Angelica keiskei Koidzumi (A. keiskei), as a Japanese edible herbal plant, enjoys a variety of biological activities due to the presence of numerous active compounds, especially flavonoids. This study aims for the optimization of ultrasound-assisted extraction (UAE) for flavonoids in A. keiskei and their antioxidant activity by using the response surface methodology (RSM). Single-factor experiments and a four-factor three-level Box–Behnken design (BBD) were performed to explore the effects of the following parameters on flavonoid extraction and antioxidant activity evaluation: ultrasonic temperature (X1), ultrasonic time (X2), ethanol concentration (X3) and liquid–solid ratio (X4). The optimum conditions of the combination of total flavonoid content (TFC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity (DPPH-RSC) and ferric-reducing antioxidant power (FRAP) were as follows: X1 = 80 °C, X2 = 4 min, X3 = 78%, X4 = 35 mL/g, respectively. The experimental results provide a theoretical basis for the extensive utilization of A. keiskei and flavonoids extraction from A. keiskei as a potential source of antioxidants.

The present study was designed to establish and optimize a method for extracting natural bioactive compounds from Thymus serpyllum which possess antioxidant, antimicrobial, antispasmotic and stimulant properties. Ultrasound-assisted extraction (UAE) is a well-established method in the processing of plant material, particularly for extraction of bioactive substances such as polyphenols. The influential factors including extraction time (3, 7 and 10 minutes), solid:solvent ratio (1:10, 1:20 and 1:30) and particle size (0.3, 0.7 and 1.5 mm), have been studied to optimize the extraction process, while using 30% ethanol as an extraction medium and amplitude set to 65%. The yield of UAE was expressed via total phenol content and antioxidant activity of the obtained extracts. The optimum process paremeters were found to be: extraction time, 3 min; solid:solvent ratio, 1:30; particle size, 0.3 mm. Under these conditions, the yield of total polyphenols was raised up to 23.03 mg/L GA and the highest antioxidant activity was recorded (10.32 mmol/mg Trolox and IC50 3.00 mg/ml).

In this study, the ultrasound-assisted extraction (UAE) of flavonoid from Paeonia lactiflora seed peel was optimized by response surface methodology (RSM). Single-factor experiments and a three-factor three-level Box-Behnken design (BBD) were performed to explore the effects of the following parameters on flavonoid extraction: ethanol concentration ( X 1 ), liquid-solid ratio ( X 2 ), and ultrasonic time ( X 3 ). The results showed that the optimal flavonoid yield (10.9045 mg RE/g) was as follows: ethanol concentration 62.93%, ultrasonic time 64.56 min, and liquid-solid ratio 24.86 mL/g. The optimized extract of P. lactiflora seed shell was further analyzed by UPLC-ESI-MS/MS, and 20 main flavonoids were identified and quantified, among which protocatechuic acid, vanillic acid, 4-hydroxybenzoic acid, and 3,4-dihydroxybenzaldehyde had the highest content. Furthermore, the results of the antioxidant test showed that the P. lactiflora seed peel extract obtained under optimized UAE conditions exhibited good antioxidant activity. The experimental results showed that ultrasound-assisted extraction was a fast, efficient, and simple method for extracting active ingredients from P. lactiflora seed peel, thereby making this byproduct a promising source of compounds in food and healthcare sectors.
