The root of pasak bumi (Eurycoma longifolia, Jack) is one of plant from Indonesia known as aphrodisiac. The study about its standardized extract as aphrodisiac was show increasing libido, testosterone level, FSH and LH on male rats. This study was conducted to evaluate testosterone level and histopathological changes of the testes of male rats of standardized extract of pasak bumi root. There are 50 old male rats which ages 3-4 months old divided to five groups. Group I as negative control was administered aquadestilata. Group II as positive control was administered testosterone (Andriol®). Group III, IV and V ware given standardized extract of pasak bumi root at the dose 50, 100 and 200 mg/kg body weight Respectively. The extract was given orally twice a day for six days and forty nine days and then the testes was taken out on 7th and 50th day. The histopathological of the testes was evaluated using quantitative parameter by the presence of testes weight, number and diameter of Leydig cells. Testosterone level was taken on 7th and 50th day used ELISA method. The results were analyzed using parametric test, one way ANOVA (P<0,05), post hoc Dunnet (2-sided) (P < 0,05). The result of this study shows that the standardized extract of pasak bumi root was not effect on histopathological changes of the testes but it could increased the level of testosterone m at the dose 100 and 200 mg/kg body weight.  Key word : Eurycoma longifolia, Jack, aphrodisiac, histopathological testes testosteron

Eurycoma longifolia Jack (Fam.: Simaroubaceae), known as Tongkat Ali (TA), has been known as a symbol of virility and sexual power for men. Metabolic profiling of the aqueous extract of E. longifolia (AEEL) using UPLC-MS/MS in both positive and negative modes allowed the identification of seventeen metabolites. The identified compounds were classified into four groups: quassinoids, alkaloids, triterpenes, and biphenylneolignans. AEEL is considered safe with oral LD50 cut-off >5000 mg/kg. Oral administration of 50, 100, 200, 400, or 800 mg/kg of AEEL for 10 consecutive days to Sprague-Dawley male rats caused significant reductions in mounting, intromission, and ejaculation latencies and increased penile erection index. AEEL increased total body weight and relative weights of seminal vesicles and prostate. Total and free serum testosterone and brain cortical and hippocampal dopamine content was significantly elevated in treated groups with no significant effects on serotonin or noradrenaline content.

Four herbal prescription medicines, Chi-Pao-Mei-Jan-Tan, Gui-Fu-Ba-Wei-Wan, Huan-Shao-Tan; and San Tsai-Feng-Sui-Tan, were tested for their effects on sexual behavior in aged rats. Crude liquid extracts of these herbs were administered to the rats daily through oral tubing for 14 days. All four herbal prescriptions showed some effects in restoration of mount and intromission behaviors, but there was no effect on restoration of ejaculation in 26 month old rats that had exhibited no copulatory activity (no mount, intromission and ejaculation) previously. The effects of Chi-Pao-Mei-Jan-Tan were further tested in 26 month old rats with low mount and intromission activities but without ejaculation behavior, and in 15 month old rats (middle-age group) that showed normal mount and intrornission behavior but no ejaculation activity. Chi-Pao-Mei-Jan-Tan was effective in improving the frequency of both mount and intromission, but failed to restore the ejaculation activity of the old rats with low mount and intromission behaviors. It was, however, very effective in restoration of ejaculation activity in middle-aged rats that exhibited normal mount and intromission behaviors. Serum testosterone (T) levels of Chi-Pao-Mei-Jan- Tan in tested old and middle-aged rats were determined by radioimmunoassay, and showed no difference before and after treatment. Our findings demonstrated that the four herbal prescriptions had some effects in restoration of mount and intromission behaviors, but not ejaculation activity in old rats, and that Chi-Pao-Mei-Jan-Tan was very effective in restoration of ejaculation activity in middle-aged rats. The promotional effect of Chi-Pao-Mei-Jan-Tan on copulatory behavior was not correlated with serum T levels.

Background: Raphia vinifera (Arecaceae) is a medicinal plant commonly used as a sexual enhancer. Objective: To investigate the aphrodisiac potential of aqueous extract (AE) and methanolic extract (ME) of R. vinifera in sexually experienced male rats. Materials and Methods: Thirty male Wistar rats were randomly distributed into six groups (5 rats per group) and administered for 14 days with distilled water (10 ml/kg), sildenafil citrate (1.44 mg/kg), and AE or ME of R. vinifera (100 or 500 mg/kg). The copulatory activity was tested on days 0, 7, and 14 using receptive females. Further, on day 14, rats were sacrificed and biochemical analyses (testosterone, total protein, and acid phosphatase) were performed. Results: Sildenafil citrate significantly decreased the intromission latency (day 14, p = 0.04) and frequency (days 7 and 14, p = 0.03) but increased the mount frequency (day 14, p = 0.04), compared with control. Remarkably, R. vinifera enhanced the sexual activity by significantly decreasing the intromission latency (AE and ME, 500 mg/kg, day 14, p = 0.04) and increasing the mount frequency (AE and ME, 100 mg/kg, day 7, p = 0.02) compared with control. Moreover, R. vinifera improved plasmatic (AE, 100 mg/kg, p = 0.03; AE, 500 mg/kg, p = 0.001; ME, 100 mg/kg, p = 0.01) and testicular (AE, 100 mg/kg, p = 0.001; AE, 500 mg/kg, p = 0.01; ME, 100 mg/kg, p = 0.001; ME, 500 mg/kg, p = 0.01) testosterone levels as well as plasmatic total proteins concentration (ME, 500 mg/kg, p = 0.04). Conclusion: These findings showed that R. vinifera possesses an aphrodisiac property which could further justify its folkloric use in traditional medicine as a sexual enhancer.

Eurycoma longifolia has been known for its aphrodisiac effects in male. Our previous study showed that with the dosage of 8 mg/kg bodyweight (BW) Eurycoma longifolia root extract increased plasma total testosterone levels in male rats when given for 14 days. Oestrogen is a potent inhibitor for testosterone production and spermatogenesis. The aim of the present study is to determine the effects of Eurycoma longifolia (8 mg/kg BW) on the spermatogenic cell count and sperm count of testosterone-suppressed male rats. Adult male Sprague-Dawley healthy male rats weighed 200–250 g were treated with, either control vehicle (no active ingredients given), oestradiol (500 mg/kg BW), Eurycoma longifolia (8 mg/kg BW) or combination of Eurycoma longifolia and oestradiol for fourteen consecutive days. Results showed that sperm count and spermatogenic cell count were increased in Eurycoma longifolia treated group compared with control group (P < 0.05) and to oestradiol treated group (P < 0.05). While in oestradiol treated group, sperm count and spermatogenic cell count were reduced significantly (P < 0.05) compared with control group. Combination of Eurycoma longifolia and oestradiol did not affect sperm count and spermatogenic cell count when compared with control group but they were significantly increased compared with oestradiol treated group (P < 0.05). Thus, the study has shown that Eurycoma longifolia is potentially capable to suppress harmful effects of oestradiol on spermatogenesis and sperm counts in healthy male rats when given for fourteen consecutive days.

Background: Treatment with buceng combination of Eurycoma longifolia Jack and Pimpinella alpine Molk has been proven to increase testosterone level, decrease apoptosis and caspase3 expression. Bcl2 is an antiapoptotic protein found in cytoplasm which inhibits cells apoptosis. This study was aimed to investigate the effect of buceng on Bcl2 expression on penile and prostate tissues of the rats. Methods: In this experimental study, 24 male Sprague Dawley rats of 90 days old, weighing ± 300 grams, were randomly assigned into four groups. Group A, normal rats. Group B, castrated rats and treated with buceng 100 mg/day, per oral (Cast-Bcg); Group C, castrated rats and treated with 2 ml of water as placebo against buceng (Cast-Plac). Group D, castrated rats, treated with mesterolone 6.75 mg/day, per oral, as exogenous testosterone (Cast-Mest). All rats were treated for 30 days. Manova test was used to analyze the different expression of Bcl2 among groups with significance level at p ≤ 0.05. Results: Castration was associated with significant decrease of Bcl2 expression in the penile and prostate tissues (53.0 and 50.9%, respectively) compared to normal rats (82.6 and 84.2%, respectively, p < 0.001). Treatment with mesterolone reversed Bcl2 expression (77.1 and 78.1%) to a near normal level. The same level of Bcl2 expression was also observed with buceng treatment (73.8 and 78.2%).Conclusion: The treatment with buceng could enhance Bcl2 expression in penile and prostate tissues, comparable to normal rats and mesterolone treated rats.
