Abstract Western Sandpipers (Calidris mauri) have been previously shown to undergo seasonal changes in the fatty acid composition of their fat stores, even though they do not show the marked seasonal variation in diet common to many migratory passerines. We investigated the effect of dietary fatty acid composition on the fatty acid composition of adipose tissue in captive Western Sandpipers by feeding birds experimental diets with different fatty acid composition. In addition, we determined the effect of total percentage of fat content of the diet (5 vs. 10%) on fatty acid composition of depot fat. Birds maintained normal body mass (24–27 g) throughout all experimental treatments. Most adipose fatty acids were sensitive to dietary manipulation to some extent. Changes in fatty acid composition of the diet had the largest effect on adipose tissue composition for the essential polyunsaturated fatty acid linoleate (18:2), whereas it had the least effect for the monounsaturated fatty acid oleate (18:1). The saturated fatty acid palmitate (16:0) demonstrated an intermediate capacity to alter fatty acid composition of adipose tissue. Total amount of fat in the diet did not influence the effect of diet on fatty acid deposition. Results of dietary manipulations in this study suggest that diet does explain some of the variation in fatty acid composition observed during migration in Western Sandpipers, but that certain fatty acids can be modulated independently of diet (probably through de novo synthesis, postabsorption modification, or both).

SummaryPlatelet aggregation and its relation to fatty acid composition of platelets, plasma and adipose tissue was determined in 196 randomly selected, free-living, 40-49-year-old men in two regions of Finland (east and southwest) with a nearly twofold difference in the IHD rate.There were no significant east-southwest differences in platelet aggregation induced with ADP, thrombin or epinephrine. ADP-induced platelet secondary aggregation showed significant negative associations with all C20-C22 ω3-fatty acids in platelets (r = -0.26 - -0.40) and with the platelet 20: 5ω3/20: 4ω 6 and ω3/ ω6 ratios, but significant positive correlations with the contents of 18:2 in adipose tissue (r = 0.20) and plasma triglycerides (TG) (r = 0.29). Epinephrine-induced aggregation correlated negatively with 20: 5ω 3 in plasma cholesteryl esters (CE) (r = -0.23) and TG (r = -0.29), and positively with the total percentage of saturated fatty acids in platelets (r = 0.33), but had no significant correlations with any of the ω6-fatty acids. Thrombin-induced aggregation correlated negatively with the ω3/6ω ratio in adipose tissue (r = -0.25) and the 20: 3ω6/20: 4ω 6 ratio in plasma CE (r = -0.27) and free fatty acids (FFA) (r = -0.23), and positively with adipose tissue 18:2 (r = 0.23) and 20:4ω6 (r = 0.22) in plasma phospholipids (PL).The percentages of prostanoid precursors in platelet lipids, i. e. 20: 3ω 6, 20: 4ω 6 and 20 :5ω 3, correlated best with the same fatty acids in plasma CE (r = 0.32 - 0.77) and PL (r = 0.28 - 0.74). Platelet 20: 5ω 3 had highly significant negative correlations with the percentage of 18:2 in adipose tissue and all plasma lipid fractions (r = -0.35 - -0.44).These results suggest that, among a free-living population, relatively small changes in the fatty acid composition of plasma and platelets may be reflected in significant differences in platelet aggregation, and that an increase in linoleate-rich vegetable fat in the diet may not affect platelet function favourably unless it is accompanied by an adequate supply of ω3 fatty acids.

The fatty acid composition of newly emerged Ips paraconfusus Lanier shows no sexual dimorphism and is approximately as follows: C14:0, 0.5%; C16:0, 23.0%; C16:1, 6%; C18:0, 3%; C18:1, 55%; C18:2, 9%; C18:3, 2%. Both sexes, but particularly the female, use up fatty acids, particularly the monounsaturated acids, during reproduction. Isotope from 1-14C-acetate injected into newly emerged females appeared in all saturated and monounsaturated fatty acids within 30 min. There was evidence of de novo synthesis of C14:0 and C16:0, chain elongation of C16:0 to C18:0, and desaturation of C16:0 and C18:0 to yield C16:1 and C18:1 respectively.

The aim of this study was to provide information on the fatty acid profile of different adipose depots - subcutaneous (upper and inner backfat layers) and intramuscular (m. Longissimus dorsi) in East Balkan pigs. The animals were reared in free-range conditions and slaughtered at an average live weight of 107?1.65kg. The results of the study showed that the various adipose tissues in pigs have different lipid metabolism and hence differ in their fatty acid composition. Intramuscular fat had significantly higher content of the saturated C16:0 and C18:0 (P<0.001), as well as the C16:1 (P<0.001) than the subcutaneous fat. In regards to the content of the polyunsaturated fatty acids, the latter displayed considerably higher content of both C18:2 and C18:3 (P<0.001) in comparison to the intramuscular fat in m. Longissimus dorsi. The differences between the subcutaneous and intramuscular adipose tissue in the individual fatty acids determined the similar trend of change in the total content of saturated and polyunsaturated fatty acids. Significant differences between the backfat layers were detected for C16:1, C18:0 and C18:3 (P<0.001). Stearic acid (C18:0) displayed higher content of the inner, while both C16:1 and C18:3 had higher proportion in the outer backfat layer in the East Balkan pigs. Except for C20:2, the long chain polyunsaturated n-6 and n-3 fatty acids had significantly higher proportions in the intramuscular fat, however no differences were determined between the two backfat layers.

Compared to meat from other animals lamb contains high levels of saturated fat, particularly stearic acid which comprises 18% of the total fatty acids (Enser et al, 1996). This stearic acid can be desaturated in the tissue by stearoyl coenzyme A desaturase (SCD) to produce oleic acid. In sheep SCD is produced from a single gene and the levels of SCD mRNA in the tissue correlate well with oleic acid (Ward et al, 1998, Barber et al, 2000) suggesting that an upregulation of SCD activity may increase the relative proportions of unsaturated and saturated fatty acids and so significantly improve the nutritional quality of sheep meat. Our recent studies have shown that insulin increases SCD mRNA levels and monounsaturated fatty acid synthesis in cultured ovine adipose tissue explants (Daniel et al, 2001). The present study was designed to investigate whether feeding a diet believed to manipulate SCD mRNA concentrations would significantly alter the fatty acid composition of lamb.

The benefits of long chain polyunsaturated fatty acids (PUFA) to human health, especially those of the n-3 series are now widely recognised. In a previous experiment (Wachira et al. 1998) supplementing diets with whole linseed or fish oil increased n-3 fatty acid levels in lamb muscle. To raise these further the whole linseed can be treated with formaldehyde to increase protection in the rumen. Dietary antioxidants such as vitamin E can control lipid oxidation but information on their effects on lamb performance and fatty acid composition is limited. The current experiments investigated the effects of different dietary PUFA sources and vitamin E levels on growth and fatty acid composition in two sheep breeds. Detailed results of the effects of vitamin E are presented in the accompanying abstract by Enser et al.

The fatty acid composition of cell membrane phospholipids does not remain constant after de novo biosynthesis, but undergoes continual remodelling. One of the major routes for remodelling probably includes the deacylation-reacylation steps of the Lands Pathway. This has been shown to be important for the incorporation of long chain, polyunsaturated fatty acids into phospholipids by liver and brain. An understanding of the mechanisms involved in these processes in platelets is especially important in light of the large stores of arachidonic acid (AA) in platelet phospholipids and the role of AA in hemostasis and thrombosis. Previous results from this laboratory have shown that the turnover of radioactive AA, 8,11,14-eicosatrienoic and 5,8,11,14,17-eicosapentaenoic acids in the phospholipids of resting platelets is more rapid than the turnover of radioactive C16 and C18 saturated and unsaturated fatty acids. However, little is known about how fatty acids, especially AA and its homologues, are incorporated into platelet phospholipids during de novo biosynthesis or how they are exchanged during remodelling.At least three enzymes are involved in the deacylation- reacylation of phospholipids: phospholipase A2; acyl CoA synthetase; and acyl CoA transferase. We have studied acyl CoA transferase and have found considerable activity in human platelet membranes. Experiments are in progress to determine the substrate specificity and other properties of this enzyme.

The effect of safflower oil supplement (200 mL/day) on voluntary feed intake and fatty acid composition of plasma, blood platelets, subcutaneous adipose tissue, and M. longissimus dorsi muscle was studied in Brahman steers consuming a low quality hay. The initial mean liveweight (LW) of the steers was 197 kg. Mean LW gain over 50 days was 20 kg for all steers, but voluntary feed intake (5.63 kg/day) was reduced by 1.02 kg/day with safflower oil. Supplementation increased the plasma concentrations of unsaturated fatty acids, indicating a less complete hydrogenation of dietary safflower oil than of hay by rumen microorganisms. Safflower oil altered the fatty acid (FA) profiles of platelet and muscle phospholipids. with increases in the proportions of monounsaturated acids (MFA) and polyunsaturated fatty acids (PFA) and associated decreases in the proportions of saturated fatty acids (SFA). The neutral lipids of adipose tissue consisted of 60% SFA and 38% MFA in steers fed hay alone, compared with 53% SFA and 45% MFA in supplemented steers. Safflower oil had no effect on the total fat content of muscle; the concentration of PFA was not significantly altered, but SFA decreased 17% in parallel with a 23% increase in MFA. The results support proposals that dietary oil supplements may be used to reduce forage intake during dry season or drought, to conserve body protein and to modify the FA composition of meat phospholipids which may, in turn, influence meat quality of ruminants consuming low quality roughage diets.

The effects of diet on the composition and properties of adipose tissue in relation to lymph nodes were studied in adult guinea-pigs. The proportions of monoenoic triacylglycerol fatty acids were constant in all sites in adipose tissue of similarly fed guinea-pigs, but were substantially greater in samples from guinea-pigs fed on suet-enriched chow. Triacylglycerols in adipose tissue from near nodes contained significantly fewer saturated fatty acids, and significantly more 18:2n−6 and 18:3n−3 than those in samples from sites remote from nodes within the same depot. Depots that interact most strongly with lymphoid cellsin vitrohad the largest and most consistent within-depot differences. The gradients of triacylgiycerol fatty acid composition with distance from lymph nodes in two small intermuscular depots were similar in guinea-pigs fed on plain or suet-enriched chow. These findings are consistent with the hypothesis that adipose tissue around lymph nodes is specialized for local interactions with the lymphoid cells therein, and help to explain the variability of serial or duplicate measurements of adipose tissue composition. When cultured alone, lipopolysaccharide-stimulated lymph node lymphoid cells from suet-fed guinea-pigs incorporated as much labelled thymidine as the controls. Adipose tissue explants from suet-fed guinea-pigs inhibited lymphocyte proliferation much less than those of the controls, although the site-specific differences were similar. The pattern of site-specific differences in glycerol released from explants incubated alone was generally similar for both dietary groups, but except in the popliteal depot, the increases following co-culturing with lymphoid cells were smaller for samples from suet-fed guinea-pigs. These experiments show that minor changes in the fatty acid composition of the diet can substantially alter the interactions between adipose tissue and lymphoid cells.

1. Samples of subcutaneous (inguinal) and perinephric adipose tissue were obtained, at slaughter, from each of twenty male calves. Three were neonatal animals, three were 3 days old and two were fed on reconstituted milk to appetite until they weighed 100 kg. The other twelve calves were given milk until they reached 50 kg live weight; concentrates were then included in the diet until, at 60 kg live weight, six calves were slaughtered. The remaining six calves were raised to 100 kg on concentrates alone. The weight of the empty reticulo-rumen of each slaughtered calf was recorded.2. The component fatty acids of the adipose tissue triglycerides of the neonatal and 3-day-old calves were very similar; about 80% consisted of oleic acid (18:1) and palmitic acid (16:0) and the remainder comprised stearic acid (18:0), palmitoleic acid (16:1) and myristic acid (14:0), together with very small amounts of other acids which, in the glycerides of the 3-day-old calves, included some evidently of colostral origin. The perinephric glycerides of both these groups of calves were somewhat more unsaturated than were those of subcutaneous adipose tissue.3. The continued consumption of milk by the calves slaughtered at 60 kg live weight was reflected in the presence of enhanced proportions of 14:0, 18:2, 17:0 and 17:1 in the depot triglycerides and, in addition, very small amounts of branched-chain acids and trans 18:1 were detected. A similar fatty acid pattern was observed in the triglycerides of the calves which were given milk only until they were 100 kg live weight. In all these calves only limited growth of the rumen took place.4. By contrast, the calves which were raised on solid feed from 60 kg to 100 kg and in which rumen development had taken place had depot triglycerides whose fatty acid composition resembled that found in adult animals. Increased proportions of stearic acid accompanied by relatively large amounts of trans 18:1 were present, evidently as a result of the assimilation of the products of bacterial modification of dietary fatty acids in the rumen.5. Regardless of the age of the calves and the over-all fatty acid composition of their tissue triglycerides, the intramolecular disposition of the fatty acids was similar in that saturated components were present esterified mainly in positions 1 and 3, and unsaturated acids for the most part in position 2; the only major exception to this distribution pattern was in respect of trans 18:1 which, when present, was preferentially esterified to the primary alcoholic groups of the glycerol moiety as if it were a saturated acid.
