Efficient SNP-based tests of association for quantitative phenotypes using pooled DNA

AbstractWhat makes a flagship species effective in engaging conservation donors? Large, charismatic mammals are typically selected as ambassadors, but a few studies suggest butterflies—and monarchs in particular—may be even more appealing. To gather more information about people’s responses to monarchs, we conducted an empirical study of member submissions to a successful conservation campaign, the Monarch Story Campaign, conducted by the Environmental Defense Fund (EDF). The set of 691 stories along with their associated demographic and donation data was analyzed in a mixed-methods study using qualitative analysis and tests of association. The results showed that people often described encounters with monarchs in childhood and as adults. They expressed strong, positive emotions, and lauded the monarch’s beauty and other “awe-inspiring” qualities and expressed wonder at their lifecycle (i.e., metamorphosis and migration). They also raised conservation themes of distress at monarch loss, calls for action, and caretaking, such as being “fragile” and “in need.” Sharing personal encounters was associated with current efforts to save the species and more past financial donations, while a second pattern tied more donations to awe at the monarch’s mass migration. These results imply that conservation campaigns built around species people encounter may build lifelong awareness, concern, and actions towards conservation.

Abstract Epidemiology relies on understanding the distribution of pathogens which often can be detected through DNA-based techniques, such as quantitative Polymerase Chain Reaction (qPCR). Typically, the DNA of each individual sample is separately extracted and undergoes qPCR analysis. However, when performing field surveys and long-term monitoring, a large fraction of the samples is generally expected to be negative, especially in geographical areas still considered free of the pathogen. If pathogen detection within a population – rather than determining its individual prevalence – is the focus, work load and monetary costs can be reduced by pooling samples for DNA extraction. We test and refine a user-friendly technique where skin swabs can be pooled during DNA extraction to detect the amphibian chytrid fungi, Batrachochytrium dendrobatidis and B. salamandrivorans (Bsal). We extracted pools with different numbers of samples (from one to four swabs), without increasing reaction volumes, and each pool had one sample inoculated with a predetermined zoospore amount. Pool size did not reduce the ability to detect the two fungi, except if inoculated with extremely low zoospore amounts (one zoospore). We confirm that pooled DNA extraction of cutaneous swabs can substantially reduce processing time and costs without minimizing detection sensitivity. This is of relevance especially for the new emerging pathogen Bsal, for which pooled DNA extraction had so far not been tested and massive monitoring efforts in putatively unaffected regions are underway.

Purpose: The aims of this study were to describe the demographic characteristics of pediatric dentistry patients undergoing dental rehabilitation under general anesthesia (DRGA) at UNC-Chapel Hill during the last 13 years and identify factors associated with multiple (1 versus 2 or more) DRGA visits during that timeframe. Study design: Administrative claims data were used to identify children and adolescents (age <18 years) who underwent DRGA between 1/1/2002 and 12/31/2014 at the UNC Hospitals system. Information on children's age, sex and all treatment-associated CDT codes were collected. Descriptive statistics and bivariate tests of association were used for data analyses. Results: There were 4,413 DRGAs among 3,973 children (median age=4 years 8 months, males=55%) during the study period. The annual rate of DRGAs increased over time, peaking (n=447) in 2013. Overall, 9% of children had ≥2 visits with repeat rates up to 18%. There was no association between children's sex and receipt of one versus multiple DRGAs; however, craniofacial cases were more likely (p<0.0005) to have multiple DRGAs compared to non-craniofacial ones. Conclusion: DRGAs are on the increase—with the exception of craniofacial and special health care needs patients, multiple DRGAs may be reflective of sub-optimal adherence to preventive and continuing care recommendations.
