Pregnancy rate and prolificacy after artificial insemination in ewes following synchronisation with prostaglandin, sponges, or sponges with bactericide

Pregnancy rate and prolificacy were studied in ewes after cycle synchronisation by either progestagen sponges plus equine chorionic gonadotrophin (eCG) or by three injections of prostaglandin (PG). We also tested whether there was any advantage in treating the sponges with antibiotic before insertion. In Experiment 1, 207 Corriedale ewes were treated with intravaginal sponges for 14 days and given 250 IU eCG at sponge withdrawal. For half of the ewes, the sponges had been sprayed with chlortetracycline whereas the other half received untreated sponges. Ewes were ranked within each group based on the amount of mucus and odour of the sponges at the time of withdrawal (score 0 = none, + = mild, ++ = abundant) and the result was compared with pregnancy rate. An average of 155 ± 5.5 million spermatozoa (range 96–248 million) were deposited in the vagina 55 h after sponge withdrawal. The actual dose was measured for each ewe so the dose effect could be studied. Adding antibiotics reduced the amount of mucus (57% of ewes with score 0 in the antibiotic group versus 31% in the Control group; P < 0.01) and odour (98% of ewes with score 0 in the Antibiotic group versus 11% in the Control group; P < 0.001) but had no effect on pregnancy rate (58% for the Antibiotic group versus 48% of controls; P > 0.05) at any sperm dose. In Experiment 2, Merino ewes were treated with intravaginal sponges for 14 days and given 200 IU eCG at sponge removal (n = 100), or were subjected to three PG injections 7 days apart (n = 100). Intrauterine insemination with 200 million sperm was carried out 53 h after the end of synchronisation treatment. Pregnancy rate was higher in the sponge plus eCG group than in the PG group (85 versus 47%; P < 0.001) but prolificacy was similar (1.34 versus 1.38; P > 0.05). We conclude that, under the conditions of these experiments, synchronisation with sponges plus eCG and PG resulted in similar prolificacy, but pregnancy rate was significantly lower with the three PG injections. There seems to be no benefit for pregnancy rate of pretreating sponges with chlortetracycline.

In all domestic species, acceptable pregnancy rate with frozen-thawed semen is 20-30% through simply cervical and vaginal insemination. Researchers have proved that achievable high results can be expected only from intrauterine insemination in ewes. Unlike the cow, and goat, the cervix of ewes is a formidable barrier to penetrate for transcervical intrauterine insemination. It is not only due to the physical size of the canal and a narrow luminal diameter (even at estrus), but also due to its caudally facing eccentric series of four to eight funnel-like rings. The major aim of this study was to evaluate a comparison of transcervical and laparoscopic intrauterine artificial insemination techniques with frozen –thawed semen on the reproductive performance of ewes.58ewes of 3-4 years of age, so called Altanbulag subspecies of breed were selected and equally divided into 2 groups for the AI methods from October 15-20, 2013 in the breeding station of Research Institute of Animal Husbandry in Hongor sum, Darhan uul province. The estrus was synchronized with EAZI-BREED TMCIDR in which the progesterone content was 0.3 g. After 12 days, the CIDR were removed, PMSG of 400 and 200 IU were administrated intramuscularly per ewe. The total success of synchronization rate was 93.3%. The results has shown the possibilities of laparoscopic and transcervical AI techniques in 55.17%, 41.37% (p>0.05) of pregnancy rates respectively, with frozen-thawed semen in ewes by ultrasonography diagnosis.Mongolian Journal of Agricultural Sciences Vol.13(2) 2014: 43-47

Background: Polycystic ovarian syndrome (PCOS) is among the important causes of infertility in young women. Premature luteinizing hormone (LH) surge (PLS) is one of its complications. PLS can reduce the quality of oocytes and therefore decrease the success of intrauterine insemination (IUI). Letrozole, a non-steroidal aromatase inhibitor, prevents LH surge. In this study, we aim to evaluate the effects of letrozole on preventing premature LH surge in clomiphene-resistant patients with PCOS undergoing IUI. Materials and Methods: In this randomized clinical trial, 131 patients who were developed with PCOS were selected for IUI cycle, divided into two groups randomly: control group (n=67) and letrozole group (n=64). Incidence of premature LH surge, pregnancy, abortion and ongoing pregnancy rate, endometrial thickness and number of follicles were measured in both groups. Results: No significant difference was seen between mean ages in the two groups; 11.9% of the control group and 21.9% of the letrozole group became pregnant (P =0.005); furthermore, premature LH surge was seen in 4.7% of the letrozole group and 8.9% of the control group (P =0.003). E2 and Endometrial thickness was higher in letrozole group; however, LH was significantly higher in the control group (P =0.026). Conclusion: Administration of letrozole in clomiphene-resistant patients with PCO undergoing IUI cycle can decrease the incidence of PLS. In addition, it can increase pregnancy rate significantly. Therefore, using letrozole is more reasonable in patients who have not responded to clomiphene or are hypersensitive. [GMJ.2015;4(3):104-11]

ABSTRACT: The aim of this study was to reduce the waiting voluntary period in dairy cows by administration of two different hormonal protocols for estrus synchronization and fixed-time artificial insemination (FTAI). One hundred twenty-eight Holstein dairy cows were studied. Three groups (G) were carried out: G1 (44 treated), G2 (34 treated) and G3 (50 as control). In the treated cows the following hormonal protocols was applied between 40 and 45 days post partum: G1- Day 0 (D0) - bovine intravaginal device containing progesterone (P4) plus 1.0 mg of estradiol benzoate (EB), D8 - removal of the P4, plus 0.5 mg of cloprostenol plus 400 IU of equine chorionic gonadotropin (eCG), D9 - 1.0 mg of EB and FTAI after 36 hours. G2 - D0 - P4 intravaginal plus 50 mcg of gonadotropin-releasing hormone (GnRH), D7 - removal of the P4 plus cloprostenol plus 400 IU eCG; D9 - 50 mcg of GnRH and FTAI 16 hours after. G3 – Control group inseminated 12 hours after the visual estrus. Results: G1 and G2 were inseminated (FTAI) respectively in the D55.5 and D56.8 after calving, while the G3 in the D84.5 (p<0.01).  The pregnancy rate in the G1, G2 and G3 was respectively 47.0, 46.1 and 35.8% (p>0.05).  In Conclusion: there was significant reduction in the voluntary waiting period in treated cows and higher pregnancy rate although not significant.

Artificial insemination results can be improved using fixed-timed AI (FTAI). Currently, progesterone release devices, indispensable in FTAI procedures, such as DIB® (Schering-Plough, Kenilworth, NJ, USA), are made of silicone, a nonbiodegradable biocompatible polymer. Recently, biodegradable progesterone (P4)-releasing devices made of poly(3-hydroxy)butyrate-valerate copolymer (PHB-V) and poly-ϵ-caprolactona (PCL) have been developed. To investigate the efficiency on pregnancy rate of FTAI programs using a new intravaginal device made of PHB-V (Progestar®, Innovare, São Paulo, Brazil), 240 Nellore (Bos indicus) cows, between 3 and 8 years of age, were randomly assigned to 2 experiments. In experiment 1, new P4 devices DIB (control group, DIB1; n = 60) were compared with PHB-V (Progestar1; n = 76); in experiment 2, previously used DIB (DIB2; n = 52) and PHB-V (Progestar2; n = 52) devices were compared. All groups were treated with 2 mg of estradiol benzoate i.m. (Innovare) and received the intravaginal device according to group (Day 0). Together with P4 device withdrawal (Day 8) all animals received 300IU of eCG i.m. (Novormon®, Schering-Plough) and 150 μg i.m. of D-cloprostenol (Croniben®, Biogenesis-Bagó, Buenos Aires, Argentina). On Day 9, all cows were treated with 1 mg of estradiol benzoate and the FTAI was performed 30 h later. The data were submitted to ANOVA (PROC GLM of SAS, SAS Institute, Cary, NC, USA), separating as causes of variance the effect of treatment, postpartum period (PPP1-45 until 70 days and PPP2-71 until 90 days), body condition score (BCS4, BCS5, and BCS6 using a 1 to 9 scale), cyclicity (cycling and noncycling) and clean-up bulls (5 bulls nominated with numbers 1 to 5). Significant differences were considered to have P < 0.05. In experiment 1, interactions were not significant, there was no effect of treatment on FTAI (DIB1: 66.7% v. Progestar1: 65.8%) or breeding season pregnancy rates (BSPR; DIB1: 95.0% v. Progestar1: 94.7%) and no effect of other variables on FTAI pregnancy rates (FTAIPR) and BSPR. In experiment 2 there was no effect of treatment on FTAIPR (DIB2: 61.6% v. Progestar2: 55.8%) and BSPR (DIB2: 94.2% v. Progestar2: 88.5%). As ANOVA indicated a significant effect of clean-up bulls on BSPR, Tukey test was used to split it up. A bull of the Progestar2 group had lower performance (41.7%) causing the difference (albeit nonsignificant) on BSPR. This experiment showed that Progestar and DIB had the same efficiency on pregnancy rate and both treatments have the potential to produce FTAIPR >50% and BSPR >90%. Objective. To compare pregnancy rate after controlled ovarian hyperstimulation and intrauterine insemination (COH-IUI) with no treatment in patients with endometriosis-associated infertility treated with laparoscopy.Design. A clinical cohort study.Setting. University-level tertiary care center.Patients. 238 women with various stages of endometriosis after laparoscopic treatment.Interventions. Either COH-IUI or follow-up for 12 months.Main Outcome Measures. The primary outcome measures were clinical pregnancy and live birth rate. Predictive factors evaluated were female age, maternal BMI, and duration of infertility.Results. The pregnancy rate attained after the integrated laparoscopy–COH-IUI approach was 53.4%, while it was significantly lower (38.5%) in the control group. Similarly, a significant difference was observed in live births (48.3% versus 34.2%). Patients with severe endometriosis were less likely to achieve pregnancy (38%) and live birth (35%) than their counterparts with milder forms (57% and 53%).Conclusions. In patients with endometriosis-based infertility, surgery followed by COH-IUI is more effective than surgery alone.

The goal of this study was to evaluate the effects of strategic insulin administration on follicular growth, progesterone production, and pregnancy rate in beef cattle. Two experiments were conducted, in experiment 1, 215 cows crossbreed cows (Bos indicus × Bos taurus) (Control Group, n = 109; Insulin Group, n = 106) were submitted to fixed-time artificial insemination (TAI) with application of 2 mg of estradiol benzoate and a vaginal device containing progesterone on day 0. On day 9, the device was removed and 0.5 mg of estradiol cypionate was administered with 12.5 mg of dinoprost and 0.25 UI kg-1 of insulin for the treated animals. On day 11, artificial insemination (AI) was performed on all animals. On days 9 and 11 according to the protocol, was evaluated follicular growth and estrus manifestation, and 30 days after AI the pregnancy rate. In experiment 2, 85 cows were utilized, including Aberdeen Angus, Hereford, and crossbreeds (Control Group, n = 49 and Insulin Group, n = 36), and were submitted to the protocol for fixed-time embryo transfer (FTET), and progesterone concentrations were evaluated. On day 0, the cows received 2 mg of estradiol benzoate and a vaginal device containing progesterone. On day 5 of the protocol, 0.150 mg of cloprostenol was administered. On day 8, the vaginal device was removed and 20 mg of purified porcine pituitary extract and 0.25 UI kg-1 of insulin was given to the animals in the treatment group, and on day 9, 1 mg of estradiol benzoate was administered. On day 17, an embryo transfer was performed and blood collection for evaluation of progesterone levels in animals submitted to FTET were collected. In experiment 1, the follicular growth rate and the pregnancy rate were similar between groups (P > 0.05) and in experiment 2, the production of P4 was not different between the animals that received the insulin application and the control group. The utilization of a single dose of insulin for beef cows did not affect the ovulatory follicular diameter, progesterone concentrations, or pregnancy rate.

ABSTRACT This study aimed to determine the efficiency of estradiol cypionate (EC) as an ovulation inducer in a Timed Artificial Insemination protocol. 69 buffalo cows received an intravaginal progesterone device and 2mg of estradiol benzoate (EB) at D0. On D9, the intravaginal device was removed and 0.53mg of prostaglandin (PGF2α) and 400UI of equine chorionic gonadotrophin (eCG) were applied. The cows were distributed into two groups: the first group received 1mg of EC (ECG) in D9, and the second group received 1mg of EB (EBG) in D10. Inseminations occurred on D11. Ovarian activity and pregnancy diagnosis were analyzed by ultrasonography. There was no difference (P>0.05) in follicular diameter (9.6 ± 0.89mm vs. 10.7 ± 1.12mm; P=0.06), in ovulation rate (90.9% vs. 90.9%; P=1) and pregnancy rate (58.8% vs. 62.9%; P=0.79), however, buffalo cows from the ECG treatment have less time between P4 removal and ovulation when compared to EBG buffalos (37.4h vs. 52.8h; P=0.001), respectively. Thus, it was concluded that the implantation of TAI in the floodplain of Amazonas is feasible and the use of EC results in successful rates, similar to EB.

A fibreoptic endoscope procedure for non-surgical deep intrauterine insemination in non-sedated sows has been reported. However, the endoscope is an expensive and fragile instrument, and is unsuitable for use under field conditions. The aim of this study was to determine the minimum number of spermatozoa required to maintain optimal fertility using a flexible catheter (1.8 m in length, 4 mm in diameter) for deep intrauterine insemination in 2-6 parity non-sedated sows. Crossbred sows were treated with eCG 24 h after weaning and with hCG 72 h later to induce oestrus. Deep intrauterine insemination was performed 36 h after hCG treatment in 117, 126, 60 and 69 sows with 15.0, 5.0, 2.5 or 1.0 x 10(7) spermatozoa in 10 ml, respectively. Weaned sows (n = 147) not treated with hormones and used for standard artificial insemination (AI) (two inseminations per oestrus with 3 x 10(9) spermatozoa in 100 ml) served as controls. The flexible catheter was passed successfully through the cervix into one uterine horn in 95.4% of the sows in an average of 3.7 +/- 0.09 min. Farrowing rates after deep intrauterine insemination with 15 or 5 x 10(7) spermatozoa did not differ from those of the control group (82.9, 76.2 and 83.0%, respectively), but a significant decrease (P < 0.001) was observed in sows inseminated with 2.5 or 1.0 x 10(7) spermatozoa (46.7 and 39.1%, respectively). In contrast, the number of spermatozoa inseminated did not affect prolificacy. Laparotomy revealed that the tip of the flexible catheter reached approximately the anterior third of the uterine horn. Although deep intrauterine insemination was performed in only one uterine horn, the percentages of embryos collected from the tip of both uterine horns 2 days after deep insemination were not significantly different. The results show that in comparison with standard AI, a 20-60-fold reduction in the number of spermatozoa inseminated and an 8-10-fold reduction in the dose volume can be achieved without decreasing fertility when semen is deposited non-surgically into the upper first third of one uterine horn.
