AbstractIn this study, we used 10 healthy sheep, which gave birth to healthy twins. Stool samples were collected from mothers and their offspring 3 times during the study (0, 28 and 56 day postpartum). Milk samples were taken from the mothers at the same time. RT PCR analysis of faeces and milk was performed in order to assess the level of bacteria from the Firmicutes and Bacteroidetes phyla including the family Lactobacillaceae (phylum Firmicutes). The composition of mother's milk was also analyzed and their BCS. The data were compiled statistically. The obtained results showed that the level of the studied groups of bacteria may change due to the change of diet. Additionally, there were significant differences between lambs and mothers in the levels of the studied groups of bacteria. Analysis also shown that in the digestive system of mothers was a smaller disproportion in the level of the studied bacterial phyla than in lambs. The results also indicated the occurrence of differences in the bacterial composition at the individual level, both in ewes and their offspring. Additionally, in the conducted experiment, there were differences in the level of Firmicutes and Bacteroidetes groups depending on the sex.

Exercise significantly affects the body of both animals and humans, including the composition of the digestive microbiome. This study aimed to determine the changes in the composition of the most numerous bacterial phyla (Firmicutes and Bacteroidetes, as well as the level of the Lactobacillaceae family) in the digestive system of horses under the influence of physical effort. The study included a group of 17 Thoroughbred racehorses at the age of 3 years, fed the same forage, from whom feces samples were collected individually before and 48 h after physical effort. The obtained samples were subjected to DNA isolation and RT-PCR analysis. The results showed a significant increase in the level of both phyla after exercise compared to the state before physical effort; there were no such differences in the level of facultative aerobes, i.e., the Lactobacillaceae family (although a decreasing tendency was found after exercise). In addition, the analysis of the level of the studied phyla indicates individual differences in horses’ response to the effort.

ABSTRACTBifidobacteria are a major microbial component of infant gut microbiota, which is believed to promote health benefits for the host and stimulate maturation of the immune system. Despite their perceived importance, very little is known about the natural development of and possible correlations between bifidobacteria in human populations. To address this knowledge gap, we analyzed stool samples from a randomly selected healthy cohort of 87 infants and their mothers with >90% of vaginal delivery and nearly 100% breast-feeding at 4 months. Fecal material was sampled during pregnancy, at 3 and 10 days, at 4 months, and at 1 and 2 years after birth. Stool samples were predicted to be rich in the speciesBifidobacterium adolescentis,B. bifidum,B. dentium,B. breve, andB. longum. Due to high variation, we did not identify a clear age-related structure at the individual level. Within the population as a whole, however, there were clear age-related successions. Negative correlations between theB. longumgroup andB. adolescentiswere detected in adults and in 1- and 2-year-old children, whereas negative correlations betweenB. longumandB. brevewere characteristic for newborns and 4-month-old infants. The highly structured age-related development of and correlation networks between bifidobacterial species during the first 2 years of life mirrors their different or competing nutritional requirements, which in turn may be associated with specific biological functions in the development of healthy gut.

Abstract Abstract 2389 Osteolineage cells (OLCs) have been shown to participate in a regulatory bone marrow microenvironment for the hematopoietic stem and progenitor cells (HSPCs) – the endosteal niche. Our previous experiments using live animal imaging have demonstrated that single transplanted HSPCs preferentially home in close proximity to the individual OLCs. We hypothesized that these HSPC-proximal cells represent a distinct subpopulation of OLCs, which is specifically involved in a non-cell autonomous regulation of HSPC quiescence and self-renewal. To test this hypothesis, we developed a novel experimental platform, which allows visualization of HSPC-OLC cell pairs in-vivo and retrieval of the individual OLCs for molecular analysis. We intravenously injected DiI labeled adult bone marrow-derived FACS-sorted Lin−Sca1+c-kit+CD34−Flk2− HSPCs into irradiated newborn collagen 2.3GFP mouse recipients; in this transgenic strain, the majority of the OLCs are labeled with green fluorescent protein (GFP). 48 hours later, we sacrificed the animals and obtained fresh unfixed sections of femoral trabecular bone. Using a combination of differential interference contrast fluorescent microscopy, in-situ enzymatic digestion and micromanipulation, we harvested individual GFP-positive OLCs located within 2 cell diameters (“niche” OLCs) or greater than 5 cell diameters (“control” OLCs) from single DiI-bright HSPCs. Following reverse transcription and cDNA amplification with 29 cycles of PCR, as per the single cell RNA-Seq protocol (Tang et al, Nature Protocols 2010), we performed real-time RT-PCR analysis of 31 samples – 15 niche cells and 16 controls - for the OLC signature genes (osteocalcin, osterix) and for the genes implicated in playing a functional role in the HSPC-OLC cell interaction (osteopontin, CXCL12, angiopoietin 1). Transcripts for GAPDH, collagen 1 and GFP served as positive controls for the amplification. As expected, all cells were positive for GFP and over 85% cells expressed collagen 1. Osteopontin and CXCL12 were expressed at a similar level and frequency in the niche and control OLCs. However, we found that angiopoietin 1 transcripts were detected exclusively in the niche OLCs (3/15 versus 0/16, p <0.05 by Chi-squared). Moreover, niche OLCs were enriched for the osterix-positive cells (7/15 versus 2/16, p <0.05 by Chi-squared) and expressed a lower level of osteocalcin, as normalized for GAPDH expression (1.13 vs. 0.97, p< 0.05 by t-test). Our results suggest that niche OLCs may have a distinct molecular signature and reside within a population of very immature OLCs, as evidenced by the osterix + osteocalcin low phenotype. Further unbiased transcriptome characterization of these cells using genome-wide RNA-Seq assay is therefore likely to provide more evidence in support of our hypothesis and reveal novel non-cell autonomous regulators of HSPC quiescence. To our knowledge, this approach represents the first attempt to define molecular heterogeneity in-vivo at a single cell level using the micro-anatomical relationship between two heterologous cell types. Many researchers assume a one-dimensionality of gender ideology constructs and/or stability of dimensions across countries and time, yet these assumptions have rarely been tested. WE apply factor analyses on two waves of the International Social Survey Programme in 2002 and 2012, and comparable European countries to test this. Our results show that gender ideologies can be distinguished into distinctive domains that relate to mother’s employment, women’s work, men’s role in the family, and finally women’s breadwinning. These dimensions have be found to be relatively stable across countries and time. Results from regression models investigating different aspects of the gender division of labour suggest that distinguishing dimensions is less important when considering gender ideologies at the individual level but can make a big difference when examining gender culture at the country level.

The Scanian Economic-Demographic Database (SEDD) is a high-quality longitudinal data resource spanning the period 1646-1967. It covers all individuals born in or migrated to the city of Landskrona and five rural parishes in western Scania in southern Sweden. The entire population present in the area is fully covered after 1813. At the individual level, SEDD combines various demographic and socioeconomic records, including causes of death, place of birth and geographic data on the place of residence within a parish. At the family level, the data contain a combination of demographic records and information on occupation, landholding and income. The data for 1813-1967 was structured in the model of the Intermediate Data Structure (IDS). In addition to storing source data in the SEDD IDS tables, a wide range of individual- and context-level variables were constructed, which means that most types of analyses using SEDD can be conducted without the need of further elaboration of the data. This article discusses the source material, linkage methods, and structure of the database.

Family research aims to explore family processes, but is often limited to the examination of unidirectional processes. As the behavior of one person has consequences that go beyond that one individual, Family functioning should be investigated in its full complexity. The Social Relations Model (SRM; Kenny &amp; La Voie, 1984) is a conceptual and analytical model which can disentangle family data from a round-robin design at three different levels: the individual level (actor and partner effects), the dyadic level (relationship effects) and the family level (family effect). Its statistical complexity may however be a hurdle for family researchers. The user-friendly R package fSRM performs almost automatically those rather complex SRM analyses and introduces new possibilities for assessing differences between SRM-means and between SRM-variances, both within and between groups of families. Using family data on negative processes, different type of research questions are formulated and corresponding analyses with fSRM are presented.

Endozoochory (dispersion with passage through the digestive system of animals) is one of the most important processes. It is included in the ecological process known as dispersive mutualism, benefit the species involved. The most used method to evaluate the physiological quality of seeds is carried out based on the traditional germination test. However, electrical conductivity tests with seeds of forest species are still scarce, especially those developed with the individual method, which provide greater fidelity to the results so that through this one can define the test by the mass methods. The current work aimed to investigate the vigor, from the electrical conductivity of the seeds of Tournefortia paniculata Cham. after passage through the digestive tract of a ruby-crowned tanager Tachyphonus coronatus (Vieillot, 1822). An expedition was conducted in a fragmented area of Atlantic Forest in process of reforestation in the campus of the IFRJ. After the capture of the passerine with mist nets, proceeded the identification of seeds shed in its feces. The experimental design was completely randomized with one treatments with five replicates. The results allowed concluded that the seed of the T. paniculata Cham. after digestive tract of the ruby-crowned tanager presented high electrical conductivity in three of its replicates at the individual level.

Germline mutations in the DNA mismatch repair (MMR) genes cause Lynch syndrome (LS). Insertions of retrotransposons in MMR genes have been reported as a rare cause of LS. Here, we present a novel SINE-VNTR-Alu (SVA) insertion in exon 12 of MSH2 in an individual with early-onset colorectal cancer and strong LS family history. RT-PCR analysis indicated a larger aberrant MSH2 transcript in one of the family members. MSK-IMPACT next-generation sequencing testing and long-range PCR revealed an insertion in MSH2 exon 12 at the c.1972 position in an antisense orientation. The insertion was further characterized as an SVA element approximately 3 kb in length, belonging to the SVA_F1 family of retrotransposons.

The wars and armed conflicts in many regions of the world and especially in the Middle East have an eloquent impact on all life's happenings most especially in areas affected. Wars destroys community infrastructure such as health, education and other social services sectors. As of present, the ongoing conflicts in the Middle East had left a significant impact on the family and the individual in the areas affected. In the family, it has lead to disintegration and roles change. It is also lead to forced migration and asylum. At the individual level, it have impacted negatively in terms of psychology, social and physical wellbeing. Women and children are the most affected by traumatic events of war and they are the most vulnerable to all types of exploitation and abuse. The devastating effects of war on this vulnerable group can hardly be overemphasized. This paper discusses the problems faced by children and women in war and conflict zones. It also explores physical, mental health, social and educational crisis experienced by women and children in the Middle Eastern society. A society that is known to have witnessed many civil wars and armed conflicts. In addition, the study also discusses social and health services which must be provided to women and children in conflict affected area from the social work perspective.
