Microbiological characterisation and impact of suspended solids on pathogen removal from wastewaters in dairy processing factories

In this Research Communication we investigate the microbiological profile of 12 dairy wastewater streams from three contrasting Irish dairy processing factories to determine whether faecal indicators/pathogens were present and in turn, whether disinfection may be required for potential water reuse within the factory. Subsequently, the impact of suspended solids on the inactivation efficiency of Escherichia coli via two means of ultravoilet (UV) disinfection; flow-through pulsed UV (PUV) and continuous low pressure UV (LPUV) disinfection was analysed. Faecal indicators total coliforms and E. coli were detected in 10 out of the 12 samples collected at the dairy processing factories while pathogenic bacteria Listeria monocytogenes was detected in all samples collected at 2 out of the 3 factories. Salmonella spp. was undetected in all samples. The results also indicated that organic dairy wastewater solids had an impact on the performance efficiency of the PUV system and, to a lesser extent, the LPUV system. The findings indicate that the targeting of key pathogens would be required to enable wastewater reuse (and indeed effluent discharges if regulation continues to become more stringent) and that LPUV may offer a more robust disinfection method as it appears to be less susceptible to the presence of suspended solids.

The objective of this work was to study the prevalence of antibiotic resistance phenotypes among total coliforms (TC), E. coli, E. faecalis and Salmonella spp. in the sewage treated effluents generated from three sewage treatment plants in Penang Malaysia. Among the isolates tested, TC and E. coli occurred high resistance for cephalexin (100 and 90.47%), ampicillin (80.93 and 95.23%) and ciprofloxacin (19.06 and 14.3%) compared to E. faecalis (42.86, 71.4 and 4.7%) and Salmonella spp. (59.8, 47.46 and 14.3%) respectively. All E. coli strains, 76.18% of TC, 66.66% of E. faecalis and 35% of Salmonella spp. were multi-resistant.

The Gaza Strip suffers severe constraints in water supply due to its location, confinement, high population density and semi-arid coastal climate. To improve water and agricultural resources, a study was undertaken to show the requirements in planning and management for wastewater treatment, irrigation conveyance and aquifer recharge to meet high technical standards and sustainable economic benefits. Particular attention is paid to economic, financial and socioeconomic analysis. This paper discusses the impact that wastewater reuse will have on the water resources as part of the overall water balance in the Gaza Strip.

Prebiotics are food components that are selectively fermented by beneficial microbiota and which confer a health benefit. The aim of the study was to select a prebiotic for the chosen probiotic strains to create a synbiotic. The impact of prebiotics (inulin, maltodextrin, corn starch, β-glucan, and apple pectin) on five Lactobacillus spp. strains’ growth and metabolites synthesis (lactic, acetic, propionic, and butyric acids, ethanol, and acetaldehyde) was tested by the plate count method and by high-performance liquid chromatography, respectively. Moreover, the differences in the ratio of D(−) and L(+) lactate isomers produced by Lactobacillus spp., as well as variations in the probiotics’ enzymatic profiles associated with the prebiotic used for cultivation, were determined with a Megazyme rapid assay kit and API® ZYM assay, accordingly. Finally, the influence of the carbon source (prebiotic) used on the antagonistic activity of the probiotic strains towards pathogenic bacteria, such as Salmonella spp. or Listeria monocytogenes was analyzed in the co-cultures. The results showed that the growth, metabolic profile, and antagonistic activity of the probiotics towards selected pathogens were the most favorable when 2% (w/v) of inulin was used. Therefore, the combination of inulin with selected probiotics is a promising synbiotic mixture.

This study investigated the impact of waterfowl on the bacteriological quality of village ponds in East Yorkshire, north-east England. Water and sediment samples were collected from ponds with and without resident ducks and geese; faecal indicator and potentially pathogenic bacteria were assayed by membrane filtration and by selective enrichment. Escherichia coli, faecal streptococci and, to a degree, Clostridium perfringens were more abundant in ponds with waterfowl; Salmonella was isolated in June–August from the sediment of a pond with waterfowl. The results suggested that the bacteriological quality of village ponds might be adversely affected by waterfowl. All water samples from ponds with waterfowl had faecal indicators at higher concentrations than EU requirements for bathing waters. Although these ponds are not bathing waters we suggest skin contact and accidental ingestion of water should be avoided.

Abstract Industrial wastewater reuse is a major measure to mitigate the depletion of available freshwater resources in the catchments around industrial areas and to prevent possible future water shortages and the resulting problems for industry, economy and society. Combining a set of environmental aspects and economic aspects of different wastewater treatment technologies, the authors developed a model-based approach for planning and evaluating water reuse concepts in industrial parks. This paper is based on an exemplary Model Industrial Park. The results based on data primarily calculated for Germany show that, for the majority of the indicators, the installation of the Water Reuse Plant seems to be beneficial for all examined reuse options. Considering the economic dimension, due to economies of scale, reuse options with larger volumes of treated water are preferable since the costs per m3 of reuse water are reduced by up to 33%. On the other hand, the environmentally preferable option depends on the respective indicator, e.g. for freshwater eutrophication, the higher the reuse factor, the lower the impact, leading to reductions between 8 and 12%. For climate change, the best option is dependent on the reuse purpose leading to reductions between 8 and 52%.

The study probed into reducing faecal indicators and pathogenic bacteria, heavy metals and β-lactam antibiotics, from four types of secondary effluents by bioaugmentation process, which was conducted with Bacillus subtilis strain at 45 °C. As a result, faecal indicators and pathogenic bacteria were reduced due to the effect of thermal treatment process (45 °C), while the removal of heavy metals and β-lactam antibiotics was performed through the functions of bioaccumulation and biodegradation processes of B. subtilis. Faecal coliform met the guidelines outlined by WHO and US EPA standards after 4 and 16 days, respectively. Salmonella spp. and Staphylococcus aureus were reduced to below the detection limits without renewed growth in the final effluents determined by using a culture-based method. Furthermore, 13.5% and 56.1% of cephalexin had been removed, respectively, from secondary effluents containing 1 g of cephalexin L−1 (secondary effluent 3), as well as 1 g of cephalexin L−1 and 10 mg of Ni2+ L−1 (secondary effluent 4) after 16 days. The treatment process, eventually, successfully removed 96.6% and 66.3% of Ni2+ ions from the secondary effluents containing 10 mg of Ni2+ L−1 (secondary effluent 2) and E4, respectively. The bioaugmentation process improved the quality of secondary effluents.

Abstract The need to cultivate effluent-irrigated rice is paramount and synonymous with treated wastewater reuse, recycling and water resources management. A trial in a peri-urban set-up with a low-cost decentralized wastewater treatment system (DEWATS) was carried out in the 2017 and 2018 cropping seasons to assess the effect of irrigation water reuse management techniques on the yield and water productivity of rice. It was hypothesized that anaerobic baffled reactor (ABR) irrigation water management does not have an effect on the yield of peri-urban grown rice. The impacts of irrigation water management techniques were very significant (P < 0.001) on the number of irrigation events, amount of irrigation and daily water balance. The impact was not significant (P > 0.05) on the tiller numbers per plant; it was however significant (P < 0.05) on the panicle numbers per plant. The effects were not significant (P > 0.05) on the plant height but significant (P < 0.05) on the yield rice for both trials. The effect was also significant (P < 0.05) on water productivity. The result proved that the hypothesis be rejected. It could be concluded that significant potential exists for applying wastewater reuse for non-drinking applications such as irrigation.

Climate change may cause increased microbial growth in water sources and more knowledge is required on how this may affect the hygienic water quality, i.e., whether increased occurrence of cyanobacteria and algae may stimulate the growth rate of opportunistic pathogenic bacteria. Laboratory experiments were performed to investigate if the presence of the cyanobacteria Anabanea lemmermannii and Microcystis aeruginosa affected the survival and growth rate of the opportunistic pathogenic bacteria Aeromonas hydrophila and Pseudomonas aeruginosa, and the faecal indicators Escherichia coli and coliforms. Cyanobacteria were cultured in bottles containing the nutrient-poor medium O2. Sewage, A. hydrophila or P. aeruginosa was added to cyanobacterial cultures and the bacterial growth and survival was followed. E. coli and coliforms from sewage died within few days and the decay rate was not affected by the presence of cyanobacteria. The presence of Anabaena stimulated the growth rate of P. aeruginosa, but had no effect on the growth rate of A. hydrophila. Microcystis had no effect on the growth rate of P. aeruginosa and an inhibiting effect on the growth rate of A. hydrophila.

Dairy Wastewater Sludge (DWS) is sediment from milk processing. Nutritional content of DWS can be used as feed ingredients. The effects of nutrition in DWS are tested on the growth of broiler as well as its specific impact on the development of microflora on broiler digestion. The research methods used experimental design. Tapioca by-product (onggok) is used as DWS binder while Aspergillus niger fermentation is applied to improve nutritional content. The rate of addition fermented DWS in the ration was evaluated through measuring weight gain for 35 days and microflora quality in the cecum by counting the number of non-pathogenic and pathogenic bacteria in the cecum at the end of the research period. The research data were analyzed by ANOVA with Duncan's multiple range test. The results showed that the addition of 20% fermented DWS in rations resulted in the highest body weight gain and could suppress the growth of pathogenic bacteria (Salmonella spp., E. coli, and Enterobacteriaceae) in caecal. The ratio of non pathogenic and pathogenic bacteria increased proportional to the addition of fermented DWS levels in the ration.   Different caecum weight of broiler with different fermented DWS levels was a reprentation of microorganism activity in caecum.  This condition can illustrate the good health status of livestock so as to optimize the growth of broiler.

Aim. To estimate the impact of in vivo secretions of water plantain (Alisma plantago-aquatica) on the popula- tions of pathogenic bacteria Erysipelothrix rhusiopathiae. Methods. The plants were isolated from their natural conditions, the roots were washed from the substrate residues and cultivated in laboratory conditions for 10 days to heal the damage. Then the water was changed; seven days later the selected samples were sterilized using fi lters with 0.2 μm pore diameter. The dilution of water plantain root diffusates in the experimental samples was 1:10–1:10,000. The initial density of E. rhusiopathiae bacteria populations was the same for both experimental and control samples. The estimation of the results was conducted 48 hours later. Results. When the dilution of root diffusates was 1:10, the density of erysipelothrixes in the experimental samples was 11.26 times higher than that of the control, on average, the dilution of 1:100 − 6.16 times higher, 1:1000 – 3.22 times higher, 1:10,000 – 1.81 times higher, respectively. Conclusions. The plants of A. plantago-aquatica species are capable of affecting the populations of E. rhusiopathiae pathogenic bacteria via the secretion of biologically active substances into the environment. The consequences of this interaction are positive for the abovementioned bacteria, which is demon- strated by the increase in the density of their populations in the experiment compared to the control. The intensity of the stimulating effect on the populations of E. rhusiopathiae in the root diffusates of A. plantago-aquatica is re- ciprocally dependent on the degree of their dilution. The investigated impact of water plantain on erysipelothrixes should be related to the topical type of biocenotic connections, the formation of which between the test species in the ecosystems might promote maintaining the potential of natural focus of rabies.
