Increased oxidative stress and inflammatory markers contrasting with the activation of the cholinergic anti-inflammatory pathway in patients with metabolic syndrome

AbstractBeneficial effects of probiotics have been reported on body weight, lipid and carbohydrate metabolism, inflammatory state and oxidative stress in healthy subjects and in many metabolic and inflammatory diseases. The aim of this study was to evaluate the effects ofBifidobacterium lactisHN019 on inflammatory state and nitro-oxidative stress in patients with and without the metabolic syndrome (MetS). The usual diets of the thirty-three subjects were supplemented with probiotic milk for 90 d. Inflammatory markers and oxidative measurements were performed. In relation to the baseline values, subjects in both groups showed a decrease in homocysteine (P=0·02 andP=0·03, respectively), hydroperoxides (P=0·02 andP=0·01, respectively) and IL-6 levels (P=0·02). Increases in adiponectin (P=0·04) and nitric oxide metabolites (NOx,P=0·001) levels were only seen in the group with the MetS in relation to the baseline values, whereas only the individuals without the MetS had increases in total radical-trapping antioxidant parameter levels (P=0·002). In conclusion,B. lactisHN019 have several beneficial effects on inflammatory and oxidative biomarkers in healthy subjects and the MetS patients. Patients with the MetS showed a specific improvement in adiponectin and NOx levels, whereas a specific favourable effect was shown in the antioxidant defenses in healthy subjects. If the results obtained in the present study are confirmed, supplementation of fermented milk with probiotics in healthy subjects and patients with the MetS must be further discussed.

AbstractBackgroundThe metabolic syndrome (MetS) is an obesity-driven disorder with pandemic proportions and limited treatment options. Oxidative stress, low-grade inflammation and altered autonomic regulation, are important components of MetS pathophysiology. We recently reported that galantamine, an acetylcholinesterase inhibitor and an FDA-approved drug (for Alzheimer’s disease) alleviates the inflammatory state in MetS subjects. Here we examined the effects of galantamine on oxidative stress in parallel with inflammatory and cardio-metabolic parameters in subjects with MetS.MethodsThe effects of galantamine treatment, 8 mg daily for 4 weeks, followed by 16 mg daily for 8 weeks or placebo were studied in randomly assigned subjects with MetS (n=22 per group) of both genders. Oxidative stress, including superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase activities, lipid and protein peroxidation, and nitrite levels were analyzed before and at the end of the treatment. In addition, plasma cytokine and adipokine levels, insulin resistance (HOMA-IR) and other relevant cardio-metabolic indices were analyzed. Autonomic regulation was also examined by heart rate variability (HRV) before treatment, and at every 4 weeks of treatment.ResultsGalantamine treatment significantly increased antioxidant enzyme activities, including SOD (+1.65 USOD/mg protein, [95% CI 0.39 to 2.92], P=0.004) and CAT (+0.93 nmol/mg, [95% CI 0.34 to 1.51], P=0.011), decreased lipid peroxidation (thiobarbituric acid reactive substances, -5.45 pmol/mg, [95% CI -10.97 to 0.067], P=0.053) and systemic nitrite levels (-0.05 nit/mg protein, [95% CI -0.21 to 0.10], P=0.038) compared with placebo. In addition, galantamine significantly alleviated the inflammatory state and insulin resistance, and decreased the low frequency/high frequency ratio of HRV, following 8 and 12 weeks of drug treatment.ConclusionLow-dose galantamine alleviates oxidative stress, alongside beneficial anti-inflammatory, and metabolic effects, and modulates autonomic regulation in subjects with MetS. These findings are of considerable interest for further studies with galantamine to ameliorate MetS pathophysiology.

Metabolic syndrome is an emerging global threat as a major health burden. It is widely presumed that Metabolic syndrome is associated with a low grade chronic inflammatory phenomenon. This inflammatory state is due to the imbalance between the pro and anti-inflammatory cytokines. Studies have been performed on various inflammatory markers in metabolic syndrome like hsCRP, TNF-alpha, Adiponectin, IL-6, IL-10. Articles were chosen from indexed journals from various search engines. Pro inflammatory cytokines like hsCRP, TNF – alpha, Interleukin -6 were found to be increased and anti-inflammatory cytokines like Interleukin – 10 were reduced in metabolic syndrome.

Metabolic syndrome (MetS) is a risk factor for cognitive deterioration and frailty in older adults. In this regard it has been shown that oxidative stress (OxS) and chronic inflammation are involved in the pathophysiology of these alterations. Harmless antioxidant and anti-inflammatory therapeutic alternatives have been proposed, such as the consumption of Sechium edule (chayote), but the evidence is inconclusive. For this reason, an exploratory study of a single group chosen by convenience sampling, including 12 older adults, with an average age of 71 ± 6 years (10 women and 2 men) with a diagnosis of MetS according to the National Cholesterol Education Program Adult Treatment Panel III (NCEP/ATP III) criteria. This exploratory study aimed to determine the effect of the consumption of the dried fruit powder supplement of Sechium edule var. nigrum spinosum (500 mg, 3 times per day) for six weeks on the markers of OxS in elderly adults with MetS. All participants’ OxS markers were measured before and after treatment. There was a statistically significant decrease in the concentration of lipoperoxides (baseline, 0.289 ± 0.04 vs. post-treatment, 0.234 ± 0.06 μmol/L, p < 0.05), together with a significant increase in total antioxidant status (baseline, 0.97 ± 0.18 vs. post-treatment, 1.2 ± 0.12 mmol/L, p < 0.05). In this sense, the oxidative stress index showed a statistically significant decrease (baseline, 1.7 ± 0.78 vs. post-treatment, 0.75 ± 0.87, p < 0.05). A statistically significant decrease in the concentration of TNF-α after treatment was also found (baseline, 5.3 ± 1.4 vs. post-treatment, 3.5 ± 1.3, p < 0.05).Our findings suggest that the consumption of the dry fruit of Sechium edule has an antioxidant and anti-inflammatory effect in older adults with metabolic syndrome.

The present study aims to investigate the protective effect of bees’ honey against metanil-yellow-induced hepatotoxicity in rats. Rats were divided into 7 groups: control group; three groups treated with 50, 100, and 200 mg/kg metanil yellow, and three groups treated with metanil yellow plus2.5 mg·kg-1·day-1bees’ honey for 8 weeks. The obtained data showed that the antioxidant/anti-inflammatory activity of bees’ honey reduced the oxidative stress in the liver tissue and downregulated the inflammatory markers. In addition, the elevated levels of AGE and the activated NF-κB in the metanil-yellow-treated animals were significantly attenuated. Moreover, the levels of TNF-αand IL-1βwere significantly attenuated as a result of bees’ honey administration. Furthermore, the histopathological examination of the liver showed that bees’ honey reduced fatty degeneration, cytoplasmic vacuolization, and necrosis in metanil-yellow-treated rats. In conclusion, the obtained data suggest that bees’ honey has hepatoprotective effect on acute liver injuries induced by metanil-yellowin vivo, and the results suggested that the effect of bees’ honey against metanil yellow-induced liver damage is related to its antioxidant/anti-inflammatory properties which attenuate the activation of NF-κB and its controlled genes like TNF-αand IL-1β.

Objective: This investigation was undertaken to determine whether a grape seed extract (GSE) that is rich in mono-, oligo- and poly- meric polyphenols would modify postprandial oxidative stress and inflammation in individuals with the metabolic syndrome (MetS).Background: MetS is known to be associated with impaired glucose tolerance and poor glycemic control. Consumption of a meal high in readily available carbohydrates and fat causes postprandial increases in glycemia and lipidemia and markers of oxidative stress, inflammation and insulin resistance.Materials/methods: After an overnight fast, twelve subjects with MetS (5 men and 7 women) consumed a breakfast meal high in fat and carbohydrate in a cross-over design. A GSE (300 mg) or placebo capsule was administrated 1 hr before the meal (-1 hr). Changes in plasma insulin, glucose, oxidative stress and inflammatory markers were measured hourly for 6 hr. Results: Plasma hydrophilic oxygen radical absorbance capacity (ORAC) measured as the positive incremental area under the curve (-1 to 5 hr) was significantly increased when the meal was preceded by GSE compared with placebo (P<0.05). Meal-induced increases in oxidized LDL were attenuated with GSE (P<0.05). Plasma glucose concentrations (area under the curve from -1 to 5 hr) were also significantly lower when the meal was preceded by GSE (P<0.05) while the insulin concentrations remained unchanged (P>0.05). No changes in inflammatory markers were evident. Conclusion: These data suggest that GSE enhances postprandial plasma antioxidant status and reduces the glycemic response to a meal, high in fat and carbohydrate in subjects with the MetS.
