AbstractMycobacterium abscessus, a multidrug-resistant nontuberculous mycobacterium, has emerged as a major pathogen affecting people with cystic fibrosis (CF). Although originally thought to be acquired independently from the environment, most individuals are infected with one of several dominant circulating clones (DCCs), indicating the presence of global transmission networks of M. abscessus. How and when these clones emerged and spread globally is unclear. Here, we use evolutionary analyses of isolates from individuals both with and without CF to reconstruct the population history, spatiotemporal spread and recent transmission networks of the DCCs. We demonstrate synchronous expansion of six unrelated DCCs in the 1960s, a period associated with major changes in CF care and survival. Each of these clones has spread globally as a result of rare intercontinental transmission events. We show that the DCCs, but not environmentally acquired isolates, exhibit a specific smoking-associated mutational signature and that current transmission networks include individuals both with and without CF. We therefore propose that the DCCs initially emerged in non-CF populations but were then amplified and spread through the CF community. While individuals with CF are probably the most permissive host, non-CF individuals continue to play a key role in transmission networks and may facilitate long-distance transmission.

Some epidemiological aspects of leishmaniasis in the municipality of Formiga, Brazil, an important touristic site, were evaluated. Those included phlebotomine sand fly vectors, canine infection, and geoprocessing analysis for determining critical transmission areas. Sand flies (224 insects) belonging to ten different species were captured. The most captured species includedLutzomyia longipalpis(35.3%),Lutzomyia cortelezzii(33.5%), andLutzomyia whitmani(18.3%). A significant correlation between sand fly densities and climatic conditions was detected. Serological diagnosis (DPP and ELISA) was performed in 570 dogs indicating a prevalence of 5.8%. After sequencing the main species circulating in the area wereLeishmania infantumandLeishmania braziliensis. Spatial analysis demonstrated that vegetation and hydrography may be related to sand fly distribution and infected dogs. The municipality of Formiga has proven leishmaniasis vectors and infected dogs indicating the circulation of the parasite in the city. Correlation of those data with environmental and human cases has identified the critical areas for control interventions (south, northeast, and northwest). In conclusion, there is current transmission of visceral and canine human cases and the city is on the risk for the appearance of cutaneous cases.

ABSTRACTTwo A/C incompatibility group (IncA/C family) plasmids from the 1960s have been sequenced and classified into the A/C2type 1 group. R16a and IP40a contain novel antibiotic resistance islands and a complete GIsul2 genomic island not previously found in the family. In the 173.1-kb R16a, the 29.9-kb antibiotic resistance island (ARI) is located in a unique backbone position not utilized by ARIs. ARIR16aconsists of Tn1, Tn6020, and Tn6333, harboring the resistance genesblaTEM-1DandaphA1band amermodule, respectively; a truncated Tn5393copy; and a gene cluster with unknown function. Plasmid IP40a is 170.4 kb in size and contains a 5.6-kb ARI inserted into thekfrAgene. ARIIP40acarryingblaTEM-1DandaphA1bgenes is composed of Tn1with a Tn6023insertion. Additionally, IP40a harbors single IS2, IS186, and Tn1000insertions scattered in the backbone; an IS150copy in GIsul2; and a complete Tn6333carrying amermodule at the position of ARIR16a. Loss of resistance markers in R16a, IP40a, and R55 was observed during stability tests. Every phenotypic change proved to be the result of recombination events involving mobile elements. Intramolecular transposition of IS copies that generated IP40a derivatives lacking large parts of the backbone could account for the formation of other family members, too. The MinION platform proved to be a valuable tool in bacterial genome sequencing since it generates long reads that span repetitive elements and facilitates full-length plasmid or chromosome assembly. Nanopore technology enables rapid characterization of large, low-copy-number plasmids and their rearrangement products.

The American Flamingo, Phoenicopterus ruber, is a charismatic bird distributed throughout the Caribbean, North and South America. Its wide distribution, the complexity of international monitoring due to its capacity for long-distance flying, and a focus mostly on local populations, make it difficult to understand the dynamics between sites. Here, we took advantage of the citizen eBird science project to present a global perspective on the distribution of the American Flamingo, and identify the potentially most important countries for breeding. We obtained 16,930 records for the Americas from the 1960s until October 2018, of which 9,283 could be used for our objectives. The eBird database indicated a considerable increase in the total number of records over the last decade (2010s), probably reflecting an increase in tourism facilities, research investment, technological advancement, interest in conservation, and the worldwide availability of eBird. We also observed a range extension in the Gulf of Mexico in the United States and a significant recolonization in the Florida Peninsula. The apparent range extension to the South is more likely to be linked to biases in the data; for example, in any given country the number of records might reflect either reporting efforts or actual numbers. eBird data confirmed that six countries host the main breeding colonies (Bahamas, Bonaire, Cuba, Ecuador, Mexico, and Venezuela). We suggest three additional countries as potential breeding areas for the species (Colombia, Curaçao, Turks and Caicos Islands) for which more field observations are necessary to support this possibility. This global appraisal of the distribution of the American Flamingo using citizen science data provides valuable information for national and international management and conservation programs such as the need to verify the species breeding status in areas where it appears to be expanding its distribution.

Despite advances in diagnosis and treatment, tuberculosis (TB) continues to be one of the essential health problems throughout the world. Turkey is considered to be endemic for TB. In this study, we analyzed the distribution of Mycobacterium species, compare the diagnostic methods, and susceptibilities to anti-tuberculosis drugs of TB isolates. The aim was to document the current status and to provide a frame of reference for future studies. In this study, 278 Mycobacterium species isolated from 7,480 patients between September 2015 and June 2019 were included. Löwenstein-Jensen medium (LJ) and MGIT 960 were used for the isolation of strains. Susceptibility to 1st-line anti-tuberculosis drugs was determined. Positivity rates in clinical samples were as follows: 1.4% for direct microscopic acid-fast bacilli (AFB) detection, 3.4% for growth on the LJ, and 3.7% for growth on MGIT-960. Two hundred thirty-three isolates were identified as Mycobacterium tuberculosis complex (MTBC) and 45 were non-tuberculous mycobacteria (NTMs). Eleven of the NTMs (24.4%) were Mycobacterium fortuitum group isolates, and eight NTMs (17.7%) were Mycobacterium abscessus complex isolates. A number of patients diagnosed with tuberculosis peaked twice between the ages of 20–31 and 60–71. A hundred and eighty-two MTBC isolates (78.1%) were susceptible to all 1st-line anti-tuberculosis drugs, while 51 isolates (21.9%) were resistant to at least one drug tested. The multidrug-resistant tuberculosis rate was 13.7% among resistant strains and 3% in all strains. The liquid cultures were better for detection of both MTBC and NTMs isolates. The data demonstrate that MTBC continues to be challenge for this country and indicates the need for continued surveillance and full-spectrum services of mycobacteriology laboratory and infectious diseases.

Compared with AC (alternating current) transmission, DC(direct current) transmission has obvious superiority at high power, long distance, submarine cable transmission and asynchronous interconnected between exchange systems etc.. This paper constructed a kind of 12 pulse bridge UHV DC transmission system that includes control protection function through MATLAB/Simulink tool. By setting the breaker parameters, this paper studied the 12 pulse bridge UHV DC transmission system AC/DC side in case of setting down time, type and cleared. The simulation results demonstrate that this system has a good effect in harmonic control and it has no power compensations in realizing HV DC process, and it can completely work as an important reference in constructing HV DC power grid.

For most of the twentieth century migration and invasion were the default explanation of material culture change in archaeology. This model was largely derived from the record of documentary history, which not only recorded the Gaulish diaspora of later prehistory but the migrations that resulted in the breakup of the Roman Empire. The equation of archaeological distributions—the formula ‘pots = people’—was a model adopted and promoted by Gordon Childe, and remained fundamental to archaeological interpretation into the 1960s. Thereafter diffusionism was discredited among British prehistorians, though less so among European archaeologists and classical or historical archaeologists. Even the Beaker phenomenon became a ‘cult package’ rather than the product of settlers, and it is only as a result of more recent isotopic and DNA analyses that the scale of settlement from the continent introducing Beakers has begun to be demonstrated. Other factors in culture contact including long-distance trade have long been evident, for example, from the distribution of finds of Baltic amber from Northern and North-Western Europe to the Mediterranean, or the distribution of continental pottery and glass via the western seaways in the post-Roman period.

A single extra-intestinal pathogenic Escherichia coli (ExPEC) clone, named sequence type (ST) 131, is responsible for millions of global antimicrobial-resistant (AMR) infections annually. Population genetics indicate that ST131 consists of different clades (i.e. A, B, and C); however, clade C is the most dominant globally. A ST131 subclade, named C1-M27, is emerging in Japan and has been responsible for the recent increase in AMR ExPEC in that country. The sequential acquisition of several virulence and AMR genes associated with mobile genetic elements during the 1960s to 1980s primed clade C (and its subclades C1 and C2) for success in the 1990s to 2000s. IncF plasmids with F1:A2:B20 and F2:A1:B replicons have shaped the evolution of the C1 and C2 subclades. It is possible that ST131 is a host generalist with different accessory gene profiles. Compensatory mutations within the core genome of this clone have counterbalanced the fitness cost associated with IncF plasmids. ST131 clade C had dramatically changed the population structure of ExPEC, but it still remains unclear which features of this clade resulted in one of the most unprecedented AMR successes of the 2000s.
