Alterations in the Effects of Warfarin in Dogs by Halofenate

SummaryThe interaction between warfarin and the new lipid lowering agent halofenate (MK 185) [2 - acetamidoethyl (p-chlorophenyl) (m-trifluoromethylphenoxy) acetate] was studied in dogs in both short- and long-term experiments. Our data suggest that halofenate potentiates the anticoagulant effect of warfarin by increasing the degradation of prothrombin (factor II) (Kdeg on placebo = 211 ± 32 × 10−4 × Hr−1 mean ± SEM; on halofenate = 268 ± 39 × 10−4 × Hr−1 mean ± SEM; P < 0.01). However, a concomitant increase in factor II synthesis of 34% results in resistance to warfarin’s effect if halofenate is administered prior to warfarin. The mean prothrombin time of 4 dogs on 2 mg of warfarin following halofenate pretreatment for 8 weeks was 74.8% ± 17.3 (SE) of the anticoagulated control dog. On 2 mg of warfarin alone, it was 133.7% ± 42.0 (P < 0.001). Cessation of halofenate from combined therapy results in a delayed augmentation of warfarin effect. These data suggest that the nature of the interaction between warfarin and drugs such as halofenate which alter the kinetics of prothrombin may depend on the sequence of administration.

An experiment, designed to test different ways of using straw with fertilizers, and involving a three course rotation of crops, was carried out at Rothamsted between 1933 and 1951. The methods of analysis developed for this experiment are described in the present paper and demonstrated using yields of potatoes.Treatment effects of interest are given by the mean yields over all years and the linear regressions of yield on time. These estimates are straightforward but the evaluation of their errors is complicated by the existence of correlations due to the recurrence of treatments on the same plots. Further complications are introduced when, as frequently happens in long-term experiments, treatment effects show real variation from year to year. A method is given for estimating standard errors which include a contribution from this variation.The various relationships between yields and the uncontrolled seasonal factors can also be examined; in the present experiment there is some indication that the effects of treatments on yields of potatoes are influenced by the dates of planting.In other circumstances the analysis requires modifications, some of which are briefly considered.

Abstract. A generic system dynamics watershed (GSDW) model is developed and applied to five reconstructed watersheds located in the Athabasca mining basin, Alberta, Canada, and one natural watershed (boreal forest) located in Saskatchewan, Canada, to simulate various hydrological processes in reconstructed and natural watersheds. This paper uses the root mean square error (RMSE), the mean absolute relative error (MARE), and the correlation coefficient (R) as the main performance indicators, in addition to the visual comparison. For the South Bison Hills (SBH), South West Sand Storage (SWSS) and Old Aspen (OA) simulated soil moisture, the RMSE values ranges between 2.5–4.8 mm, and the MARE ranges from 7% to 18%, except for the D2-cover it was 26% for the validation year. The R statistics ranges from 0.3 to 0.77 during the validation period. The error between the measured and simulated cumulative actual evapotranspiration (AET) flux for the SWSS, SBH, and the OA sites were 2%, 5%, and 8%, respectively. The developed GSDW model enables the investigation of the utility of different soil cover designs and evaluation of their performance. The model is capable of capturing the dynamics of water balance components, and may used to conduct short- and long- term predictions under different climate scenarios.

Objective: We examined short and long-term outcomes of bariatric surgery in patients with obesity and type 1 diabetes mellitus (T1DM). Methods: We reviewed the records of all adults insured by Maccabi Healthcare Services during 2010 -2015, with body mass index (BMI) ⩾30 kg/m2 and T1DM; and compared weight reduction and glucose control according to the performance of bariatric surgery. BMI and glycated hemoglobin (HbA1c) levels were extracted for baseline and every 6 months, for a mean 3.5 years. Results: Of 52 patients, 26(50%) underwent bariatric surgery. Those who underwent surgery were more often female and with a longer duration of diabetes. Immediately postoperative, 4(15%) developed diabetic ketoacidosis, while 6(23%) experienced severe hypoglycemic episodes. The mean BMI decreased among surgery patients: from 39.5±4.4 to 30.1±5.0 kg/m2 ( p < 0.0001); and increased among those who did not undergo surgery: from 33.6±3.9 to 35.1±4.4 kg/m2 ( p = 0.49). The mean HbA1c level decreased during the first 6 months postoperative: from 8.5±0.9% to 7.9±0.9%; however, at the end of follow-up, was similar to baseline, 8.6±2.0% (p = 0.87). For patients who did not undergo surgery, the mean HbA1c increased from 7.9±1.9% to 8.6±1.5% ( p = 0.09). Conclusions: Among individuals with obesity and T1DM, weight loss was successful after bariatric surgery, but glucose control did not improve. The postoperative risks of diabetic ketoacidosis and severe hypoglycemic episodes should be considering when performing bariatric surgery in this population.

BACKGROUND & OBJECTIVE: Peptic ulcer disease (PUD) is one of the common presentations in the medical as well as Gastroenterology clinics and it can end up in various short- and long-term complications among which osteoporosis is highly under rated. Therefore, the present study was designed to determine the frequency of osteoporosis in cases having PUD. METHODOLOGY: A cross-sectional study was conducted at the Department of Medicine during 1st January 2020 to 30th June 2020 in Sheikh Zayed Hospital, Lahore. The diagnosed cases of PUD on the basis of history and endoscopic findings with the age range of 20 to 60 years were included. Osteoporosis was labelled as yes, where a T-score of ≤ -2.5 on DEXA scan was noted. RESULTS: In this study, there were a total of 139 cases, out of which 75 (53.95%) were males and 64 (46.05%) females. The mean age of the participants was 48.11±9.43 years. Osteoporosis was observed in 14 (10.07%) out of 139 cases. Osteoporosis was seen in 05 (6.67%) males vs 09 (14.06%) females in their respective groups with p= 0.148. Osteoporosis was observed in 4 (5.80%) cases with a duration of PUD up to 2 years and 10 (14.28%) cases with duration more than this with p= 0.096 CONCLUSION: Osteoporosis is not uncommon in cases with Peptic ulcer disease, and there is no significant association with any of the confounders of this study.

Aim of study: to evaluate the results of draining and resection interventions in patients with pancreatic cystic lesions. Materials and methods. A retrospective analysis of the treatment of 50 patients with pancreatic cystic lesions of various etiologies was conducted. All patients included in the study underwent surgical treatment, namely 20 (40.0%) resection interventions (RI) and 30 (60.0%) draining interventions (DI). Comparison of short- and long-term outcomes of treatment between the two groups was carried out. Results. There were no statistically significant differences in body mass index in the compared groups. The average duration of surgery in DI group was significantly lower than in RI group (142.5±4.9 and 278.5±6.9, p<0.001). The mean intraoperative blood loss, as well as the length of hospitalization, was significantly lower in DI group as compared with RI (390±28.4 ml, 500±27.4 ml, p=0.008; 8.5±0.9 days, 13.8±3.9 days, p<0.001). The severity of postoperative complications was proved to be significantly higher in RI group (p<0.05). However, when analyzing long-term results, the attacks-free course of the disease was observed only in RI group. Conclusion. If technically possible, DI is more preferable than RI in patients with pancreatic pseudocysts. RI should be performed in patients with cystic neoplasia.

Abstract The postmenopausal period is characterized by a decrease in the hormonal supply which is associated with Alzheimer's Disease (AD). Vitamin D is neuroprotective and can be used in combination with pre-existing medications to improve its effects. The objective was to evaluate the effect of vitamin D associated with memantine and donepezil in female mice submitted to ovariectomy (OVX) for 5 months and subjected to an AD-induced dementia model. Animals were divided into 5 groups who received 17 days of treatment and were subjected to behavioral tests. The animals underwent euthanasia at 18th day. OVX groups exhibit reduced levels of E2 and triple treatment group had high levels of vitamin D. The induction of dementia with OVX induced short- and long-term spatial and habituation memories damage. Also, induced reduction of BDNF and IL-4 levels in hippocampus, and increasing levels of TNFα in hippocampus and of IL-1β in hippocampus and frontal cortex of animals, as well as a significant increase on GFAP immunoreactivity. Triple-association treatment reversed the effects of long-term spatial and habituation memories damage, as well as reversed changes in TNFα, IL-1β, IL-4 and GFAP immunoreactivity levels in hippocampus of treated animals. Therapeutic association has beneficial effects on memory and inflammation parameters in female mice subjected to OVX and the AD animal model of dementia.
