. Poor understanding of alternative sources can hamper plans for addressing water scarcity and supply resilience. The potential of three alternative supply systems in Lilongwe, Malawi and Sharm El-Sheikh, Egypt are compared using a fast, data-light assessment approach. Lilongwe water supply is based on unsustainable use of source water, while Sharm depends primarily on desalination. Both locations experience shortages due to poor system performance and service inequity. Alternative supply systems are shown to potentially contribute to supply augmentation/diversification, improving service and system resilience. There are considerable seasonal variations to consider, especially regarding storage of water. Social preferences could limit the uptake/demand for alternative water. One important conclusion is the value in addressing public perceptions of alternative systems, and assessing water end use in order to site systems appropriately. Other issues surround financing, encouraging uptake and addressing institutional/governance aspects surrounding equitable distribution. A further consideration is whether demand reductions might yield shorter-term improvements in performance without the need to institute potentially expensive alternative water strategies. Reducing non-revenue water is a priority. Such measures should be undertaken with alternative supply enhancement to reduce inequity of supply, improve system performance and increase resilience to future changes.

Abstract During the summer of 1994, we compared the physical and nutrient characteristics of the three main tributaries of Cootes Paradise: Spencer, Chedoke and Borer’s creeks. On all sampling occasions, concentrations of CHL α and nutrients were always lowest in Borer’s Creek and highest in Chedoke Creek. There were generally 10-fold higher CHL α concentrations and 2 to 10 times higher levels of nitrogen and phosphorus in Chedoke Creek compared with Spencer Creek. Despite this, the light environment did not differ significantly between Spencer and Chedoke creeks because the low algal biomass in Spencer Creek was balanced by a relatively high loading of inorganic sediments from the watershed. Laboratory experiments indicated that sediments from Chedoke Creek released up to 10 µg/g of soluble phosphorus per gram (dry weight) of sediment, compared with only 2 µg/g from Spencer Creek. By contrast, sediment samples from Spencer Creek contained levels of polycyclic aromatic hydrocarbon that were as high as or higher than those from Chedoke Creek, and much higher than those found in Borer’s Creek. The distribution of normalized PAH concentrations suggests a common source of PAHs in all three tributaries, most likely automobile exhaust, since there were high concentrations of fluoranthene and pyrene, both of which are derivatives of engine combustion.

This book is about what metaphors mean and do within Buddhist texts. More specifically, it is about the fundamental Buddhist ambivalence toward language, which is seen as obstructive and yet necessary for liberation, as well as the ingenious response to this tension that one Buddhist philosophical school—the early Indian Yogācāra (3rd–6th century CE)—proposed by arguing that all language use is in fact metaphorical (upacāra). Exploring the profound implications of this claim, the book presents the full-fledged Yogācāra theory of meaning—one that is not merely linguistic, but also perceptual.Despite the overwhelming visibility of figurative language in Buddhist philosophical texts, its role and use have received relatively little attention in scholarship to date. This book is the first sustained and systematic attempt to present an indigenous Buddhist philosophical theory of metaphor. By grounding the Yogācāra’s pan-metaphorical claim in its broader intellectual context, both Buddhist and non-Buddhist, the discussion reveals an intense Indian philosophical conversation about metaphor and language that reached across sectarian lines, and it also demonstrates its potential contribution to contemporary philosophical discussions of related topics. The analysis of this theory of metaphor radically reframes the Yogācāra controversy with the Madhyamaka; sheds light on the school’s application of particular metaphors, as well as its unique understanding of experience; and establishes the place of Sthiramati as an original Buddhist thinker of note in his own right, alongside Asaṅga and Vasubandhu.

A matter raised consistently by eminent personalities asked to report on atrocities in Africa, such as former South African President Thabo Mbeki, is the utilization of traditional justice mechanisms known to Africans. Their use has been limited to Gacaca courts in Rwanda, set up in haste and subject to much criticism. However, there exist several types and models of traditional justice mechanisms at the African level. The contribution of these sui generis mechanisms towards accountability for heinous crimes is largely unaddressed in academic literature. This chapter intends to fill this gap by exploring their potential contribution towards accountability for heinous crimes, alongside the International Criminal Court (ICC). In short, the chapter explores how community-based judicial mechanisms and the ICC, two types of accountability mechanisms with different methodologies and approaches, can work side by side to eradicate impunity regarding, and also to prevent, mass atrocities on the African continent.

Both RNA-binding proteins (RBPs) and translation are increasingly implicated in several neurodegenerative diseases, but their specific roles in promoting disease are not yet fully defined. This chapter critically evaluates the evidence that altered translation of specific mRNAs mediated by RNA-binding proteins plays an important role in driving specific neurodegenerative diseases. First, diseases are discussed where a causal role for RNA-binding proteins in disease appears solid, but whether this involves altered translation is less clear. The main foci here are TAR DNA-binding protein (TDP-43) and fused in sarcoma (FUS) in amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Subsequently, diseases are presented where altered translation is believed to contribute, but involvement of RNA-binding proteins is less clear. These include Huntington’s and other repeat expansion disorders such as fragile X tremor/ataxia syndrome (FXTAS), where repeat-induced non-AUG-initiated (RAN) translation is a focus. The potential contribution of both canonical and non-canonical RBPs to altered translation in Parkinson’s disease is discussed. The chapter closes by proposing key research frontiers for the field to explore and outlining methodological advances that could help to address them.

Seven taxa of raptorial birds were experimentally fed a controlled sample of 50 house mice (Mus musculus). Bones recovered from the pellets were examined for interspecies variability in preservation to assess the potential contribution of specific raptors to patterning in fossil assemblages. Quantitative analyses demonstrate that patterns in bone fragmentation may assist in the identification of particular raptor species as depositional agents in small mammal assemblages.
