Histopathological Changes of Organs (Lungs, Liver, Kidney, and Brain) After Using Two Types of AgiCoat and Acticoat Nanosilver Dressings on Deep Second-Degree Burn in Rat

Abstract Prevention of infections is a very important issue in treating the burn wounds. The nanosilver dressings have many promising advantages, but absorption of silver ions and its adverse effects to the body were always a question. The aim of this study was to compare Silver serum levels and acute toxic effects of nanosilver on histopathology of organs (lungs, liver, kidney, spleen, and brain) in two types of AgiCoat and Acticoat (nanosilver) dressings on second-degree deep burn in rat. This is an experimental study conducted in our animal laboratory. We divided 24 Sprague–Dawley male rats weighing 300 to 350 randomly into two groups. After anesthesia, a second deep-degree burn was made over dorsal skins of rats by standard method. For group A, Agicoat and, for group B, Acticoat dressings were used. The dressings were changed every 3 days with AgiCoat and Acticoat, respectively. After 14 days, we got blood samples and tissue samples taken from heart, liver, kidneys, spleen, lungs, and brain and a sample from dorsal skin of the rat for histopathological examinations. The results showed that the levels of serum silver in both groups were significantly higher than the standard level (1.22 part per million (PM); AgiCoat, P = .017; Acticoat, P = .000), but there was no significant difference between the groups (P = .551). Examination of the relationship between the level of serum silver and histopathological changes in liver showed that hepatotoxicity of AgiCoat was higher compared with Acticoat and the difference was significant (P = .002). There were no pathological changes in brain, kidneys, spleen, heart, and lungs. Wound healing was faster in Acticoat group. The nanosilver dressings can cause toxicity in liver but not in kidney, brain, spleen, heart, and lungs. Liver pathology and hepatotoxicity were more prominent in AgiCoat group. Wound healing was faster in Acticoat group.

Abstract Introduction Burn is one of the most severe traumas that causes coagulative destruction of the skin. The use of various products that accelerate wound healing in patients with burn may affect the patient’s survival and reduce the complications that may be seen. In the present study we aimed effects of subcutaneous ozone injection on second degree burn wound. Methods A total of 72 Sprague-Dawley male rats included in the study were divided randomly into three groups (control group (CG), silver sulfadiazine group (SG), ozone group (OG)) and each group was divided randomly two subgroups (as sacrificed on d7 and on d14).A deep second degree scald burns were created on the lower back. In CG subcutaneous 0.9% serum saline was injected daily into the burn area. In SG, burns were dressed with silver sulfadiazine daily and in OG subcutaneous ozone was injected daily into the burn area. Tissue hydroxyproline level measurement and histopathological evaluation were done. Results When the groups were compared in terms of weight change, no significant difference was found on the 7th and 14th days. In the evaluation made in terms of tissue hydroxyproline, tissue hydroxyproline level in OG was found to be significantly higher on both the 7th and 14th days (p < 0.001). In histopathological evaluations, it was determined that wound healing in OG was significantly higher than in the other groups. Conclusions According to the results, subcutaneous ozone therapy is more effective than silver sulphadiazine in the healing process of second-degree burn wounds and it can be safely used in the treatment of burn wounds.

ackground: This study aimed to compare sheep burnt wool and human amniotic membrane (AM) on second-degree burn wound healing in rats. Materials and Methods: Seventy-two adult male rats of Sprague Dawley underwent general anesthesia, and a deep second-degree burn was created on their skin by a hot iron plate. Afterward, human AM, silver sulfadiazine ointment (SSD), and sheep burned wool were used on wound area for burn treatment. On days 7, 14, and 21 of the experiment, the rats were sacrificed, and histopathological assessments were done. Results: Human AM, in comparison with other groups, significantly (P<0.05) showed better improvement in all pathologic variables. Burned wool showed significant improvement compared to the control group on day 7 in the angiogenesis, on day 14 in granulation tissue formation and epithelial formation, and on day 21 in new epithelial formation (P<0.05). Burned wool compared with SSD ointment in granulation tissue formation improved significantly (P<0.05) on days 7 and 14. Also, SSD ointment in comparison with the control group significantly improved (P<0.05) granulation tissue formation and macrophage on day 7. Conclusion: Human AM has a significant effect on the treatment of second-degree burn. Burned wool has a better effect on wound healing than SSD ointment and negative control group without treatment in terms of granulation tissue and epithelium formation. [GMJ.2020;9:e1759]

Background. For centuries, medicinal plants have been extensively used in wound healing of burn injuries. The aim of this study is to analyze comparatively the effects of curcumin and Hypericum perforatum (HP) on second-degree burn wounds in rats. Materials and Methods. This experimental study was conducted on 24 male Sprague-Dawley rats with second-degree burns. The animals were randomly divided into three groups. The burns were treated with curcumin (Group B) and Hypericum perforatum (Group C) with daily application. Group A was considered as the control group and received no medication. Histological parameters, reepithelization, granulation tissue formation, inflammation, and angiogenesis were assessed after the scar biopsy at the end of research. Results. All histological parameters of the control group showed statistically significant difference than the other groups (p<0.05). There was no statistically significant difference between Groups B and C in terms of reepithelization and inflammation (p=0.351, p=0.067). In the curcumin group compared with the HP group, a significant difference was observed for two parameters of fibrosis and angiogenesis (p<0.05). Conclusion. Both curcumin and Hypericum perforatum oil are effective in burn wound healing. Our findings showed a better quality of healing in curcumin-treated rats.

Burn wound healing process consists of three phases of inflammation, proliferation and remodelling. The number of fibroblasts has increased during the proliferation phase. Oxidative stress in burn may disrupt proliferation through TGF-β inhibition, result in decreased number of fibroblasts and poor burn wound healing. Genistein in edamame seeds has anti-inflammatory, antibacterial and antioxidant properties. Edamame contains vitamins A, C, and E which is also have antioxidant properties. Edamame has been suggested to protect burn wound healing from oxidative stress. This study aimed to understand the effectiveness of edamame seed (Glycine max L. Merril) ethanolic extract (ED) to fibroblast count in second degree burn wound healing. This is true experimental research with post test only control group design. The study was conducted at University of Jember during November until December 2017. The sample of this research was 24 rat models of second degree burn which was made by attaching hot metal (60 ° C) for 5 seconds on the shaved back of the rats. The sample was divided into 6 groups with topical treatment, among others: positive control group (silver sulfadiazine), negative control group (Na CMC 0.5%), group with ED 20%, ED 40%, ED 60%, and ED 80%. Treatment was given for 15 days and skin sample was taken on the 16th day. Observation of the number of fibroblasts was done by taking pictures of histhologic preparations in Hematoxylin and Eosin staining under 400x magnification of light microscope. The number of fibroblast of each picture was processed using Image-J software. In one way ANOVA test, significance value of 0.011 (p≤0,05) indicated that there was at least a significant difference between 2 groups. LSD’s post hoc analysis showed that there was a significant difference in the group ED 20, 40, and 60% compared with negative control.   Keyword: burn wound healing, vegetable soybean, genistein, fibroblast count, silver sulfadiazine

Many researches have been exerted to find an application dressing for wound healing and also attain a considerable microbial reduction in burn wounds. In this study, the healing effect of a mixed herbal ointment (containing Myrtus, honey, Aloe vera, and pseudomonas phage) on the healing process of second degree burn wounds infected with Pseudomonas aeruginosa in comparison was evaluated. For this purpose, a hot metal square piece (4×2 cm, 50 g) was applied using a standard burning technique, and the applied pressure on the skin kept the same for all animals, then infected with Pseudomonas aeruginosa. Rats were randomly divided into 2 groups. Group 1 was treated with mixed herbal ointment, and group 2 received no treatment (control group). The treatment was daily, and sampling was weekly for three consecutive weeks (7, 14, and 21-day). Formalin 10% was used for tissue fixation. Wound healing in test and control groups was investigated by macroscopic and microscopic methods using Hematoxylin-Eosin staining. It wound contraction evaluation (Image J software). Macroscopic findings showed that wound contraction of the mixed herbal ointment group was significantly higher than the control group for 21 days. Hematoxylin-Eosin staining revealed that the epithelialization was considerably more completed in the mixed herbal ointment group in comparison with the control group. Also, neovascularization was significantly higher in the mixed herbal ointment group. The comparative results demonstrated that the mixed herbal ointment group had a significant difference (P<0.05) with a non-treated group (control). Therefore, the mixed herbal ointment is suggested as a suitable candidate for the treatment of second-degree burn wounds infected to pseudomonas aeruginosa.

Abstract Background Burn injuries are one of the most common sources of trauma globally that comprise a significant drain on long-term personal and healthcare cost Large surface area burn wounds are difficult to manage and may result in significant physiologic and psychological sequelae . The aim of this study was to compare the effectiveness of Aloe Vera gel with Nitrofurazone ointment in the healing of superficial second-degree burn wounds.Methods The present study was a split body controlled, randomized clinical trial. The sample was recruited from patients with superficial second-degree burn wound who were prescribed to treat with 2% Nitrofurazone ointment. Thirty patients with at least two burn, each burn on an alternate side of the body, entered the study — samples allocated to two groups which received Aloe Vera gel or 2% Nitrofurazone ointment on their burns. Bates-Jensen Wound assessment tool (BWAT) was used to evaluate the healing of burns. The burns were evaluated before, one, two and three weeks after the beginning of treatment.Results The mean ± SD of BWAT scores in Aloe Vera zones were 30.32 ± 3.28, 27.33 ± 3.38, 21.33 ± 3.13, 16.12 ± 2.16 respectively (F(2, 65.07) =440.00, p=0.001). The mean ± SD of BWAT scores in Nitrofurazone ointment zones were 30.51 ± 3.79, 28.45 ± 3.49, 23.36 ± 2.89, 19.23 ± 2.11 (F(1, 52.00) =228.00, p=0.001).Conclusions There is a significant difference in (BWAT) scores between intervention and control groups. Aloe Vera gel was as effective as Nitrofurazone ointment in the treatment of superficial second-degree burns.Trial registration IRCT2014113020151N1. Background: The aim of this study was to compare consequences in single and triple testicular biopsy by biopty gun in pubertal rats using histological and immunohistochemical analysis. Methods: Thirty-two Sprague-Dawley male rats were used as the experimental model. The rats were randomly divided into three study groups. The rats from the first group (n = 12) received a single-biopsy of upper pole of the left testis, while the rats from the second group (n = 10) received triple-biopsy of upper and lower poles and lateral surface of left testis. The third group (n = 10) was a control group. On the eightieth day after the biopsy in all rats bilateral orchiectomy and funiculectomy were performed to obtain testicular tissue and sperm for analysis. The consequences of the puncture were observed by pathohistology, immunohistochemistry and semen analysis. Results: The results of the study showed lower percentage of sperm count (14.5 mill/mL vs. 16 mill/mL, p = 0.130), sperm motility (24.6% vs. 32.7%, p > 0.05), abnormal sperm (30% vs. 27%, p > 0.05), atrophic tubules (21% vs. 6%, p < 0.001), volume (1.7 mL vs. 2.28 mL, p < 0.01) and apoptotic index (1.56 vs. 1.19, p = 0.650) in the testes with a triple-biopsy compared to the testes with a single-biopsy. Semen analysis showed a borderline significant difference between the group with triple-biopsy where sperm count was lower than it in the control group (14.5 mill/mL vs. 17.5 mill/mL, p = 0.05). A single-biopsy has little effect on the testis, especially on overall fertility. A triple-biopsy showed higher degree of the testicular damage but without a significant impact on overall fertility. Semen analysis showed that single- and triple-biopsies did not have a significant effect on sperm count, motility and morphology. Conclusion: Biopty gun procedure is a cheap, simple and reliable method for testicular biopsy in rats without a significant effect on sperm count, motility and morphology.
