Abstract Although cyanogen ion (CN-) plays important role in industry which also bring acute environmental pollution. More serious, trace CN- enters the human body can cause serious consequences and even death. Therefore, it is of great significance to detect trace CN- with high sensitivity. Herein, a novel aggregation-induced emission (AIE) probe C-BH was synthesized based on coumarin matrix. Probe C-BH showed high selectivity and sensitivity toward CN- by dual channel response due to the excited state intramolecular proton transfer (ESIPT). The low detection limit was calculated to be 0.05 µM. Moreover, probe C-BH was successfully used for imaging CN- in living cells and zebrafish due to its low toxicity and excellent optical properties.

A novel rhodamine B-based lysosomal pH probe RML was developed. RML responded to acidic pH with short response time, high selectivity and high sensitivity and could detect lysosomal pH change in living cells.

AbstractThe carboxyl-terminal domain of RNA polymerase II (RNAP2) is phosphorylated during transcription in eukaryotic cells. While residue-specific phosphorylation has been mapped with exquisite spatial resolution along the 1D genome in a population of fixed cells using immunoprecipitation-based assays, the timing, kinetics, and spatial organization of phosphorylation along a single-copy gene have not yet been measured in living cells. Here, we achieve this by combining multi-color, single-molecule microscopy with fluorescent antibody-based probes that specifically bind to different phosphorylated forms of endogenous RNAP2 in living cells. Applying this methodology to a single-copy HIV-1 reporter gene provides live-cell evidence for heterogeneity in the distribution of RNAP2 along the length of the gene as well as Serine 5 phosphorylated RNAP2 clusters that remain separated in both space and time from nascent mRNA synthesis. Computational models determine that 5 to 40 RNAP2 cluster around the promoter during a typical transcriptional burst, with most phosphorylated at Serine 5 within 6 seconds of arrival and roughly half escaping the promoter in ~1.5 minutes. Taken together, our data provide live-cell support for the notion of efficient transcription clusters that transiently form around promoters and contain high concentrations of RNAP2 phosphorylated at Serine 5.

Rhodamine B thiohydrazide (RBS) was firstly employed as turn-on fluorescent probe for hypochlorite in aqueous solution and living cells. It exhibits a stable response to hypochlorite from 1.0×10-6to 1.0×10-5M with a detection limit of 3.3×10-7M. The response of this probe to hypochlorite is fast and highly selective compared with other reactive oxygen species (such as.OH,1O2, H2O2) and other common anions (such as X-, ClO2-, ClO4-, NO3-, NO2-, OH-, Ac-, CO32-, SO42-).
