Studies of Blood Lipid Fractions of Quiescent Rheumatic Fever Patients and Their Siblings

Abstract Three phospholipid fractions have been determined in plasmas of quiescent rheumatic children and their siblings of similar ages. The method for determining blood lecithin gave reproducible results, and the lipid fraction appeared indicative of difference in levels of blood lipids between rheumatic and nonrheumatic children. In this series of lecithin determinations only one rheumatic subject had a high normal level and only one sibling (presumably nonrheumatic) had a low level. The difference in mean nonfasting lecithin plasma levels between rheumatic subjects and siblings was 0.36 mMol./L. Similarly, the lecithin level of each of the nonrheumatic siblings in this series averaged 0.41 mMol./L. higher than that of the corresponding quiescent rheumatic child. In both of these differences P <0.05.

Background: Previous studies showed an anti-atherosclerotic effect of PADMA 28, an herbal formula based on Tibetan medicine. As the mechanisms of action are not fully understood, we investigated whether PADMA 28 may lower blood lipids and lipid oxidisability, and affect early endothelial dysfunction. Patients and methods: Sixty otherwise healthy subjects with total cholesterol ≥5.2 mmol/l and < 8.0 mmol/l were randomly assigned to placebo or PADMA 28, 3 x 2 capsules daily, for 4 weeks (double-blind). Blood lipids (total, LDL-, and HDL-cholesterol, triglycerides, Apo-lipoprotein A1 and B) and ex vivo lipid oxidisability were measured before and after treatment. In a subset of 24 subjects, endothelial function was assessed using venous occlusion plethysmography with intraarterial infusion of acetylcholine. Isolated LDL and plasma both untreated and pre-treated with PADMA 28 extract were oxidised by the radical generator AAPH. Conjugated diene formation was measured at 245 nm. Results: Blood lipids did not change during the study in both groups. In contrast to previous reports in mild hypercholesterolaemia, no endothelial dysfunction was seen and, consequently, was not influenced by therapy. Ex vivo blood lipid oxidisability was significantly reduced with PADMA 28 (area under curve: 5.29 ± 1.62 to 4.99 ± 1.46, p = 0.01), and remained unchanged in the placebo group (5.33 ± 1.88 to 5.18 ± 1.78, p > 0.1). This effect persisted one week after cessation of medication. In vitro experiments confirmed the prevention of lipid peroxidation in the presence of PADMA 28 extracts. Persistent protection was also seen for LDL isolated from PADMA 28-pretreated blood after being subjected to rigorous purification. Conclusions: This study suggests that the inhibition of blood lipid oxidisability by PADMA 28 may play a role in its anti-atherosclerotic effect.

SummaryWe have studied 155 subjects, 48 normals, 36 diabetics without complications, 44 with complications and 27 patients with macroangiopathy. β-Thromboglobulin (β-TG) and platelet factor 4 (PF4) are elevated in the patients groups. There is no correlation between the plasma levels of β-TG and the stages of either retinopathy or macroangiopathy or nephropathy. The difference is more marked between normals and diabetics with neuropathy (p = 0.026). The aggregation response to ADP and platelet activating factor (PAF) is enhanced at lower stimulator concentration. Using the β-TG, PF4 and aggregation values the discriminant analysis allows a distinction of several subgroups especially with nephropathy and neuropathy (Table 6).

AbstractThe role of high-mobility group box-1 (HMGB1) in outcome prediction in sepsis is controversial. Furthermore, its association with necroptosis, a programmed cell necrosis mechanism, is still unclear. The purpose of this study is to identify the association between the plasma levels of HMGB1 and the severity and clinical outcomes of sepsis, and to examine the correlation between HMGB1 and key executors of necroptosis including receptor-interacting kinase 3 (RIPK3) and mixed lineage kinase domain-like- (MLKL) proteins. Plasma HMGB1, RIPK3, and MLKL levels were measured with the enzyme-linked immunosorbent assay from the derivation cohort of 188 prospectively enrolled, critically-ill patients between April 2014 and December 2016, and from the validation cohort of 77 patients with sepsis between January 2017 and January 2019. In the derivation cohort, the plasma HMGB1 levels of the control (n = 46, 24.5%), sepsis (n = 58, 30.9%), and septic shock (n = 84, 44.7%) groups were significantly increased (P < 0.001). A difference in mortality between high (≥ 5.9 ng/mL) and low (< 5.9 ng/mL) HMGB1 levels was observed up to 90 days (Log-rank test, P = 0.009). There were positive linear correlations of plasma HMGB1 with RIPK3 (R2 = 0.61, P < 0.001) and MLKL (R2 = 0.7890, P < 0.001). The difference in mortality and correlation of HMGB1 levels with RIPK3 and MLKL were confirmed in the validation cohort. Plasma levels of HMGB1 were associated with the severity and mortality attributed to sepsis. They were correlated with RIPK3 and MLKL, thus suggesting an association of HMGB1 with necroptosis.

Microalgae are at the start of the food chain, and many are known producers of a significant amount of lipids with essential fatty acids. However, the bioactivity of microalgal lipids for anti-inflammatory and antithrombotic activities have rarely been investigated. Therefore, for a sustainable source of the above bioactive lipids, the present study was undertaken. The total lipids of microalga Chlorococcum sp., isolated from the Irish coast, were fractionated into neutral-, glyco-, and phospho-lipids, and were tested in vitro for their anti-inflammatory and antithrombotic activities. All tested lipid fractions showed strong anti-platelet-activating factor (PAF) and antithrombin activities in human platelets (half maximal inhibitory concentration (IC50) values ranging ~25–200 μg of lipid) with the highest activities in glyco- and phospho-lipid fractions. The structural analysis of the bioactive lipid fraction-2 revealed the presence of specific sulfoquinovosyl diacylglycerols (SQDG) bioactive molecules and the HexCer-t36:2 (t18:1/18:1 and 18:2/18:0) cerebrosides with a phytosphingosine (4-hydrosphinganine) base, while fraction-3 contained bioactive phosphatidylcholine (PC) and phosphatidylethanolamine (PE) molecules. These novel bioactive lipids of Chlorococcum sp. with putative health benefits may indicate that marine microalgae can be a sustainable alternative source for bioactive lipids production for food supplements and nutraceutical applications. However, further studies are required towards the commercial technology pathways development and biosafety analysis for the use of the microalga.

Babesia divergens was cultivated in RPMI 1640 (25 mM HEPES) supplemented with 10% human serum (RPMI-10% HS) with a high percentage of parasitized erythrocytes (PPE) ([ges ]40%). Standardization of in vitro tests, purification of exoantigens, biochemical studies and the safety of the culture handler motivated the development of a serum-free defined medium. Removal of serum greatly reduced the PPE but, after a period of adaptation, the culture was continuous and the parasite was able to develop a 3% routine PPE. Addition of vitamins or reduced glutathione in basal medium (RPMI) did not improve the PPE. The supplementation of basal medium with lipidic carrier (Albumax I or bovine serum albumin–Cohn's fraction V) promoted the growth of B. divergens with high PPE (>30%) close to those obtained in RPMI–10% HS. Neither protein nor lipid fractions alone were able to restore the growth of B. divergens. Nevertheless, the whole lipid fraction from serum or Albumax I added to delipidated albumin partially restored the growth (7% PPE), indicating that the presentation of specific lipids by a carrier is crucial for the parasite. All the data indicate that Albumax I can replace human serum offering the advantages of safety, standardization for chemosensitivity tests, and exoantigen purification.

In reply to the question raised by Dr. Morgan, it is now well established that urea plays an important part in the concentration of urine. Starting at a very low level of urea excretion, the urinary concentration maxima (as obtained during prolonged fluid deprivation, or following sustained vasopressin stimulation) is less than normal. When the rate of urea excretion is increased to the normal level, the maximal urinary concentration ability increases, but when the rate of urea excreted increases markedly, the urinary concentration maxima once again decreases.

Individuals’ perceptions of their knowledge can have an important role in shaping their cognition and influencing their behaviour. However, there has been a scarcity of studies in biology on how perceived knowledge relates to actual knowledge. The focus of this article is on quantitative results analysing and interpreting student teachers’ perceived knowledge of biological content in relation to their actual animal and species name knowledge linked to the ecosystem in which they live. K-means cluster analysis and ANOVA were used. The results show a high- and low-level perceived knowledge cluster group among the participants. They further indicate that the difference in actual animal and species name knowledge betweenthese cluster groups remained the same during the five years of the study. The student teachers with a higher level of perceived knowledge tended to have better actual animal and species name knowledge than those in the low-level group. The actual animal name knowledge in these cluster groups was similar with regard to the local Finnish ecosystems but differed concerning the exotic species by year. The year that the participants enrolled in the study programme had an impact on their actual animal and species name knowledge. Strategies for coping with work-related demands and maintaining engagement in one’s career would be important additions to the teacher education curriculum.

Abstract. A regional-scale dust model is used to simulate Saharan dust emissions and atmospheric distributions in the years 2007 and 2008. The model results are compared to dust source activation events compiled from infrared dust index imagery from the geostationary Meteosat Second Generation (MSG) satellite. The observed morning maximum in dust source activation frequencies indicates that the breakdown of nocturnal low-level jets is responsible for a considerable number of dust source activation events in the Sahara. The comparison shows that the time of the day of the onset of dust emission is delayed in the model compared to the observations. Also, the simulated number of dust emission events associated with nocturnal low level jets in mountainous regions is underestimated in the model. The MSG dust index observations indicate a strong increase in dust source activation frequencies in the year 2008 compared to 2007, the difference between the two years is less pronounced in the model. The quantitative comparison of simulated dust optical thicknesses with observations at stations of the sunphotometer network AERONET shows, however, good agreement for both years, indicating that the number of observed dust activation events is only of limited use for estimating actual dust emission fluxes in the Sahara.

AbstractObjective:  Postpartum obesity leads to long-term maternal obesity and promotes drastic health complications. Low glycemic index diet is suggested to have a beneficial impact on blood lipid levels. Therefore, we conducted a study to explore the effect of low glycemic diet on blood lipid profile in obese postpartum women.Methods:  In a randomised controlled trial, 38 obese postpartum women in intervention and 36 obese post-partum women in control group were analysed. Subjects in the intervention group were assigned low glycemic index diet and to follow this protocol for 12 weeks and the control group was advised to continue their routine diet.Results:  Low glycemic diet had a positive impact on low density lipoprotein (LDL), triglyceride and highdensity lipoprotein (HDL) concentration (p < 0.05). A strong positive correlation was observed between the glycemic index and LDL (mg/dl), (r = 0.57; p = 0.02) and between glycemic index and triglyceride (mg/dl), (r = 0.51; p = 0.01) in control and intervention group. A negative correlation was identified between glycemic index and HDL (mg/dl), (r = -0.45; p = 0.01).Conclusion:  Study concluded that low glycemic index diet reduced low density lipoprotein and triglyceride level and increased HDL level in blood; further more a significant association was found between glycemic index and blood lipids profile.
