Aim: We aimed to explore ceruloplasmin (CP) expression in clear cell renal cell carcinoma (ccRCC). Materials & methods: CP was analyzed in biofluid samples of 63 ccRCC patients, divided into three grading groups, and immunohistochemically, in 308 ccRCC. Results: Significant differences of mean plasma and urine CP levels in different grading groups were found. CP immunoreactivity was significantly linked to high-grade disease. Log rank tests showed a significant shorter overall survival rate in CP-positive cases (all p < 0.05). Conclusion: CP protein levels in biofluid samples confirmed differential CP expressions, depending on nuclear grade in ccRCC as previously seen in RNA expression analysis. CP expression was linked to high-grade disease and reduced survival rate in RCC.

Abstract Background Fever, as an adverse event, is well documented in a wide array of drugs including multiple tyrosine kinase inhibitors however, it is not a previously well described consequence of the novel multi-targeted tyrosine kinase inhibitor, cabozantinib. Case presentation In this paper we document the first detailed review of high-grade fevers in a 54 year old male (Caucasian) with a background of metastatic clear cell renal cell carcinoma recently commenced on cabozantinib. After detailed investigation, we exclude infection and other common causes of fever as the causative agent and further, definitively resolve the recurrent fever by ceasing cabozantinib and starting a short course of oral corticosteroids. Conclusions We have demonstrated that cabozantinib should always be considered in the aetiology of high-grade fever in relevant patients. Further, we demonstrate that temporary cessation of cabozantinib and a course of short-term steroids can induce resolution of fever and allow for recommencement of cabozantinib safely thereafter.

Abstract Background To develop a hypoxia-associated long non-coding (lncRNA) signature for risk stratification in clear cell renal cell carcinoma (ccRCC). Methods The Cancer Genome Atlas Kidney Renal Clear Cell Carcinoma (TCGA KIRC) dataset was downloaded and analyzed. Results Four prognostic hypoxia-associated lncRNAs were used for signature construction. The four lncRNAs were downregulated in high grade, advanced stage, and high-risk ccRCC. ccRCC patients in the high-risk group had a worse prognosis than those in the low-risk group. And the risk score was significantly higher in high grade and advanced stage. The signature had an independent and long-standing prognosis prediction ability up to 10-year follow-up. Notably, the risk score was significantly positively correlated with the infiltration abundances of six immune cells from the Tumor IMmune Estimation Resource (TIMER). The gene set enrichment analysis (GSEA) also suggested that the signature was involved in the metabolism and tumorigenesis, which were closely related to the hypoxic tumor microenvironment. Ultimately, a nomogram was built to predict the individual long-term survival possibility. Conclusions We have developed a new hypoxia-associated lncRNA signature, representing a promising tool for risk stratification tool in ccRCC. It might serve as a prognostic index to facilitate personalized counseling for treatment.
