Polychlorinated biphenyls (PCBs) are a ubiquitous class of environmental pollutants with toxic and hepatocellular effects, including accumulation of fat, proliferated smooth endoplasmic recticulum (SER), and concentric membrane arrays (CMAs) (1-3). The CMAs appear to be a membrane storage and degeneration organelle composed of a large number of concentric membrane layers usually surrounding one or more lipid droplets often with internalized membrane fragments (3). The present study documents liver alteration after a short term single dose exposure to PCBs with high chlorine content, and correlates them with reported animal weights and central nervous system (CNS) measures. In the brain PCB congeners were concentrated in particular regions (4) while catecholamine concentrations were decreased (4-6). Urinary levels of homovanillic acid a dopamine metabolite were evaluated (7).Wistar rats were gavaged with corn oil (6 controls), or with a 1:1 mixture of Aroclor 1254 and 1260 in corn oil at 500 or 1000 mg total PCB/kg (6 at each level).

The contamination of poultry tissues by total organochlorine (OC) pesticides and polychlorinated biphenyls (PCBs) in Slovenia was examined from a public health perspective. As part of systematic veterinary and sanitary monitoring of foods of animal origin from 1997 to 1999, 87 samples of fat tissue were examined for the presence of α-HCH, lindane, heptachlor, and total DDT (plus DDE and DDD) and 88 samples of fat tissue were examined for the presence of total PCBs (sum of PCB congeners). The levels of poultry tissue contamination in six regions of Slovenia were determined. Levels of OC pesticides were below detection limits in 86.2% of analyses performed. Total PCB levels for most samples (59.1%) of poultry fat tissue were in the 0.002- to 0.02-mg/kg range. The level of contamination with OC pesticides and total PCBs in poultry fat tissue was not influenced by the type of poultry and was the highest in samples from industrial areas, which have been ecologically disturbed. All samples met Slovenian and European Union tolerances.

The biological half-life [Formula: see text] of 31 dichloro- to decachloro-biphenyl congeners were monitored for 105 d in adult rainbow trout (Salmo gairdneri) that were exposed to a single oral dose. In whole fish [Formula: see text] increased from 5 d to no apparent elimination as the number of chlorines on the biphenyl increased. This structure–activity relationship was not as evident in muscle where [Formula: see text] ranged from < 5 to 127 d. We suggest the decline in muscle resulted from decreasing lipid levels and the redistribution of congeners within the fish. From structure–activity analysis of [Formula: see text] in whole fish we conclude that elimination is enhanced for those congeners with lower chlorine content, with no chlorine substitutions in the ortho positions, and those with two unsubstituted carbons that are adjacent (vicinal) on the biphenyl. A significant decline in total PCB content in whole fish, equivalent to a [Formula: see text] of 219 d, was partly due to the composition of the PCB mixture administered, and the selective elimination of the lower chlorinated biphenyls.

Cells of Pseudomonas strain LB400, grown on biphenyl, glucose, or glycerol, transformed polychlorinated biphenyl (PCB) congeners into chlorobenzoic acid (CBA) metabolites. Transformation of the PCB congeners, 2,3-chlorobiphenyl (CBP), 2,2'-CBP, 2,5,4'-CBP, and 2,4,2',4'-CBP, produced the metabolites, 2,3-CBA, 2-CBA, 4-CBA, and 2,4-CBA, respectively. Rates and extents of PCB transformation and metabolite formation were highest with biphenyl-grown cells. Intermediate rates of metabolite production were observed with glycerol-grown cells, and lowest rates of production were found with glucose-grown cells. Regardless of carbon source, the rate of degradation of congeners was faster than the rate of production of CBAs. Relative rates of PCB transformation and metabolite production from different congeners with cells grown on a particular substrate followed the same general order, 2,3-CBA (from 2,3-CBP) > 2-CBA (from 2,2'-CBP) > 4-CBA (from 2,5,4'-CBP) > 2,4-CBA (from 2,4,2',4'-CBP). Pseudomonas strain LB400 appeared unable to grow on any of the chlorobenzoic acids. However, Pseudomonas strain LB400 cells grown on biphenyl appeared capable of degrading 2-CBA and 2,3-CBA but not 4-CBA nor 2,4-CBA. Cells grown on glycerol appeared unable to metabolize any CBAs.Key words: polychlorinated biphenyls, metabolites, Pseudomonas LB400.

Abstract A previously published gel permeation chromatographic (GPC) procedure for determination of organophosphorous insecticides is used to determine 24 organochlorine (OC) pesticide and 9 polychlorinated biphenyl (PCB) residues in 8 fatty preserved foods of vegetable and animal origin. Samples are extracted with methylene chloride and cleaned up by automated GPC with a Biobeads SX3 column and a methylene chloride–cyclohexane (15 + 85) eluant. Compounds are quantitated by gas chromatography/mass spectrometry (GC/MS) with selected ion monitoring using a DB-5 column. Average recoveries of OC pesticides from samples fortified at 0.010–0.080 mg/kg ranged from 73 to 98%. Average recoveries of 9 PCB congeners spiked at 2 levels (0.0025 and 0.020 mg/kg) were between 85 and 104%. Quantitation limits were between 0.002 and 0.025 mg/kg, depending on the compound. The purified extracts were analyzed further by GC with electron capture detection (ECD), and the results were compared with those obtained by MS. Determination of some OC pesticides and PCB congeners was not possible by ECD because of matrix interference.
