Microencapsulation of clove essential oil improves its antifungal activity against Penicillium digitatum in vitro and green mould on Navel oranges

Ten bacteria isolate (4 Bacillus subtilis, 2 Bacillus pumilus, 2 Bacillus cereus, 1 Bacillus megaterium, and 1 Agrobacterium radiobacter) were tested in vitro for antagonistic properties against Penicillium digitatum, the causal agent of citrus green mould. The effect of these bacteria was also observed on mycelial growth, spore germination, and spore production of the pathogenic fungus in broth culture. Extracellular enzyme activities of the bacteria were determined. According to the results of in vitro antagonistic tests and enzymes activities, the most promising bacteria were Bacillus subtilis and Agrobacterium radiobacter. These bacteria were tested for disease suppression on lemon fruits. In addition, these bacterial isolates also showed remarkable antifungal activity against the pathogen on lemon fruits. The results of this study showed that Bacillus subtilis and Agrobacterium radiobacter showed remarkable antifungal activity against the pathogen. Chitinase and glucanase enzyme activity of all the tested bacteria was positive. Protease enzyme activity was positive in all tested bacteria with the exception of Agrobacterium radiobacter. In addition, all bacteria inhibited mycelial growth and spore germination (except Agrobacterium radiobacter) of the fungus. Bacillus subtilis, Bacillus cereus, and Agrobacterium radiobacter inhibited spore production in broth culture. Bacillus subtilis and Agrobacterium radiobacter were tested on lemon fruits significantly reduced disease severity. Consequently, these isolates can be used as new biocontrol agents in controlling the post-harvest decay of citrus fruits caused by Penicillium digitatum.

Abstract The objective of the present paper was the assessment of the chemical composition of the essential oils from four Asteraceae species with a considerable food, medicinal, and agricultural value, collected in Egypt, together with their in vitro inhibitory activity against molds and yeasts. The essential oil of Launaea cornuta flowers was also evaluated for the first time, but because of its very low yield (<0.01%), no antifungal test was performed.

Background. Echinops kebericho is an endemic medicinal plant in Ethiopia widely used in the treatment of infectious and noninfectious diseases. Essential oils are known for their antibacterial, antifungal, antiviral, insecticidal, and antioxidant properties. This study evaluated the antifungal activity of essential oil from E. kebericho against four common pathogenic fungi and two standard strains. Methods. The essential oil was obtained by hydrodistillation. The antifungal screening was done by agar well diffusion method. Minimal inhibitory concentrations (MICs) were determined by broth microdilution. Minimal fungicidal concentrations (MFCs) were determined by subculturing fungal strains with no visible growth onto a Sabouraud dextrose agar (SDA) plate. Results. Candida albicans and Cryptococcus neoformans were highly sensitive while Aspergillus flavus did not show sensitivity up to 1 mg/ml of essential oil; MICs ranged from 0.083 mg/ml to 0.208 mg/ml. Concentration and fungal species showed significant dose-dependent associations ( p < 0.0001 ) with antifungal activity. The MICs of essential oil were comparable to those of the standard drug (fluconazole) against C. glabrata and C. krusei. The lowest MFC of the essential oil was observed against Candida parapsilosis (0.145 mg/ml) while the highest MFC was against Candida krusei (0.667 mg/ml). Conclusion. Echinops kebericho essential oil showed noteworthy antifungal activity against Cryptococcus neoformans, Candida albicans, and Candida glabrata and could be a potential candidate for further antifungal drug development.

The increase in dermatophytoses and the fact that some patients do not respond well to therapy make it necessary to find new antifungal agents. As part of our ongoing studies on medicinal plants from Iran, we studied antidermatophytic activities of Ferula latisecta essential oil, which had shown considerable antifungal activity in preliminary antimicrobial screening. Antifungal activity was evaluated by determination of MIC values using the agar dilution method on type strains of Candida albicans and dermatophytes. The composition of the oil was characterized by GC and GC/MS analyses. The essential oil was rich in polysulfides (75.2%) and exhibited good activity against Trichophyton rubrum and T. verrucosom for about three weeks, with a MIC value 96 μg/mL. The oil showed antifungal activity, especially against dermatophytes, and the activity is probably related to the sulfur-containing components of the oil. This study has identified that the polysulfides-rich essential oil of Ferula latisecta fruits has activity against a range of human pathogenic dermatophytes, justifying future clinical trials to validate its use as a therapeutic alternative for dermatophytosis.

In this study, Carum carvi L. essential oil (CEO) and Origanum majorana L. essential oil (MEO) was steam-distillated under reduced pressure. We henceforth obtained three fractions for each essential oil: CF1, CF2, CF3, MF1, MF2, and MF3. Then, these fractions were characterized using the gas chromatography–mass spectrometry (GC-MS) technique. The results indicated that some fractions were rich in oxygenated compounds (i.e., CF2, CF3, MF2, and MF3) with concentrations ranging from 79.21% to 98.56%. Therefore, the influence of the chemical composition of the essential oils on their antifungal activity was studied. For this purpose, three food spoilage fungi were isolated, identified, and inoculated in vitro, in order to measure the antifungal activity of CEO, MEO, and their fractions. The results showed that stronger fungi growth inhibitions (FGI) (above 95%) were found in fractions with higher percentages of oxygenated compounds, especially with (−)-carvone and terpin-4-ol as the major components. Firstly, this work reveals that the free-terpenes hydrocarbons fractions obtained from MEO present higher antifungal activity than the raw essential oil against two families of fungi. Then, it suggests that the isolation of (−)-carvone (97.15 ± 5.97%) from CEO via vacuum distillation can be employed successfully to improve antifungal activity by killing fungi (FGI = 100%). This study highlights that separation under reduced pressure is a simple green method to obtain fractions or to isolate compounds with higher biological activity useful for pharmaceutical products or natural additives in formulations.

The essential oil obtained by the hydro-distillation of the leaves of Mentha arvensis Linnaeus was evaluated for its antifungal activity against the causal agent of the Alternaria blight of tomatoes, i.e., Alternaria alternata (Fries) Keissler. The antifungal activity of the mentha essential oil was assessed both in vitro and in vivo. The chemical composition of the mentha oil was also identified by GCMS analysis. The in vitro test revealed that the maximum inhibition in the mycelial growth (93.6%) and conidia germination (90.6%) was at the highest concentration (40 µL/mL), furthermore, it was found that the inhibition of the mycelial growth and conidia germination was dose dependent. The in vivo test proved that the application of the mentha essential oil (40 µL/mL) significantly increased the plant height (84.6%), fresh weight (81.5%) and dry weight (80.0%) when compared to the untreated tomato plants. The disease incidence was 3.5 in the untreated plants, while it was 0.93 for the mentha essential oil treated plants and was 0.08 in the carbendazim treated plants. The GC-MS analysis of the mentha essential oil identified 18 compounds in total, among which the percentage of menthol was the highest (69.2%). The mentha essential oil was successful in managing the Alternaria leaf blight in the tomato plants. Therefore, it can be explored further for the development of a natural fungicide.
