Bovine immunoglobulin G, β-lactoglobulin, α-lactalbumin and serum albumin in colostrum and milk during the early post partum period

Colostrum and milk samples from 60 Holstein–Friesian cows were analysed for concentrations and yields of immunoglobulin G (IgG), β-lactoglobulin (β-lg), α-lactalbumin (α-la) and serum albumin (BSA) throughout the first 16 milkings post partum (8 d of lactation) using a single radial immunodiffusion assay. Concentrations (mg/ml, means±SD) at first milking were IgG 59·8±28·5, β-lg 14·3±4·6, α-la 2·04±0·6, BSA 1·21±0·44. Large variations were recorded for IgG concentrations (15·3–176·2 mg/ml) and yields (0·2–925 g). Cows in their first lactation produced significantly lower concentrations and yields of colostral IgG than cows in later lactations. A colostral yield of IgG below the 100 g required to prevent calf hypo-γ-globulinaemia was found in 18·3% of the cows. The concentrations of IgG, β-lg and BSA dropped abruptly in subsequent milkings and α-la concentration decreased slowly. The mean IgG concentration was <2 mg/ml after eight milkings and <1 mg/ml after fifteen milkings. However, IgG concentration did not differ significantly, at the 1% level, during milkings 11–15. The results were tabulated to make it possible to calculate the excess of whey proteins that would be obtained if early milks were illegally added to the milk supply.

Concentrations (mg/ml, means±SD) at first milking were IgG 47·9±25·5, β-lg 30·7±10·4, α-la 2·77±0·82 and CSA 2·97±2·46 mg/ml. Large variations were recorded for IgG concentrations (19·9–94·5 mg/ml) and β-lg (9·3–49·8 mg/ml). Concentrations of IgG, β-lg and CSA dropped abruptly in the subsequent milkings and α-la concentration decreased slowly. Mean IgG concentration was <2 mg/ml after 7 milkings and <1 mg/ml after 11 milkings. However, IgG concentration does not differ significantly, at the 1% level, from milkings 7–14. The contribution of β-lg to the increase in whey proteins in early milks was greater than that of IgG from milkings 5 to 14. The results were tabulated to make it possible to calculate the excess of whey proteins that would be obtained if early milks were illegally added to milk supply.

AbstractIn order to investigate the rationale of manipulating post-ovulation progesterone (P4) concentrations, luteal activity was measured in 10 Holstein-Friesian cows treated with a progesterone-releasing device (CIDR, 1·9 g P4) inserted on the morning of day 2 post-oestrus for a period of 7 days and compared with 10 untreated control animals. Milk samples were collected daily during afternoon milking from 7 days pre- to 24 days post-oestrus and P4 concentrations were measured using enzyme-linked immuno-sorbent assay (ELISA). The milk P4 profiles were used to assess the effects of early post-oestrus P4 elevation on five intervals of the luteal phase. In the treated (T) and control (C) animals, the mean interval from oestrus to P4 rise (≥3 μg/l), was 2·38 (s.e. 0·18) and 4·90 (s.e. 0·28) days, respectively, (P < 0·01). The mean interval from oestrus to the attainment of peak P4 concentrations was not significantly different (P > 0·05) between the T (14·75 (s.e. 1-62) days) and С (14·30 (s.e. 0·70) days) animals, with cows in the T group showing a more variable (F7 9 = 4-30, P < 0·05) interval to the occurrence of the peak. The interval from peak P4 to corpus luteum (CL) regression (when P4 fell below ≥3 μg/l) was not significantly different between the T (4·13 (s.e. 1·30 days) and С (5·60 (s.e. 0·88) days) groups. Furthermore, CIDR insertion did not alter luteal phase length, number of days with P4 concentrations ≥3 μg/l (T = 16·50 (s.e. 0·80) v.C = 15·00 (s.e. 0·47) days) or the interval from oestrus to CL regression (T = 18·88 (s.e. 0·79) v.C = 19·90 (s.e. 0·41) days). We conclude that treatment with CIDR 2 days after oestrus successfully increased the availability of P4 to cows without significantly affecting luteal phase characteristics.

AbstractThe objective of the present study was to evaluate the efficacy of a prostaglandin F2α(PGF2α) and progesterone (P4) treatment in initiating oestrus and ovulation post partum (PP) in Holstein-Friesian cows. Using four herds, the treatment protocol consisted of a single intra-muscular injection of PGF2α(Estrumate) between days 12 to 14 PP followed 48 h later by progesterone treatment via intra-vaginal CIDR insertion for a period of 7 days. Milk samples for progesterone determination were collected three times weekly from 7 to 65 days PP. The ovarian activity and reproductive performance of treated (T, no. = 153) animals and untreated control cows (C, no. = 315) were compared. Treatment was effective (P< 0·001) in reducing the mean interval to PP commencement of luteal activity from 29·62 (s.e. 0·82) days to 22·09 (s.e. 0·70) days. The mean interval to first PP oestrus in the T animals was significantly reduced (P< 0·001) from 55·62 (s.e. 1·58) days to 44·91 (s.e. 1·44) days and the incidence of silent ovulation in cycles between days 21 to 65 PP was reduced (P< 0·001) from 56·28% to 42·27%. In two herds under one management regime and with a similar block-calving pattern (no. = 280 animals), the treatment protocol was beneficial to the overall reproductive performance as there was a significant shortening in the mean interval to first PP service (75·82 (s.e. 1·93) v. 80·86 (s.e. 1·32) days) and in the interval to PP conception (83·07 (s.e. 2·49) v. 88·90 (s.e. 1·95) days), bothP< 0·05.

Colostrum and milk samples from twelve Tunisian camels were analysed for concentration of immunoglobulin G (IgG), α-lactalbumin (α-la), serum albumin (CSA) and lactoferrin throughout the first 14 milkings post partum (7 days of lactation) using single radial immunodiffusion assay. Concentrations (mg/ml, means±SD) at first milking were IgG, 100·7±60·4; α-la, 2·2±0·7; CSA, 8·5±3·6 and lactoferrin, 1·2±0·3. Large variations were recorded for IgG and CSA concentrations (11·8–211·1 mg/ml and 2·9–13·8 mg/ml respectively) Concentrations of IgG and CSA dropped abruptly in the subsequent milkings while α-la concentration increased until milking 5 and then decreased slowly. Lactoferrin dropped only from milking 7. Mean IgG concentrations were 3·6 and 2·5 mg/ml at milking 9 and 13 respectively. However, IgG concentration did not differ significantly, at the 1% level, from milkings 11 to 14. The contribution of CSA to the increase in whey proteins in early milks was greater than that described in the bovine and caprine species.

<em>Longissimus thoracis</em> and <em>lumborum</em> (LTL) and <em>Gluteus medius</em> (Gm) muscles of culled dairy cows, differing in production status (Ps) at slaughter and carcass weight were assessed for intramuscular fat and myoglobin, color, shear force (SF) and sensorial characteristics, after being aged for 2, 7, 14, 28 and 42 days. Meat from dried-off cows was lighter, redder and had higher yellowness and chroma (<em>P </em>&lt; 0.05) than lactating cows. Meat of finished fed cows improved in color in relation to counterparts directly slaughtered, but not significantly. Color parameters increased with aging time and the mean values of LTL and Gm significantly differed mostly among lactating cows. SF of meat was affected by aging time, muscle type, Ps and by their respective interactions (<em>P</em> &lt; 0.001), with a greater impact in LTL muscle. Meat from LTL muscle of dried-off cows had lower SF (47.30 N) than that of lactating cows (65.61 N) (<em>P</em> &lt; 0.05), but such differences were not significant for Gm muscle. Meat tenderness of finished fed cows was higher than counterparts not finished (<em>P</em> &gt; 0.05). Main effects and the results from their interactions were significant for all sensorial attributes (<em>P</em> &lt; 0.001).

Pedigree data from national breeding value evaluations were used in calculation of the coefficient and rate of inbreeding, average coefficient and rate of relationship and generation intervals for the Finnish Ayrshire and Holstein-Friesian dairy cattle populations. The data had 1,366,555 Ayrshire and 377,869 Holstein-Friesian animals. The mean coefficient of inbreeding for Ayrshire and Holstein-Friesian animals born in the 1990s was 2.29% and 0.90%, respectively, and the trend was towards higher inbreeding values. The average coefficient of relationship, mean increase in inbreeding and generation interval was calculated for bulls born between 1976 and 1999, and for cows born between 1986 and 1999. The mean coefficient of relationship of Ayrshire bulls increased 2.22 %-units per generation and inbreeding increased 0.20 %-units per generation during the years studied. The mean coefficient of relationship of Finnish Holstein-Friesian bulls increased 0.96 %-units per generation and inbreeding 0.17 %-units per generation. The mean coefficient of relationship and inbreeding of Ayrshire cows increased 0.38 %-units and 0.31 %-units per generation, respectively. For Holstein-Friesian cows the mean coefficient of relationship and inbreeding increased 0.25 %-units and 0.11 %-units per generation, respectively. Results show that inbreeding is low and it is increasing slowly in both breeds. However, especially the coefficients of relationship of Ayrshire bulls are high in some age classes and this may lead into faster increase in coefficients of inbreeding.;

AbstractControlled breeding schemes for oestrous detection constitutes a proactive technical response that balances the infrastructural requirement for a profitable dairy operation and the demands for optimal animal performance. The present study compared (a) the reproductive response of a treatment based on a short vs longer-acting PGF2α analogue (tiaprost vs luprostiol), and (b) the reproductive response after a treatment of GnRH-PGF2α vs PGF2α alone for synchronizing dairy cows. Holstein-Friesian cows averaging 9000 kg milk/lactation and fed according to their requirements were used in the study. Cows were cyclic, at least 60 days post partum and were clinically sound before being considered for the experiments. In Experiment 1, animals were synchronised using an i.m. injection of either 15 mg of luprostiol or 0.75 mg of tiaprost, based on ultrasonic diagnosis of a corpus luteum. Animals were inseminated at observed oestrus. In Experiment 2, cows were synchronised, at random, by either an injection of 10pg ofbuserelin (day 0) followed by 0.75 mg of tiaprost at day 7 (GnRH-PGF2α) orjust 0.75 mg of tiaprost (PGF2α). For both treatments only cows with an ultrasonically detected corpus luteum were treated. Animals were inseminated at oestrus. At the time of treatment and again 3 days later, milk samples were collected and assayed for progesterone by RIA. Cows with progesterone concentrations >1 ng/ml were considered to have corpus luteum. Luteolysis was considered to have occurred when concentrations of progesterone were > 1 ng/ml at day 0 and <0.8 ng/ml at day 3. In Experiment 1, both analogues gave similar results in terms of induced luteolysis [luprostiol: 36/39 (92.3%) vs tiaprost: 36/41 (87.8%)], oestrous detection efficiency [luprostiol: 26/36 (72.2%) vs tiaprost: 30/36 (83.3%], oestrous distribution [day 2, 3 and 4, respectively: luprostiol: 26.9%, 50.0%, 19.2% vs tiaprost: 36.7%, 50.0%, 13.3%], and conception rates [luprostiol: 12/25 (48.0%) vs tiaprost: 14/28 (50.0%); P>0.05]. In Experiment 2, oestrous detection efficiency, interval to oestrus and conception rate were similar between treatments [97/149 (65.1%), 71.1 h, 43/95 (45.3%) for PGF2α vs 130/188 (69.1%), 68.2h, 65/126 (51.6%) for GnRH-PGF2α, respectively]. However the oestrous distribution was more concentrated in GnRH-PGF2α treated animals (P<0.01).

ABSTRACTTwenty-six pregnant Holstein-Friesian × zebu heifers, arranged in pairs according to age, body weight, expected calving date and amount of zebu breeding, were allocated at random to two nutritional regimens during the last 12 weeks before calving. If was planned that the animals in treatment A should calve at a pre-calving live weight of 480 kg and those in treatment B should calve at 380 kg. The diet consisted of maize silage or elephant grass silage, soya-bean meal and minerals. During the first 21 days of the post-partum period both groups were given 6 kg concentrate per head daily. After this period, the feeding level was adjusted every 14 days according to the current milk yield. The mean total milk yields were 2132 kg and 1283 kg (P < 0·05) and the lengths of lactation were 239 days and 156 days (P < 0·01) for cows in treatments A and B, respectively. There was no statistically significant difference between treatments in the intervals from calving to first heat or calving to conception.

SummaryThe interaction of sheep, horse, pig, human and guinea-pig whey proteins with fatty acids has been studied. Using gel filtration and autoradiography, it was found that sheep β-lactoglobulin and serum albumin from all species had the ability to bind fatty acids in vitro. Sheep β-lactoglobulin, isolated from milk, had ˜ 0·5 mol fatty acids bound per mol monomer protein, and albumin from sheep, horse and pig contained ˜ 4·5, 2·9 and 4·7 mol fatty acids/mol protein respectively. However, β-lactoglobulin from horse and pig milk had neither fatty acids physiologically bound nor the ability to bind them in vitro. Albumin was the only whey protein detected with bound fatty acids in these species as well as in human and guinea pig. This suggests that the ability of ruminant β-lactoglobulin to bind fatty acids was not shared by the same protein of non-ruminants.
