SummaryA study is reported of the effect of preincubation of raw and laboratory pasteurized bulk milk samples on the tyrosine value (TV) of milk and its relationship with bacterial populations. For raw samples, mean TVs before and after preincubation for 20±2 h at both 25 and 30 °C differed (P < 0·001). After preincubation at 30 °C, the TV of 47 % of raw samples exceeded that considered to represent the upper limit of variation in the TV of a normal bulk milk supply. At this temperature, however, there was no indication that proteolytic psychrotrophs were a major contributor to increased TV. With preincubation of raw samples at 25 °C, an increase to above the upper limit of variation occurred in only 20 % of samples; however, proteolytic psychrotrophs appeared more active at this temperature. In similar samples preincubated after laboratory pasteurization, large increases occurred in TV after 20 ±2 h at 30 °C, while at 25 °C the increases were much smaller and of the order found for preincubated raw samples. The results showed, however, that nonpsychrotrophic bacteria were also the principal source of proteinase giving rise to increased TVs in laboratory pasteurized samples.

Antibiotics, used to maintain healthy dairy animals, persist in milk produced by treated animals for several days after therapy. Antibiotic residues, if present in the milk supply, negatively impact suitability for processing and consumption. This is an area of concern in the dairy industry and for the general public. This study explores the incidence of antibiotic residues in milk provided to a Central Wisconsin dairy. Random, unidentified samples were obtained over a four month period from producers located within a 100-mile radius of a dairy located in Granton, Wisconsin. All samples were tested within one week of their receipt, and maintained at refrigerated temperatures at all times prior to testing. Following testing, the origin of the samples was revealed. Samples were derived from two milk populations: those intended for human consumption (bulk milk), and those from cows under surveillance following antibiotic treatment (milk removed from the commercial supply). All bulk milk samples tested negative for over 55 different antibiotic residues, while all samples from the surveillance population tested positive for antibiotic residues. Our results are consistent with those observed nationally, and suggest that dairy producers recognize the ongoing concern of antibiotic contamination in the milk supply and are taking steps to prevent antibiotic contamination in milk. Our findings also suggest the public should have minimal concern with respect to antibiotic residues in the commercial milk supply.

Background: Mastitis is one of the diseases that cause damage to dairy cattle, and coagulase positive Staphylococcus (CPS), are its main etiological agents. The production of biofilms gives these microorganisms a physiological advantage, being an important virulence factor, as it confers their resistance to the action of disinfectants used in hygiene procedures. Therefore, the objective of this study was to evaluate the capacity of CPS, isolated from milk samples from cases of bovine subclinical mastitis in the Alagoas dairy basin, biofilm production and its sensitivity to disinfectant agents used before and after dipping.Materials, Methods & Results: In the Alagoas Milk Basin Region, 10 properties were selected from the municipalities of Batalha, Cacimbinhas, Major Isidoro and Minador do Negrão, which used a mechanical milking system and supplied milk to a dairy under federal inspection. The California Mastitis Test (CMT) was performed on 1155 cows. The animals diagnosed with subclinical mastitis were classified as: mild (+); moderate (++) and severe (+++). CMT positive samples were collected from 2+, totaling 891 samples. After cleaning the papillary ostium, 2 to 5 mL of milk was collected from each fourth breast before milking began. Subsequently, 0.5 µL of each sample was streak plated by exhaustion on 5% Sheep Blood Agar and incubated at 37°C, with readings at 24 and 48 h. The macroscopic and microscopic characteristics of the colonies, catalase and coagulase test were evaluated. Biofilm formation was determined by the microplate adhesion test. The efficacy of the sanitizers was evaluated with the active ingredients: lactic acid (2%), allantoin (0.05%), iodine (0.5%), chlorhexidine (2.0%), chlorine (2.5%), at times 15, 30 and 60 seconds. It was observed that 91.2% of the samples were able to form biofilm; Of these, 22.2%, 29.6% and 48.2% were strong, moderate and weak producers, respectively. The sensitivity profile of chlorhexidine isolates was 79.73%, 80.41% and 83.78%; iodine was 59.46%, 65.54% and 66.22% sensitivity over the evaluated time. For allantoin, the isolates presented sensitivity of 22.30%, 27.03% and 37.74%, for lactic acid, 18.92%, 20.95% and 22.30% of the samples were sensitive, and to chlorine, 12.6%, 15.58% and 17.57% of the isolates were sensitive. Moreover, it was observed that the longer the exposure to the disinfectant, the greater the effectiveness of the same.Discussion: The results are in line with literature data. Staphylococcus spp. from milk samples of animals with subclinical mastitis are able to form biofilm, thus being an important virulence factor. The heterogeneity regarding the biofilm production potential of the isolates is due to the different species of Staphylococcus spp. that, through distinct mechanisms, triggers a greater or lesser formation of this virulent factor, as well as having their adhesion tested, end up becoming more strongly attached to the plate. Among the disinfectants tested, it was confirmed that chlorhexidine and iodine are more effective than SCP, which tend to have high levels of sensitivity to these disinfectants. However, although chlorhexidine was more sensitive to SCP, its use in the field should be done with caution, as its effectiveness may be reduced in the presence of chlorinated water or excess organic matter. It is concluded that SCP isolates from bovine milk from subclinical mastitis mammary quarters were able to produce biofilms. These isolates were also in vitro sensitive to chlorhexidine and iodine, but periodic evaluation of the efficacy of disinfectants used before and after milking is necessary.

Abstract The accuracy of fat, crude protein (CP), true protein (TP), and lactose determinations of raw milk by Fourier transform infrared (FTIR) spectroscopy and by analysis with a conventional filter-based milk analyzer was assessed in 6 trials performed over a 10-month period. At each trial, 30 bulk milk samples collected from 15 European countries and 11 reconstituted milks made from raw milk components were analyzed. When calibrations were performed with reconstituted milks at each trial, accuracy standard deviations for fat, CP, TP, and lactose were, respectively, 0.050,0.048,0.035, and 0.076 g/100 g for the filter instrument and 0.047, 0.046,0.042, and 0.065 g/100 g for the FTIR instrument. When a single calibration was made instead of calibrations at each trial, accuracy standard deviations increased for the filter instrument to 0.130, 0.119,0.121, and 0.083 for fat, CP, TP, and lactose, respectively, and for the FTIR instrument to 0.082, 0.053,0.044, and 0.084 g/100 g. Because the FTIR instrument provides more spectral information related to milk composition than does the filter instrument, single-calibration FTIR analysis of milk samples collected in different seasons is more accurate. Using reconstituted milks, prepared such that there is no correlation between fat, CP, and lactose, provides a more robust calibration than using genuine bulk milk, especially when milks with unusual composition are analyzed.

The results from a shortened procedure for the direct epifluorescent filter technique (DEFT) determination of viable bacterial populations in raw milk were compared to standard plate counts. Shortening the prefiltration trypsin-Triton X-100 incubation period from 10 to 3 min enabled the completion of the analysis within 20 min. The short DEFT method results had a correlation coefficient (r) of 0.81 with plate counts. With respect to precision, the average difference between values of duplicate plate count analyses was 0.16 log units; that of the short DEFT was 0.14 log units. The slopes of the regressions equations were less than 1, indicating that a direct correlation is not achieved. Short DEFT values were 0.17 log units higher than those of plate counts on milk samples containing less than 10,000 CFU/ml. For milk samples containing counts over 10,000 CFU/ml, short DEFT values averaged only 0.05 log units above plate count readings. Daily preparation of the stain appears unnecessary since acridine orange solutions stored for up to 2 days at 4°C did not produce results significantly (P &gt; 0.05) different from those obtained with fresh solutions. The short DEFT method has potential for the assessment of the bacteriological quality of raw milk in tanker deliveries.

A healthy microbial community in the gut of piglets is critical to minimize the negative performance consequences associated with dietary and environmental changes that occur at weaning. Tonisity Px, an isotonic protein drink, is a potential alternative to balance the gut microbiota as it contains key ingredients for nourishing the small intestine. In the present study, 16 litters comprising 161 piglets were randomly allocated to a group to which Tonisity Px was provided from days 2 to 8 of age (TPX group) or to a control group, to which no Tonisity Px was provided. The TPX group also received Tonisity Px in the 3 days before and after weaning. At days 9, 17, and 30 of age, fecal and ileum samples were collected from piglets belonging to both groups and analyzed using 16S rRNA gene sequencing, semiquantitative PCR of Rotavirus serogroups, and semiquantitative Escherichia coli culture. Overall, Tonisity Px increased the abundance of beneficial bacterial populations (Lactobacillus and Bacteroides species) and reduced potentially pathogenic bacterial populations (E. coli and Prevotellaceae), in both the pre-weaning and post-weaning periods.

Ceftiofur, a third-generation cephalosporin, is one of the most used antibiotics in dairy industry. Intramuscular injection of 1 mg/kgBW ceftiofur hydrochloride (HCl) generally results in 0 hour withdrawal time for the milk in dairy cows. Nevertheless, farmers and dairy processors occasionally complain about ceftiofur-based products in case of positive result to a commercial rapid screening test for the presence of violative residues of antimicrobials (inhibitors) in the bulk milk tank. Six lactating cows were injected with a 50 mg/ml ceftiofur HCl-based product at the dosage regimen of 1 mg/kg, intramuscularly, once a day, for five consecutive days, as per label. Milk samples were then collected just before the very last injection (T0) and then at 12, 24, 36, 48, 60, 72, 84 and 96 hours after the last injection. Individual milk samples were tested using three commercial screening test kits for inhibitor residues: DelvotestSP NT, SNAP Beta-Lactam ST Plus and ROSA MRL Beta-Lactam Test. Since bulk tank is screened in real operating conditions, samples were also diluted to 1:4, 1:10 and tested again. For the Delvotest SP NT, which lowest detected concentration is close the MRL of the ceftiofur (100 µg/kg), all results were negative. For the ROSA MRL Beta-Lactam Test and the SNAP Beta-Lactam ST Plus, several samples yielded positive and doubtful results at T0 and T12. However, after dilution to 1:10, all results were negative. Consequently, when used as officially instructed, the tested 50 mg/ml ceftiofur HCl-based injectable veterinary products are safe, and milk should be free of violative residues of ceftiofur. With consideration to the low specificity and the low positive predictive value of commercial screening tests, positive reactions of the bulk milk should be interpreted as false positive or another risky usage of β-lactam-based medicines in the farm must be investigated.

Objectives The aims of this study were to describe the treatment outcomes following oral administration of a fixed dose (138 MBq; 3.7 mCi) of radioiodine in hyperthyroid cats and to examine the correlation between total thyroxine (TT4) concentrations before and after treatment. Methods This was a retrospective cohort study that documented the TT4 concentration and clinicopathological parameters at the time of diagnosis and after treatment. Logistic regression was used to assess the relationship between TT4 concentrations before and after treatment. The difference in pre- and post-treatment variables between cats that had TT4 concentrations below or within the reference interval (RI) was compared by the Mann–Whitney U-test. Results Of 161 cats, 133 (82.6%) cats had TT4 concentrations within the RI, four (2.5%) cats had TT4 concentrations above the RI and 24 (14.9%) cats had TT4 concentrations below the RI after treatment. The severity of hyperthyroidism at diagnosis, as measured by the percentage of TT4 elevation above the upper limit of the RI, had no impact on the odds of cats having low TT4 concentrations after treatment (odds ratio 1.00; 95% confidence interval 0.96–1.05; P = 0.828). Conclusions and relevance When using an orally administered fixed dose of radioiodine for the treatment of feline hyperthyroidism, TT4 concentrations at diagnosis cannot be used to predict TT4 concentrations after treatment. The proportion of cats with TT4 concentrations below the lower limit of the RI after treatment was 14.9%. Further work is required to optimise oral radioiodine dosing to achieve maximal euthyroid outcomes.
