Sambiloto mengandung flavonoid dan andrografolid yang mempunyai efek anti alergi. Penelitian ini bertujuan untuk mengetahui Pengaruh Ekstrak Sambiloto (Andrographis paniculata [Burm f.] Ness) terhadap hitung Eosinofil Serum Mencit Model Alergi Inflamasi. Jenis penelitian ini adalah eksperimental laboratorik, dengan post test only control group design. Hewan uji berupa 15 ekor mencit jantan. Mencit dibagi menjadi tiga kelompok, masing-masing terdiri dari 5 ekor. Kelompok I (kontrol) hanya diberi diet standar. Kelompok II diberi diet standar dan ekstrak sambiloto per oral dengan dosis 10mg/20 gram BB/hari. Kelompok III diberi diet standar dan antihistamin per-oral dengan dosis 0,4 mg/20 gram BB/hari. Sensitisasi mencit dengan ovalbumin (OVA). Hari ke-4 dan ke-20 dipapar OVA i.p. Dosis 0,15 cc/mencit. OVA inhalasi ke-37 secara intraorbital. Analisis data dengan uji Anova menggunakan program SPSS for Windows Release 11.5. Hasil penelitian menunjukkan rata-rata eosinofil mencit kelompok I = 0,94% kelompok II = 0,87% dan kelompok III = 1,2%. Dengan analisa statistik kelompok II dibandingkan kelompok I, kelompok III dibandingkan kelompok I dan kelompok II dibanding kelompok III menunjukkan perbedaan hitung eosinofil yang tidak bermakna (p>0,05). Ekstrak sambiloto dengan dosis 10 mg/20gram BB/hari dapat menurunkan hitung eosinofil serum pada mencit model alergi inflasi.

The effect of dietary lead exposure on immunomodulation of broilers and possible ameliorative effect of Andrographis paniculata were studied. One hundred broiler chicks were randomly divided into five equal groups of 20 each. Group T0 served as control while group T1, T2, T3 and T4 birds were provided commercial diet with lead acetate at the dose of 300 ppm in feed. Powder of dry leaves of Andrographis paniculata was mixed in the feed of group T2, T3 and T4 chicks at the dose of 2 gm, 3 gm and 4 gm/kg of feed respectively and fed for 4 weeks. Significant decrease in HI titres in T1 group and significant increase in T4 group suggested increased haemagglutination titre (HI) response. The maximum skin thickness and diameter on 24th and 48th hours was observed in control group birds (T0) and was comparable with T4 group. Microscopically spleen, thymus and bursa of fabricius revealed increase in lymphoid population in birds fed lead acetate along with Andrographis paniculata compared to T1 group. We concluded that Andrographis paniculata has immunomodulatory effect during lead toxicity in broilers.

Abstract Objectives Andrographis paniculata tablets (AS201-01) have previously been shown to have potent bioactivity as an antimalarial and to produce no unwanted side effects in animal models. Here, we present the phase 1 clinical trial conducted to evaluate the safety of AS201-01 tablets in healthy volunteers. Methods The study was a randomized, double-blind controlled cross-over, a placebo-controlled design consisting of a 4-day treatment of AS201-01 tablets. A total of 30 healthy human volunteers (16 males and 14 females) were divided into two groups, and each group was given 4 tablets, twice daily for 4 days. Group 1 received AS201-01, while group 2 received placebo tablets. Volunteers were given a physical examination before the treatment. The effects of AS201-01 on random blood glucose, biochemical, and hematological as well as urine profiles were investigated. Results There were no changes in observed parameters as a result of AS201-01 being administered. Statistical analysis showed no significant difference (p>0.05) between the test and control group regarding hematology profile, biochemical profile, and random blood glucose. Increased appetite and better sleep, which categorized as grade 1 adverse event was reported after treatment with AS201-01 tablet Conclusions The outcome supports our previous observation that the AS201-01 tablet, given twice a day for 4 days, is safe and nontoxic.

Abstract Background and aims Andrographis paniculata is an annual herbaceous plant which belongs to the Acanthaceae family. Extracts from this plant have shown hepatoprotective, anti-inflammatory and antidiabetic properties, at least in part, through activation of the nuclear receptor Peroxisome Proliferator-Activated Receptor-gamma (PPAR γ). Recent evidence has demonstrated that activation of PPARγ reduces alcohol drinking and seeking in Marchigian Sardinian (msP) alcohol-preferring rats. Methods The present study evaluated whether A. paniculata reduces alcohol drinking and relapse in msP rats by activating PPARγ. Results Oral administration of an A. paniculata dried extract (0, 15, 150 mg/kg) lowered voluntary alcohol consumption in a dose-dependent manner and achieved ~65% reduction at the dose of 450 mg/kg. Water and food consumption were not affected by the treatment. Administration of Andrographolide (5 and 10 mg/kg), the main active component of A. paniculata, also reduced alcohol drinking. This effect was suppressed by the selective PPARγ antagonist GW9662. Subsequently, we showed that oral administration of A. paniculata (0, 150, 450 mg/kg) prevented yohimbine- but not cues-induced reinstatement of alcohol seeking. Conclusions Results point to A. paniculata-mediated PPARγactivation as a possible therapeutic strategy to treat alcohol use disorder.
