Aim. Cytokines and chemokines may be involved in the onset of oral ulcer in Behcet’s disease. The aim of our study is to assess the cytotoxic effects of proinflammatory cytokines and chemokines on reconstructed oral mucosal cell line (TR146) when treated with different concentrations of neutrophil elastase (NE). Objective. For this purpose, a culture of the oral mucosal model (OMM) prepared from a cell line derived from an oral squamous cell carcinoma of buccal mucosa (TR146) is treated with different concentrations of neutrophil elastase. The cultures were incubated for 4- and 24-hour intervals and designed as follows: culture + artificial saliva served as the negative control; culture + 0.01% SLS (sodium lauryl sulphate) served as the positive control; and culture + NE (10, 50, 100, and 200 nM) served as the treated group. Materials and Methods. We used sandwich ELISA technique to isolate IL-1β (interleukin 1β), IL-8, and TNF-α (tumor necrosis factor). Results. We found no significant level of IL-8 and TNF-α when treated with different concentrations of neutrophil elastase after 4- and 24-hour incubation. The IL-1β level was slightly higher when treated with 100 and 200 nM NE after 24 hours of incubation although a significantly high level was observed at 100 nM NE after 4 hours of incubation. Hence, we found an increase in the level of IL-1β when stimulating the reconstructed oral mucosal model (OMM) with different concentrations of NE. This is a preliminary in vitro study; however, further research is required to evaluate the cytotoxic effects of cytokines and chemokines released when treated with NE. Moreover, high concentrations of NE are recommended to stimulate the release of cytokines and chemokines against the OMM.

One of the nutritious plants to heal burns is the leaves of pohpohan (Pilea trinervia W.). This study aims to test the activity of pohpohan leaf extract (Pilea trinervia W.) on the healing of burns in rabbits. Preparation of gel is done on some concentration of pohpohan leaf extract, that is 0,5, 1, and 2%. After gel making, gel evaluation is done to know the quality of preparation, including organoleptic test, homogeneity, pH test, spreading test, sticky, and burn activity test. Burning activity test was performed on 3 male rabbits with 5 treatment sides on each rabbit, and induced with hot metal by Rafiky'sTool Pressure Test. For comparison use Bioplacenton®. Gels on the treatment of burns done 2 times a day. Parameters measured were burn diameter for 10 days. Data on burn activity test results were tested statistically using non-parametric Kruskal-Wallis test and further test of Mann Whitney. The results of gel evaluation showed that the gel containing pohpohan leaves extract concentration of 0.5, 1, and 2% fulfilled the homogeneity, pH, dispersion and stickiness requirements. The result of burn activity test showed that gel with concentration of 0,5, 1, and 2% extract pohpohan able to reduce burn burn diameter faster than control. AbstractTalas or taro leaf contains active compounds in the form of flavonoids and saponins that can accelerate the process of epidermal tissue re-epitalization and infiltration of inflammatory cells in the wound area. The purpose of this study was to compare the healing activities of the n-hexane fraction, methanol fraction, and taro leaf extract in New Zealand rabbits. The extraction method using maceration, the fractionation method used liquid-liquid partition with a separating funnel and the treatment method on test animals used was the application of the n-hexane fraction, methanol fraction, and taro leaf extract on open wounds of New Zealand rabbits with positive control and negative control. The data obtained is in the form of the value of open wound narrowing on the rabbit’s back. The result of the data were analyzed by ANOVA (Analysis Of Variant) followed by the LSD (Least Significant Different) test to see which treatment gave different effect in each group. The results of the research or testing for 14 days showed that the extract, n-hexane fraction, and methanol fraction had wound healing activity on the rabbit back with statistical data showing significant differences between treatment groups. The extract had the most optimal wound healing activity compared to other treatment groups. Keywords: talas leaf, extract, fraction, wound AbstrakDaun talas mengandung senyawa aktif berupa flavonoid dan saponin yang mampu mempercepat proses reepitalisasi jaringan epidermis dan infiltrasi sel-sel radang pada daerah luka. Ficus deltoidea (Mas Cotek) is one of the popular herbs that has been used traditionally to alleviate illnesses. The present study aimed to investigate the in vitro wound healing activity of  F. deltoidea leaf extract on skin cell. Cell proliferative and migration assay were done on the Human Skin Fibroblast cell (HSF 1184) which were treated with different concentrations of F. deltoidea leaf extract. The data of wound closure were collected at time intervals of 0, 6, 12 and 24 hours and analyzed using ImageJ™ software. MTT assay revealed that the F. deltoidea leaf extract could induce cell proliferation at a dose dependent manner. F. deltoidea leaf extract significantly accelerated the wound closure process in comparison to cells treated with ascorbic acid and untreated cells in scratch assay. Following 24 hours of incubation, cells treated with 50 mg/mL of F. deltoidea leaf extract showed remarkable proliferative and wound closure effect with 143.67% and 5.96%, respectively as compared to other concentrations. Collectively, these findings suggested that F. deltoidea leaf extract possesses wound healing potential and may be useful for the development of efficient wound healing drug.
