Endogenous amino acid flows at the terminal ileum of broilers, layers and adult roosters

AbstractEndogenous flows of nitrogen and amino acids at the terminal ileum of broilers (6 weeks old), layers (70 weeks old) and adult roosters (70 weeks old) were determined using the peptide alimentation method. The ileal endogenous output of nitrogen and total amino acids in broilers, layers and roosters, expressed as mg/kg dry matter intake, were similar (F > 0-05). Endogenous flows were similar (F > 0-05) for nine of the 17 amino acids analysed, but the flows of serine, glutamic acid, proline, alanine, isoleucine, tyrosine, arginine and methionine differed (P < 0-05) among the classes of chickens. The amino acid profile of endogenous protein, expressed asg/100 g crude protein, did not differ (F > 0-05) between the three classes of chickens, except for serine, glutamic acid, proline and isoleucine. The concentrations of proline were higher (F < 0-05) in broilers, compared with the other two groups. The concentrations of glutamic acid in layers were lower (F < 0-05) than the other two groups. The concentrations of serine and isoleucine were higher (F < 0-05) in roosters than the other two groups. In all three groups, the most abundant amino acid in the ileal endogenous protein was glutamic acid, followed by aspartic acid, proline, serine, glycine and threonine. The present study provides estimates for endogenous amino acid flow at the terminal ileum in broilers, layers and adult roosters under normal physiological conditions.

The aim of the present study was to establish whether feeding broiler chickens with diets containing increasing dietary peptide concentrations would cause increases in ileal endogenous amino acid flow. The flow of N and most amino acids increased quadratically (P < 0·05 to 0·001) with increasing dietary concentrations of peptides. The exceptions were the flow of threonine, serine, glycine, tyrosine and cystine, which increased linearly (P < 0·001) with dietary peptide levels. Another notable exception to the general trend was the flow of proline, which was significantly higher (P < 0·01) in birds fed the protein-free diet. The amino acid profile of endogenous protein, expressed as proportion of crude protein, indicated that the ratios of threonine, glutamic acid, proline, glycine, leucine, histidine, arginine and cystine were influenced (P < 0·05) with increasing dietary peptide concentrations. In general, compared with the protein-free diet, the ratios of threonine and arginine in endogenous protein were lower (P < 0·05) and those of glutamic acid, glycine and histidine were greater (P < 0·05) in diets with high concentrations of peptides. The ratio of proline was found to decrease (P < 0·05) with increasing dietary peptide concentrations. These changes in the amino acid profile of endogenous protein are probably reflective of changes in the output of one or more of the components of endogenous protein. Overall, the present results demonstrated that increasing dietary peptide concentrations increased the flow of endogenous amino acid flow at the terminal ileum of broiler chickens in a dose-dependent manner and also caused changes in the composition of endogenous protein. The observed changes in endogenous amino flow will influence the maintenance requirements for amino acids and also have implications for the calculation of true digestibility coefficient of feedstuffs.

AbstractIleal digestibility of amino acids in barley-based diets with increasing inclusion of lucerne leaf meal (LLM; 0, 50, 100 and 200 g/kg) was studied in a change-over experiment with post-valve T-caecum cannulated growing pigs. The apparent Heal digestibility of crude protein, as well as of most of the essential (EAA) and non-essential amino acids (NEAA) was not significantly affected by the dietary inclusion of LLM. The exceptions were cystine, methionine, phenylalanine, glutamic acid and serine, all of which showed a significant (P< 0·05) reduction in apparent Heal digestibility with increasing inclusion of LLM. In contrast, the calculated true Heal digestibility of all EAA (with the exception of arginine and lysine) and the NEAA glutamic acid and serine were significantly (P< 0·05) reduced with the inclusion of LLM. Associated with an increased crude protein intake, there was a significant (P< 0·05) increase in the amount of absorbed EAA when the proportion of LLM was increased in the diet. With increasing proportion of fibre in the diet, there was a significant (P< 0·05) increase in the Heal flow of glucosamine.It can be concluded from the present data that the inclusion of LLM in a barley-based diet for growing pigs will result in an improvement in the absorbed amino-acid profile due to a significant increase in the absorption of all of the EAA.

The recovery and composition of endogenous protein at the terminal ileum of broiler chickens were determined at 14 and 42 days post-hatching using the peptide alimentation method. The ileal endogenous flows of nitrogen and amino acids, expressed as mg/kg dry matter intake, differed (P < 0.05–0.01) between the two age groups, with flows increasing with age, except for lysine, histidine and glycine. The flows of lysine and histidine were unaffected (P > 0.05) by age, whereas a tendency (P = 0.07) for increased loss with age was observed for glycine. The amino acid profile of ileal endogenous protein, expressed as g/100 g crude protein, did not differ (P > 0.05) between 14-day- and 42-day-old broilers, suggesting that the relative proportions of the individual sources that contribute to the endogenous protein were similar at both ages. In both age groups, the most abundant amino acids in the ileal endogenous protein were glutamic acid, aspartic acid, proline, serine, glycine, and threonine. The present findings suggest that, when determining true digestibility, corrections using endogenous amino acid flows determined with broilers of a particular age to apparent amino acid digestibility values determined with birds of a different age would clearly result in less accurate true digestibility estimates.

Free amino acids in cacao beans are important precursors to the aroma and flavor of chocolate. In this research, we used inferential and explanatory statistical techniques to verify the effect of different edaphic crop conditions on the free amino acid profile of PH-16 dry cacao beans. The decreasing order of free amino acids in PH-16 dry cacao beans is leucine, phenylalanine, glutamic acid, alanine, asparagine, tyrosine, gamma-aminobutyric acid, valine, isoleucine, glutamine, lysine, aspartic acid, serine, tryptophan, threonine, glycine. With the exception of lysine, no other free amino acid showed a significant difference between means of different edaphic conditions under the ANOVA F-test. The hydrophobic free amino acids provided the largest contribution to the explained variance with 58.01% of the first dimension of the principal component analysis. Glutamic acid stands out in the second dimension with 13.09%. Due to the stability of the biochemical profile of free amino acids in this clonal variety, it is recommended that cacao producers consider the genotype as the primary source of variation in the quality of cacao beans and ultimately the chocolate to be produced.

The aim of the present study was to compare the protein-free diet, guanidinated casein (GuC) and enzyme hydrolysed casein (EHC) methods for the quantification of endogenous amino acid (AA) flow in the avian ileum. Growing broiler chickens (5 weeks old) were used. All three assay diets were based on dextrose, and in the GuC and EHC diets GuC or EHC were the sole source of N. Endogenous AA flows determined with the use of protein-free diet were considerably lower (P>0·05) than those determined by the GuC and EHC methods. The total endogenous AA flows determined by the GuC and EHC methods were almost 3-fold greater (P>0·05) than those determined by the protein-free diet. The endogenous AA values obtained from GuC and EHC methods were similar (P<0·05), except for the flow of arginine, which was lower (P>0·05) in the EHC method. Glutamic acid, aspartic acid, threonine and glycine were the predominant endogenous AA present in digesta from the distal ileum. The contents of methionine, histidine and cystine were lower compared with other AA. The method of determination had no effect on the AA composition of endogenous protein, except for threonine, glutamic acid, lysine, arginine and cystine. The concentrations of threonine and arginine were lower (P>0·05) and that of lysine was higher (P>0·05) with the EHC method compared with the other two methods. The concentration of glutamic acid was greater (P0·05) and that of cystine was lower (P>0·05) in the EHC and GuC methods compared with the protein-free diet method. The results showed that the ileal endogenous flows of N and AA are markedly enhanced by the presence of protein and peptides, above those determined following feeding of a protein-free diet. It is concluded that the use of EHC and GuC methods enables the measurement of ileal endogenous losses in chickens under normal physiological conditions.

The amino acid profile of 11 samples of tiger nuts ( Cyperus esculentusL.) grown in the area of “L'Horta Nord” in Valencia (Spain) and one sample of African origin were determined, along with the amino acid contents of 10 samples of natural orgeat from Valencia. Protein was hydrolysed by hydrochloric acid at 110 °C for 23 h, and amino acids were derivatised with AQC and determined by RP-HPLC with fluorescence detection. The chromatographic conditions were optimised. The analytical parameters (detection and quantification limits, precision and accuracy) showed the method to be sufficiently sensitive and reproducible for determining amino acids resistant to acid hydrolysis in tiger nuts and orgeat. Arginine was the most abundant amino acid in both tiger nuts and orgeat and the lowest contents corresponding to histidine and tyrosine. The essential amino acid contents of tiger nuts and orgeat protein were greater than those proposed in the protein standard for adults by the FAO/WHO, with the exception of histidine. No significant differences were found among the arginine, lysine and isoleucine amino acid contents in tiger nuts from Valencia, Alboraya and Alm‡ssera; nor were they found among amino acids in tiger nuts from Valencia and Alm‡ssera, with the exception of tyrosine.

Knowledge of the profile of amino acids of the rumen-undegradable protein can help in the formulation of diets to provide amino acids that complement microbial protein as well as supply amino acids, which are most limiting for milk production. Three non-lactating cows fitted with rumen cannulas were used to determine the effect ofin siturumen degradation on crude protein and amino acid profile of rumen-undegraded protein of feedstuffs. The obtained values of rumen degradability of crude protein with significant difference (P< 0.001) between feeds ranged from 20.3 to 76.3% (mean 62.0 ± 17.9%) and values of total amino acids ranged from 30.9% in corn gluten meal to 83.8% in corn gluten feed (mean 67.5 ± 16.4%). Anin vitromodified 3-step method was used to determine intestinal digestibility. Intestinal digestibility of undegraded protein varied from 54.5 ± 1.4% in raw soybean to 95.2 ± 1.0% in corn gluten feed. The absorbable amino acid profile of rumen-undegraded protein for each feedstuff was compared with profiles of the original feedstuff and the rumen-exposed undegraded protein. Absorbable lysine (9.3 ± 1.1 g/kg of crude protein) was higher in products of soybean and sunflower cake. Corn gluten feed and meal supplied more absorbable methionine (3.6 ± 0.6 g/kg of crude protein). This study showed that the digestibility factor of crude protein and amino acid based onin situandin vitromethods for thermal treatment of protein feeds can be used in models to optimize the amino acid nutrition of dairy cows and expand knowledge about rumen degradability and ileal digestibility of amino acids in feedstuffs.

The proximate and amino acid compositions of Artocarpus altilis and Bucchholzia coriacea were investigated using standard analytical techniques. The respective proximate composition (%) for the Artocarpus altilis and Bucchholzia coriacea samples were: Moisture (5.91 and 5.67); ash (4.39 and 5.13); crude fat (4.42 and 2.76); crude protein (12.27 and 14.78); crude fibre (7.10 and 6.27); carbohydrate (65.91 and 65.39). The calculated fatty acids and metabolizable energy for the Artocarpus altilis and Bucchholzia coriacea samples were 3.54 and 2.21 %; 1492.60 and 1465.01 kJ/100 g, respectively. The most abundant minerals in Artocarpus altilis and Bucchholzia were potassium (658.42 and 369.64 mg/100 g) and phosphorus (382.55 and 261.38 mg/100 g), respectively. The amino acid profile revealed that Artocarpus altilis and Bucchholzia coriacea samples contained nutritionally useful quantities of most of the essential amino acids. The total essential amino acids (TEAA) (with His) were 31.14 and 34.40 g/100 g crude protein for the Artocarpus altilis and Bucchholzia coriacea samples, respectively. The first limiting amino acid was Met + Cys (TSAA) for all the samples and calculated isoeletric points (ρl) were 3.52 and 4.05 for Artocarpus altilis and Bucchholzia coriacea, respectively.Bangladesh J. Abstract:The objective of this work was to evaluate the apparent digestibility coefficients of nutrients, energy, and amino acids of nontoxic and detoxified physic nut cakes treated with solvent plus posterior extrusion, for Nile tilapia. The apparent digestibility coefficients of crude protein and gross energy were higher for detoxified than for nontoxic physic nut cake. However, the apparent digestibility coefficient of ether extract of the nontoxic physic nut cake was higher than that of the detoxified one. The apparent digestibility coefficient of amino acids of both feed ingredients was superior to 80%, except for glycine, for the nontoxic psychic nut cake, and for threonine, for the detoxified one. Nontoxic and detoxified physic nut cakes show apparent digestibility coefficient values equivalent to those of the other evaluated oilseeds and potential for inclusion in Nile tilapia diets.

1. Pigs prepared with re-entrant ileal cannulas were used to determine the ileal availability of amino acids in nine cereal grains including five wheats, sorghum (Sorghum vulgare Pers.), maize, barley and Triticale.2. The average true availability of amino acids in these grains was 0·88 but there were consistent differences in availability among amino acids. Generally, lysine and threonine were among the least available amino acids while glutamic acid and arginine were among the most available amino acids in cereal protein.3. There was as much variation in amino acid availability within a grain species (wheat) as among the other grains.
