Effects of waterlogging, rootstock and salinity on Na, Cl and K concentrations of the leaf and root, and shoot growth of sultana grapevines

Potted Sultana vines ( Vitis vinifera L. cv. Sultana) on own-roots or grafted onto Ramsey, Harmony, Schwarzmann or 1613 rootstocks were irrigated with solutions containing 1, 10, 20, 40 or 60 mM NaCl. Half the vines had free-draining rootzones and the other half were waterlogged for the first week in a 2-week cycle. The vines were harvested after seven cycles. Raising the irrigation salinity from 1 to 60 mM caused growth to decline by 47% in vines with free-draining rootzones and by 61% in vines with waterlogged rootzones. Under saline conditions, the use of chloride excluding rootstock reduced leaf chloride concentration by 60% in vines with free-draining rootzones but by only 18% in vines with waterlogged rootzones. Waterlogging decreased the root chloride concentration in all rootstocks. The leaf potassium concentration was reduced by waterlogging at irrigation salinities less than 20 mM NaCl and increased by waterlogging at higher salinities. Waterlogging altered the relative effects of rootstock on leaf potassium.

История виноградарства на Дону насчитывает несколько веков, местные сорта винограда многообразны и специфичны. Микросателлитные маркеры широко используются для генотипирования сортов и подвоев винограда, при изучении происхождения сортов и анализе их родословной. Целью исследования было изучение выборки редких и малораспространенных автохтонных донских сортов и сравнение их с другими аборигенными донскими генотипами на основе данных ДНК-анализа. В исследования включены 23 стародавних донских сорта. Генотипирование проводили методом микросателлитного профилирования. В исследовании использовали микросателлитные маркеры (SSR), рекомендованные в качестве основного минимального набора для ДНК-паспортизации сортов вида Vitis vinifera L.: VVMD5, VVMD7, VVMD27, VVS2, VrZAG62 и VrZAG79. По результатам проведенного анализа все изученные образцы показали сорт-специфическую комбинацию аллелей в идентифицированных ДНК-профилях. Количество выявленных аллелей составило в среднем 8 аллелей/локус. Наибольший полиморфизм в исследовании этой группы донских сортов был обнаружен в локусе VVMD5: идентифицировано 10 аллелей на локус, наименьшее - в локусе VrZAG62: 6 аллелей/локус. Основываясь на данных SSR-анализа, степень генетического сходства сортов оценивали с использованием метода UPGMA. Кластерный анализ матрицы генетических дистанций, созданный на основе выявленных значений аллелей в шести микросателлитных локусах исследуемых сортов, определил несколько групп генотипов. Сорт Красностоп золотовский выделился в отдельную ветвь, что указывает на различия между этим генотипом и другими сортами исследуемой выборки. Наивысший уровень генетического сходства наблюдался между следующими парами сортов: Крестовский и Бургундский, Шилохвостый и Мушкетный, Кумшацкий черный и Ефремовский.The history of viticulture on the Don goes back several centuries. Local grapevine varieties are diverse and peculiar. Microsatellite markers are widely used in genotyping grapevine cultivars and rootstocks, in grapevine origin and breeding background analysis. Our study aimed to examine samples of rare and less common autochthonous Don varieties, and compare them with the other aboriginal Don genotypes using DNA data. The study involved 23 traditional Don varieties. The genotyping was done by microsatellite profiling. The study used microsatellite (SSR) markers recommended as the basic minimum set for DNA-certification of the genotypes of Vitis vinifera L.: VVMD5, VVMD7, VVMD27, VVS2, VrZAG62 and VrZAG79. Based on the findings, all the studied samples demonstrated variety-specific combination of alleles in the identified DNA profiles. The number of detected alleles on average was 8 alleles/locus. The greatest polymorphism in the studied group of Don varieties was detected in VVMD5 locus: 10 alleles per locus were identified, the smallest in VrZAG62 locus: 6 alleles/locus. UPGMA method was used to assess the extent of genetic similarity of the varieties based on SSR-genotyping data. Based on determined allele values of the studied varieties, cluster analysis of the genetic distances matrix determined several groups of genotypes. ‘Krasnostop zolotovskiy’ variety stood out as a separate branch, which indicates the difference between this genotype and the other varieties of the studied sampling. The highest level of genetic similarity was observed between the following pairs of varieties: ‘Krestovskiy’ and ‘Burgundskiy’, ‘Shilohvostyi’ and ‘Mushketnyi’, ‘Kumshatskiy chornyi’ and ‘Efremovskiy’.

The quality of grapes is determined above all by the contents of the primary and secondary metabolites. The primary metabolites involve sugars and organic acids, and just these compounds are dealt with in this study. Its objective was to analyse and critically evaluate the primary metabolites in new interspecific varieties and, based on a comparison with European varieties of grapevine (Vitis vinifera L.), to find out the similarities and also possible differences between them. The study evaluates and compares 4 conventional varieties of Vitis vinifera with 11 new interspecific cultivars. The contents and compositions of the individual sugars and acids were estimated by means of the HPLC method. Most of these varieties belong to the group with either medium or low content of malic acid, i.e. with a medium to high β ratio. This corroborates the similarity of interspecific varieties to those of V. vinifera. The cluster analysis identified the existence of two interesting groups of varieties: the first one involved the varieties Riesling, Nativa, Marlen, and Kofranka while the other group consisted of varieties Blaufränkisch, Blauer Portugieser, and Laurot. This observation also indicates similarity between Vitis vinifera L. varieties and interspecific cultivars and demonstrates that the contents of the primary metabolites (i.e. sugars and organic acids) are also comparable.

Although compost and biochar received high attention as growing media, little information is available on the potential of vineyard by-products for the production and use of composted solid digestate (CSD) and biochar (BC). In the present study, two experiments are reported on CSD and BC mixed with commercial peat (CP) for grapevine planting material production. Four doses (0, 10%, 20%, 40% vol.) of CSD and BC were assessed in the first and second experiment, respectively. CSD mixed at a dose of 10% recorded the highest values of shoot dry weight (SDW) and a fraction of total dry biomass allocated to shoot (FTS), both cropping bench-graft and bare-rooted vine. On the other hand, CSD mixed at a dose of 40% displayed the highest values of SDW and FTS, cropping two-year-old vine. BC used at a dose of 10% improved SDW, root dry weight, total dry weight, FTS, shoot diameter, and height on bare-rooted vine. The present study shows that CSD and BC, coming from the valorization of vineyard by-products, can be used in the production of innovative growing media suitable for nursery grapevine production. Further studies are needed to assess the combined applications of CSD and BC in the same growing media.

Royal palms [Roystonea regia (HBK.) O.F. Cook], coconut palms (Cocos nucifera L. `Malayan Dwarf'), queen palms [Syagrus romanzoffiana (Chamisso) Glassman], and pygmy date palms (Phoenix roebelenii O'Brien) were grown in a rhizotron to determine the patterns of root and shoot growth over a 2-year period. Roots and shoots of all four species of palms grew throughout the year, but both root and shoot growth rates were positively correlated with air and soil temperature for all but the pygmy date palms. Growth of primary roots in all four species was finite for these juvenile palms and lasted for only 5 weeks in royal palms, but ≈7 weeks in the other three species. Elongation of secondary roots lasted for only 9 weeks for coconut palms and less than half of that time for the other three species. Primary root growth rate varied from 16 mm·week-1 for coconut and pygmy date palms to 31 mm·week-1 for royal palms, while secondary root growth rates were close to 10 mm·week-1 for all species. About 25% of the total number of primary roots in these palms grew in contact with the rhizotron window, allowing the prediction of the total root number and length from the sample of roots visible in the rhizotron. Results indicated that there is no obvious season when palms should not be transplanted in southern Florida because of root inactivity.

Components of the carbon balance in the Sultana vine (Vitis vinifera L.) were examined with emphasis on the autumn, post-harvest, period. The net photosynthetic rate (PN) of leaves decreased from 0.30 to 0.03 mg CO2 m-2 s-1 from January to May (late summer to autumn). Two-thirds of this decline cocurred in the month before leaf fall. This decline in PN as senescence progressed was mainly due to increases in the 'residual', non-gas-phase, resistance to CO2 transfer. 14C-labelled assimilates were shown to move from the leaves to the perennial portions of the vine in both early and late autumn, and this 14C label reappeared in the shoot growth of the following spring. However, the level of carbohydrate reserves in shoots, roots or trunk in the autumn to spring period was not significantly affected by a severe (c. 60%) defoliation in early autumn. This level of defoliation, which the vine is able to tolerate without cumulative effects, occurs where Sultana vines are 'harvest-pruned' as part of the harvesting method of trellis drying.

Spraying Roomy Red grapes with benomyl 15 days before harvesting was effective than the control in decreasing decay percentage during 77 days cold storage(2-3°C). On the other hand, it was of meaningless effect on shattering and bleaching. Adding sodium bisulfite at a rate of 0.25 and 0.50 gms/skgs fruits just before storage minimized decay and shattering percentage. Simultaneously, no significant influence could be detected concerning its influence on bleaching. Sodium bisulffite reduced losses in grapes, weight as well as total soluble solids and acidity than the control and benomyl treatments. The studied parameters in grapes treated with benomyl and solium bisulfite as a pre and post-harvest treatment did not differ statistically than that of those treated with sodium bisulfite alone.

Rooted cuttings of grapevines (Vitis vinifera L. cv. Sultana; syn. Thompson Seedless) were grown under glasshouse conditions and supplied with dilute nutrient solution containing either 0 or 90 mM of added NaCl. Growth and photosynthetic response to salt treatment and subsequent recovery were followed over 80 days. Shoot growth and photosynthesis were reduced by salt treatment. At relatively low concentrations of leaf chloride (< c. 150 mM, on a tissue water basis), photosynthetic reduction was largely due to increased stomatal resistance. Internal disturbances were involved at higher leaf Cl- concentrations (> c. 150 mM) and included an apparent reduction in photochemical efficiency and a faster rate of photorespiration. Levels of fraction I protein, and specific activity of ribulose-1,5-bisphosphate carboxylase measured in vitro, were not reduced by salt treatment. Vines showed remarkable adaptation to salinity insofar as leaves maintained positive turgor despite leaf Cl- concentrations exceeding 300 mM, implying osmotic adjustment. Cessation of salt treatment led to an immediate decrease in leaf Cl-, a promotion of shoot growth and a progressive recovery in photosynthesis accompanied by a marked but not necessarily concurrent reduction in both stomatal and internal resistances. Leaves tolerated Cl- levels up to 200 mM (under glasshouse conditions) without sustaining permanent reduction in photosynthetic activity. New shoots formed subsequent to stress relief are not a prerequisite for Cl- retranslocation from mature leaves as decapitation at the time of stress relief did not prevent attenuation of leaf Cl- or recovery in photosynthesis.

The influence of the experimental herbicide safener BAS-145–138 on the toxicity of imazaquin and metsulfuron-methyl was investigated in two corn varieties in growth chamber bioassays. BAS-145–138 alone slightly decreased root and shoot growth but this decrease was not dependent on the safener concentration. The imbibition of seeds of both corn varieties with low levels of safener caused substantially less damage in the roots and shoots especially with the highest metsulfuron-methyl doses used in the study. In one variety, the safener significantly increased corn root tolerance to imazaquin and to metsulfuron-methyl 1.5- and 2.3-fold, respectively; the concentrations of both herbicides producing 50% inhibition were higher for safened seedlings. In the other variety, a high antidotal influence of BAS-145–138 imbibition on metsulfuron-methyl was observed in shoot growth.

<p>This study presents the results regarding the identification and discrimination of twenty seven possible clones of grapevine cultivar Korinthiaki staphis (<em>Vitis vinifera</em> L.), three biotypes of Korinthiaki lefki and the related cultivar Staphidampelo using the ampelographic description and the molecular method AFLP. The results from the statistical analysis showed that all the biotypes of cultivar Korinthiaki staphis show small distance and are grouped in the same cluster, depending on their origin, while Staphidampelo and Korinthiaki lefki are neither variants nor biotypes of the cultivar Korinthiaki staphis but different cultivars since they are very distant compared to the other biotypes and moreover, Korinthiaki lefki is in a separate cluster of the dendrogram. The ampelographic description in combination with the molecular method AFLP are effective for the study of the between and within genetic diversity of grapevine cultivars as well as for their identification and discrimination. The results of this study can constitute the base for the implementation of the clonal selection for grapevine cultivar Korinthiaki staphis and the seclusion of the desired clones.</p>
