Abstract Background Natural glycolysis encounters the decarboxylation of glucose partial oxidation product pyruvate into acetyl-CoA, where one-third of the carbon is lost at CO2. We previously constructed a carbon saving pathway, EP-bifido pathway by combining Embden-Meyerhof-Parnas Pathway, Pentose Phosphate Pathway and “bifid shunt”, to generate high yield acetyl-CoA from glucose. However, the carbon conversion rate and reducing power of this pathway was not optimal, the flux ratio of EMP pathway and pentose phosphate pathway (PPP) needs to be precisely and dynamically adjusted to improve the production of mevalonate (MVA). Result Here, we finely tuned the glycolytic flux ratio in two ways. First, we enhanced PPP flux for NADPH supply by replacing the promoter of zwf on the genome with a set of different strength promoters. Compared with the previous EP-bifido strains, the zwf-modified strains showed obvious differences in NADPH, NADH, and ATP synthesis levels. Among them, strain BP10BF accumulated 11.2 g/L of MVA after 72 h of fermentation and the molar conversion rate from glucose reached 62.2%. Second, pfkA was finely down-regulated by the clustered regularly interspaced short palindromic repeats interference (CRISPRi) system. The MVA yield of the regulated strain BiB1F was 8.53 g/L, and the conversion rate from glucose reached 68.7%. Conclusion This is the highest MVA conversion rate reported in shaken flask fermentation. The CRISPRi and promoter fine-tuning provided an effective strategy for metabolic flux redistribution in many metabolic pathways and promotes the chemicals production.

ABSTRACT We have found and analysed 16 multipeaked type-I bursts from the neutron-star low-mass X-ray binary 4U 1636 − 53 with the Rossi X-ray Timing Explorer (RXTE). One of the bursts is a rare quadruple-peaked burst that was not previously reported. All 16 bursts show a multipeaked structure not only in the X-ray light curves but also in the bolometric light curves. Most of the multipeaked bursts appear in observations during the transition from the hard to the soft state in the colour–colour diagram. We find an anticorrelation between the second peak flux and the separation time between two peaks. We also find that in the double-peaked bursts the peak-flux ratio and the temperature of the thermal component in the pre-burst spectra are correlated. This indicates that the double-peaked structure in the light curve of the bursts may be affected by enhanced accretion rate in the disc, or increased temperature of the neutron star.

ABSTRACT We present the first optical identification and confirmation of a sample of supernova remnants (SNRs) in the nearby galaxy NGC 3344. Using high spectral and spatial resolution data, obtained with the CFHT imaging Fourier transform spectrograph SITELLE, we identified about 2200 emission line regions, many of which are H ii regions, diffuse ionized gas regions, and also SNRs. Considering the stellar population and diffuse ionized gas background, which are quite important in NGC 3344, we have selected 129 SNR candidates based on four criteria for regions where the emission lines flux ratio [S ii]/H α ≥ 0.4. Emission lines of [O ii] λ3727, H β, [O iii] λλ4959,5007, H α, [N ii] λλ6548,6583, and [S ii] λλ6716,6731 have been measured to study the ionized gas properties of the SNR candidates. We adopted a self-consistent spectroscopic analysis, based on Sabbadin plots and Baldwin, Phillips & Terlevich diagrams, to confirm the shock-heated nature of the ionization mechanism in the candidates sample. With this analysis, we end up with 42 Confirmed SNRs, 45 Probable SNRs, and 42 Less likely SNRs. Using shock models, the confirmed SNRs seem to have a metallicity ranging between Large Magellanic Cloud and 2×solar. We looked for correlations between the size of the confirmed SNRs and their emission lines ratios, their galaxy environment, and their galactocentric distance: We see a trend for a metallicity gradient among the SNR population, along with some evolutionary effects.

Ti6Al4V alloy is promising biology material with outstanding properties of low density, high specific strength, and exceptional corrosion resistance. However, one of its disadvantages is the poor tribological property. In this paper Mo-N hard surface modification layers were formed on Ti6Al4V at 900°C substrate temperature by plasma reactive sputtering. The flux ratio N2/Ar is an important parameter and its influence on the composition, structure and hardness of the Mo-N layers is studied. The Mo-N layers are duplex layers, composed of diffusing layer and surface coating. The component of Mo and N elements in the diffusing layer changes gradually which can enhance the load-bearing capacity to the coating and ensure the durability of the coating. With the increase of the flux ratio N2/Ar, the content of N element in the Mo-N layers increases. The Mo-N layers were polycrystalline γ- Mo2N with (200) plane oriented parallel to the substrate surface. The surface hardness of the formed layers is in the range HK1330-1430. The hardness of the Mo-N layers increases with the increase of the flux ratio N2/Ar and the reason is that the content of N element in the Mo-N layers increases.
