Talaromyces saxoxalicus sp. nov., isolated from the limestone walls of the Old Cathedral of Coimbra, Portugal

Fungi are one of the main agents of stone biodeterioration worldwide, since they strongly interfere with its integrity, aesthetical and structural natural properties. During an experimental survey aimed to isolate fungal species contributing to the biodeterioration of the limestone walls of the Old Cathedral of Coimbra (Portuguese unesco World Heritage site), a Talaromyces species that could not be identified to any currently known species in this genus was isolated. Molecular phylogenetic analysis of the internal transcribed spacer, β-tubulin and RNA polymerase II subunit 2, placed this fungus in Talaromyces sect. Purpurei, while also pointing at its phylogenetic distinction from the remaining species in this section. Thus, a novel species, Talaromyces saxoxalicus sp. nov., is here proposed. Moreover, considering the isolation source of this fungus and in an attempt to understand its contribution to the overall stone monument biodeterioration, the species's in vitro biodeteriorative potential was also evaluated. The results highlighted that the species exhibited an in vitro biodeteriorative ability (calcium oxalate crystal formation), highlighting its potential deteriorative profile.

1. A low urinary magnesium was induced in normal volunteer subjects by giving cellulose phosphate; magnesium was added in vitro to yield urine samples of normal and high magnesium concentrations 2. After rapid evaporation of these urine samples at pH 5.3 to standard osmolality the calcium oxalate crystals were measured by microscopy and isotopic methods 3. There was a clear inverse correlation between magnesium concentration and calcium oxalate crystal formation 4. The case for treating calcium oxalate urolithiasis with magnesium is strengthened.

The ion content of drinking water might be associated with urinary stone formation, representing a keystone of conservative nephrolithiasis management. However, the effects of specific ions on calcium oxalate crystal formation and their mechanism of action are still highly controversial. We report an investigation of the effects of oligomineral waters with similar total salt amount but different ion composition on calcium oxalate (CaOx) precipitation in vitro, combining gravimetric and microscopic assays. The results suggest that the “collective” physicochemical properties of the aqueous medium, deriving from the ion combination rather than from a single ionic species, are of importance. Particularly, the ability of ions to strengthen/weaken the aqueous medium structure determines an increase/decrease in the interfacial energy, modulating the formation and growth of CaOx crystals.

Rapeseed (Brassica napus subsp. napus) is one of the autumn crops in Iran. A new fungal species isolated from internal tissues associating a typical leaf spot of Rapeseed in Dezful (southwestern Iran) was described and illustrated. The sampling and isolation were performed in the late fall of 2019 (during December), which led to obtain the colonies with the similar morphology. Molecular phylogenetic analyses based on a combined matrix of the internal transcribed spacer regions 1 and 2 including the intervening 5.8S nuclear ribosomal DNA (ITS), and partial regions of the β-tubulin (tub2) and RNA polymerase II second largest subunit (rpb2) indicated that two isolated strains represent a new Plenodomus species in family Leptosphaeriaceae (Pleosporales). These novel strains formed a sister lineage with P. biglobosus in our phylogenetic analyses, while, observed morphology is distinguished the taxa from P. biglobosus by its pycnidia in having hyphal outgrowths and lacking cylindrical neck. Accordingly, P. dezfulensis sp. nov. was introduced and its pathogenicity was investigated. Pathogenicity test with both P. dezfulensis strains on three-week seedlings of Rapeseed reproduced leaf spot symptoms. These results approve P. dezfulensis as the causal agent of leaf spot observed on Rapeseed in Dezful.

The Sassi, a UNESCO World Heritage Site and its rupestrian churches, are richly decorated and visited by thousands of visitors every year. It is important to preserve this heritage which shows signs of deterioration due to abiotic and/or biotic factors. Aiming to carry out in the future an environmental-friendly restoration, a screening of the fungi present on walls and frescoes of two rupestrian churches “Santa Lucia alle Malve” and “La Madonna dei derelitti” located, respectively, in the “Sasso Caveoso” and in the “Sasso Barisano” was performed. Isolation and characterization of fungal species from investigated sites was carried out. Total genomic DNA (gDNA) was extracted from pure fungal cultures and subsequently utilized in PCRs using primers that amplify a portion of the ribosomal DNA (ITS5/ITS4) or the β-tubulin gene (Bt2a/Bt2b). The amplicons were directly sequenced. Obtained nucleotide sequences were compared to those present in the GenBank (NCBI) showing a very high similarity (99–100%) with the following species: Parengyodontium album, Alternaria alternata, Cladosporium cladosporioides, Lecanicillium psalliotae, Meyerozyma guilliermondii and Botryotrichum atrogriseum. All sequences from this study were deposited in the EMBL database. Detailed knowledge about fungi isolated from stone is indispensable not only to counter/reduce the structural and aesthetic damage but also to protect the health of both guardians and visitors who may develop different pathologies due to the spores diffused in the environment.

Tourism is an important component of the Indonesian economy as well as a significant source of the country’s foreign exchange revenues. According to the Center of Data and Information - Ministry of Culture and Tourism, the growth of foreign visitor arrivals to Indonesia has increased rapidly by 9.61 percent since 2010 to the present. One of the most potential tourism destinations is Komodo Island located in East Nusa Tenggara. With the island’s unique qualities, which include the habitat of the Komodo dragons and beautiful and exotic marine life, it is likely to be one of the promising tourism destinations in Indonesia and in the world. In 1986, the island has been declared as a World Heritage Site by UNESCO. The Ministry of Culture and Tourism continuously promotes many of the country’s natural potential in tourism through various media: printed media, television and especially new media. However, there are challenges for the Indonesian tourism industry in facilitating entrepreneurship skills among the local people in East Nusa Tenggara. According to the Central Bureau of Statistics (2011), East Nusa Tenggara is considered as one of the poorest provinces in Indonesia where the economy is lower than the average, with a high inflation of 15%, and unemployment of 30%. This research is needed to explore further the phenomenon behind the above facts, aiming at examining the role of new media in facilitating entrepreneurship in the tourism industry in Komodo Island.
