Evaluating growth platforms and stress scenarios to assess the salt tolerance of wheat plants

Crops are routinely subjected to a combination of different abiotic stresses. Simplified platforms, stress scenarios and stress protocols are used to study salt tolerance under largely controlled and uniform conditions that are difficult to extrapolate to real arid and semiarid field conditions. To address the latter deficit, this work compares a realistic stress protocol (for salinity alone, drought alone and combined salinity plus drought stress) simulating a field environment in large containers to equivalent results from a more artificial pot environment. The work was based on two wheat cultivars known to differ in their salt tolerance (salt-sensitive Sakha 61 and salt-tolerant Sakha 93). Our results showed that previously established differences in the salt tolerances of the two wheat cultivars were no longer valid when the plants were exposed to a combined stress of salinity plus drought, regardless of the growth platform. Furthermore, in comparing a simulated field root-environment (containers) with pots, our results showed an interactive effect between the different treatments and platforms for both of the investigated cultivars. We conclude that a combined salinity + drought stress scenario and a reliable growth platform are of utmost importance in screening for salt tolerance of spring wheat. In future studies, increased emphasis should be placed on combining salinity with drought stress in well suited platforms to better mimic real field conditions where salinity is present.

Soil salinity is one of the most important abiotic stresses limiting plant growth and productivity. The breeding of salt-tolerant wheat cultivars has substantially relieved the adverse effects of salt stress. Complementing these cultivars with growth-promoting microbes has the potential to stimulate and further enhance their salt tolerance. In this study, two fungal isolates, Th4 and Th6, and one bacterial isolate, C7, were isolated. The phylogenetic analyses suggested that these isolates were closely related to Trichoderma yunnanense, Trichoderma afroharzianum, and Bacillus licheniformis, respectively. These isolates produced indole-3-acetic acid (IAA) under salt stress (200 mM). The abilities of these isolates to enhance salt tolerance were investigated by seed coatings on salt-sensitive and salt-tolerant wheat cultivars. Salt stress (S), cultivar (C), and microbial treatment (M) significantly affected water use efficiency. The interaction effect of M x S significantly correlated with all photosynthetic parameters investigated. Treatments with Trichoderma isolates enhanced net photosynthesis, water use efficiency and biomass production. Principal component analysis revealed that the influences of microbial isolates on the photosynthetic parameters of the different wheat cultivars differed substantially. This study illustrated that Trichoderma isolates enhance the growth of wheat under salt stress and demonstrated the potential of using these isolates as plant biostimulants.

Abstract This chapter discusses current progress and prospects of molecular breeding and strategies for developing better saline-tolerant sorghum (Sorghum bicolor) varieties. Most molecular breeding techniques for salt tolerance have been carried out in controlled environments where the plants were not exposed to any variation of the surrounding environment, producing reliable results. Due to the polygenic nature of salt tolerance, the identified quantitative trait loci (QTLs) could be false QTLs. Therefore, QTL validation is important in different plant populations and field conditions. Subsequently, marker validation is important before utilizing marker-assisted selection for screening salt-tolerant plants. Combining molecular breeding with conventional breeding can hasten the development of salt-tolerant sorghum varieties.

Field-based trials and genotype evaluation until yielding stage are two important steps in improving the salt tolerance of crop genotypes and identifying what parameters can be strong candidates for the better understanding of salt tolerance mechanisms in different genotypes. In this study, the salt tolerance of 18 bread wheat genotypes was evaluated under natural saline field conditions and at three saline irrigation levels (5.25, 8.35, and 11.12 dS m−1) extracted from wells. Multidimensional evaluation for salt tolerance of these genotypes was done using a set of agronomic and physio-biochemical attributes. Based on yield index under three salinity levels, the genotypes were classified into four groups ranging from salt-tolerant to salt-sensitive genotypes. The salt-tolerant genotypes exhibited values of total chlorophyll, gas exchange (net photosynthetic rate, transpiration rate, and stomatal conductance), water relation (relative water content and membrane stability index), nonenzymatic osmolytes (soluble sugar, free proline, and ascorbic acid), antioxidant enzyme activities (superoxide dismutase, catalase, and peroxidase), K+ content, and K+/Na+ ratio that were greater than those of salt-sensitive genotypes. Additionally, the salt-tolerant genotypes consistently exhibited good control of Na+ and Cl− levels and maintained lower contents of malondialdehyde and electrolyte leakage under high salinity level, compared with the salt-sensitive genotypes. Several physio-biochemical parameters showed highly positive associations with grain yield and its components, whereas negative association was observed in other parameters. Accordingly, these physio-biochemical parameters can be used as individual or complementary screening criteria for evaluating salt tolerance and improvement of bread wheat genotypes under natural saline field conditions.

AbstractSalt tolerant wheat cultivars may be used as genetic resource for wheat breeding to ensure yield stability in future. The study was aimed to select salt tolerant cultivar(s) to identify novel source of salt tolerance in local wheat germplasm. Initially, 40 local wheat cultivars were screened at 150 mM NaCl stress at seedling stage. Selected salt-tolerant (three; S-24, LU-26S and Pasban-90) and salt-sensitive (four; MH-97, Kohistan-97, Inqilab-91 and Iqbal-2000) wheat cultivars were further evaluated using growth, yield, biochemical and physiological attributes. Growth and yield of selected cultivars were reduced under salt stress due to decline in plant water status, limited uptake of macronutrients (N, P and K), reduced K+/Na+ ratio, photosynthetic pigments and quantum yield of PSII. Wheat plants tried to acclimate salt stress by osmotic adjustment (accumulation of total soluble sugars, proline and free amino acids). Degree of salinity tolerance in cvs. S-24 and LU-26S found to be associated with maintenance of K+/Na+ ratio, osmo-protectant and photosynthetic activity and can be used as donor for salt tolerance in wheat breeding program at least in Pakistan. These cultivars can be further characterized using molecular techniques to identify QTLs/genes for salt exclusion, osmo-protectant and photosynthetic activity for molecular breeding.

A cross was made between high yielding salt susceptible BINA variety (Binadhan-5) with salt tolerant rice landrace (Harkuch) to identify salt tolerant rice lines. Thirty six F3 rice lines of Binadhan-5 x Harkuch were tested for salinity tolerance at the seedling stage in hydroponic system using nutrient solution. In F3 population, six lines were found as salt tolerant and 10 lines were moderately tolerant based on phenotypic screening at the seedling stage. Twelve SSR markers were used for parental survey and among them three polymorphic SSR markers viz., OSR34, RM443 and RM169 were selected to evaluate 26 F3 rice lines for salt tolerance. With respect to marker OSR34, 15 lines were identified as salt tolerant, 9 lines were susceptible and 2 lines were heterozygous. While RM443 identified 3 tolerant, 14 susceptible and 9 heterozygous rice lines. Eight tolerant, 11 susceptible and 7 heterozygous lines were identified with the marker RM169. Seed of 42 wild accessions (Plant Introductions) of Lycopersicon pimpinellifolium Jusl., 11 cultigens (cultivated accessions) of L. esculentum Mill., and three control genotypes [LA716 (a salt-tolerant wild accession of L. pennellii Corr.), PI 174263 (a salt-tolerant cultigen), and UCT5 (a salt-sensitive breeding line)] were evaluated for germination in either 0 mm (control) or 100 mm synthetic sea salt (SSS, Na+/Ca2+ molar ratio equal to 5). Germination time increased in response to salt-stress in all genotypes, however, genotypic variation was observed. One accession of L. pimpinellifolium, LA1578, germinated as rapidly as LA716, and both germinated more rapidly than any other genotype under salt-stress. Ten accessions of L. pimpinellifolium germinated more rapidly than PI 174263 and 35 accessions germinated more rapidly than UCT5 under salt-stress. The results indicate a strong genetic potential for salt tolerance during germination within L. pimpinellifolium. Across genotypes, germination under salt-stress was positively correlated (r = 0.62, P < 0.01) with germination in the control treatment. The stability of germination response at diverse salt-stress levels was determined by evaluating germination of a subset of wild, cultivated accessions and the three control genotypes at 75, 150, and 200 mm SSS. Seeds that germinated rapidly at 75 mm also germinated rapidly at 150 mm salt. A strong correlation (r = 0.90, P < 0.01) existed between the speed of germination at these two salt-stress levels. At 200 mm salt, most accessions (76%) did not reach 50% germination by 38 days, demonstrating limited genetic potential within Lycopersicon for salt tolerance during germination at this high salinity.

Soil salinity is one of the main factors limiting cereal productivity in worldwide agriculture. Exploitation of natural variation in local barley germplasm is an effective approach to overcome yield losses. Three gene pools of North African Hordeum vulgare L. grown in Tunisia, Algeria and Egypt were evaluated at the reproductive stage under control and saline conditions. Assessment of stress tolerance was monitored using morphological, yield-related traits and phenological parameters of reproductive organs showing significant genetic variation. High heritability and positive relationships were found suggesting that some traits associated with salt tolerance could be used as selection criteria. The phenotypic correlations revealed that vegetative traits including shoot biomass, tiller number and leaf number along with yield-related traits such as spike number, one spike dry weight, grain number/plant and grain number/spike were highly positively correlated with grain yield under saline conditions. Hence, these traits can be used as reliable selection criteria to improve barley grain yield. Keeping a higher shoot biomass and longer heading and maturity periods as well as privileged filling ability might contribute to higher grain production in barley and thus could be potential target traits in barley crop breeding toward improvement of salinity tolerance. Multiple selection indices revealed that salt tolerance trait index provided a better discrimination of barley landraces allowing selection of highly salt-tolerant and highly productive genotypes under severe salinity level. Effective evaluation of salt tolerance requires an integration of selection indices to successfully identify and characterize salt tolerant lines required for valuable exploitation in the management of salt-affected areas.
