Abstract Variation in plasma metabolite profiles can provide information on physiological state and relative rates of mass change in free-living birds caught only once, and this technique could be used to compare rates of mass change (fattening) among populations or sites in relation to habitat quality. We compared plasma metabolite levels, as an index of relative refueling rates, in migratory Arctic-nesting Western Sandpipers (Calidris mauri), on a landscape scale at as many as nine sites over two years and during three different migratory stages within the Georgia Basin-Puget Sound region of British Columbia and Washington. There was significant intersite variation in plasma triglyceride levels in both years, but only for the northward migration. By contrast, there was little evidence of intrasite variability (i.e. metabolite profiles of birds using the same site were consistent between years and migratory stages), though we documented intrasite variation resulting from birds' use of different microhabitats at the same site. Plasma glycerol levels did not vary systematically among sites, though they varied among years; on average, birds had higher glycerol levels during northward migration than during southward migration. For the northward migration only, there was a positive relationship between plasma triglyceride levels and total macrofaunal prey abundance among sites. Birds using smaller sites with a lower index of mudflat exposure (mean number of kilometer-hours of mudflat exposed in a 24-h period, reflecting the opportunity to forage at each site) tended to have lower triglyceride levels. The purpose of the present study was first to identify drivers of variance in plasma metabolite profiles of cats and dogs that may affect the interpretation of nutritional metabolomic studies. A total of fourteen cats and fourteen dogs housed in environmentally enriched accommodation were fed a single batch of diet to maintain body weight. Fasting blood samples were taken on days 14, 16 and 18 of the study. Gas chromatography–mass spectrometry (GC–MS), liquid chromatography (LC)–MS/MS and solid-phase extraction–LC–MS/MS analyses were used for metabolite profiling. Principal component (PC) analysis that indicated 31 and 27 % of the variance was explained in PC1 and PC2 for cats and dogs, respectively, with most individuals occupying a unique space. As the individual was a major driver of variance in the plasma metabolome, the second objective was to identify metabolites associated with the individual variation observed. The proportion of intra- and inter-individual variance was calculated for 109 cat and 101 dog metabolites with a low intra-individual variance (sd< 0·05). Of these, fifteen cat and six dog metabolites had inter-individual variance accounting for at least 90 % of the total variance. There were four metabolites common to both species (campesterol, DHA, a cholestenol and a sphingosine moiety). Many of the metabolites with >75 % inter-individual variance were common to both species and to similar areas of metabolism. In summary, the individual is an important driver of variance in the fasted plasma metabolome, and specific metabolites and areas of metabolism may be differentially regulated by individuals in two companion animal species.

Background: Human milk oligosaccharide supplementation safely modulates fecal bifidobacteria abundance and holds the potential to manage symptoms in irritable bowel syndrome (IBS). Here, we aimed to determine the role of a 4:1 mix of 2′-O-fucosyllactose and lacto-N-neotetraose (2′FL/LNnT) on the modulation of the gut microbiota composition and host mucosal response, as well as the link between the bifidobacteria abundance and metabolite modulation, in IBS patients. Methods: Biological samples were collected from IBS patients (n = 58) at baseline and week 4 post-supplementation with placebo, 5 g or 10 g doses of 2′FL/LNnT. The gut microbiota composition, metabolite profiles and expression of genes related to host mucosal response were determined. Results: Moderate changes in fecal, but not mucosal, microbial composition (β-diversity) was observed during the intervention with higher dissimilarity observed within individuals receiving 10g 2′FL/LNnT compared to placebo. Both fecal and mucosal Bifidobacterium spp. increased after 2′FL/LNnT intake, with increased proportions of Bifidobacterium adolescentis and Bifidobacterium longum. Moreover, the intervention modulated the fecal and plasma metabolite profiles, but not the urine metabolite profile or the host mucosal response. Changes in the metabolite profiles were associated to changes in bifidobacteria abundance. Conclusion: Supplementation with 2′FL/LNnT modulated the gut microbiota, fecal and plasma metabolite profiles, but not the host mucosal response in IBS. Furthermore, the bifidogenic effect was associated with metabolite modulation. Overall, these findings support the assertion that 2′FL/LNnT supplementation modulate the intestinal microenvironment of patients with IBS, potentially related to health.
