Campylobacter fetus subsp. fetus infection can occur in female sheep, causing infertility or abortion. Despite extensive research on the effect of these bacteria on female fertility, little research has been done on the influence of C. fetus subsp. fetus on the male factor. Our objective was to examine the influence of C. fetus subsp. fetus on ram sperm. Motility index, percentage of live spermatozoa, mean αt value (an indication of the chromatin stability of the sperm cell) and percentage of sperm cells expressing the FAS receptor were measured in sperm incubated in the presence or absence of C. fetus subsp. fetus. The motility index and viability of sperm incubated with the bacteria were lower than those of untreated sperm samples after 5 h. In bacteria-incubated sperm cells, the percentage expressing FAS receptor was already significantly elevated at 15 min. Bacteria-incubated sperm showed a greater prevalence of morphological damage. The bacteria were attached to tail and acrosome regions, and the sperm damage was concentrated in both the motility and chromatin regions. Bacteria-infected sperm cells showed a decrease in motility, increase in early acrosome reaction and chromatin damage. Similar effects were induced by incubation of the sperm with supernatants from C. fetus subsp. fetus cultures. Thus this study demonstrates that C. fetus subsp. fetus has a detrimental effect on the quality of ram sperm.

AbstractApproximately half of the cases of infertility are due to male factor. In many cases the underlying cause of male infertility is not discovered and, therefore, the condition is considered idiopathic. Examination of morphology, motility, concentration and total sperm count is very important but not sufficient for complex men fertility evaluation. Sperm DNA integrity is a very important one. Sperm DNA can be fragmented by several internal or external factors. In immature sperm cells, the DNA can be repaired by reparatory mechanisms of spermatogonia or spermatocytes. However, in a haploid mature sperm cell, these fragments can not be repaired by male and the fragmented DNA is transferred to oocyte. Oocytes are able to repair male fragmented DNA after their fertilization. A quality embryo can repair damaged sperm DNA and the repair system depends on cytoplasmic and genomic quality of the oocyte. The ability of oocyte to repair sperm DNA strong depend on quality of fertilized oocytes.Running title: Oocyte and DNA repair

In recent years, the study of male factor in conjugal infertility, has received more attention since it was recognized as an important cause of infertility in 30-50% of the cases; and to the diminishing quality of semen, observed during the last decades, possibly related to environmental polution.Alterations in sperm motility has been included as a direct cause of male infertility along with the traditionally known causes. Frequently these alterations have been related to morphological changes of spermatozoa. The morphology and size of sperm cells are higly variable in human semen and abnormal forms also can be found in fertile men. Experimental data demonstrate that abnormal spermatozoa are not functional cells. There are many clinical reports that associate spermatic dismorphy with infertility. Previously reported abnormalities in the sperm morphology related to infertility include: acrosomal agenesis, defects in the dynein arms and fibrous sheath displasia. Therefore, it is important to undertake the ultrastructural study of spermatozoa to determine subcellular pathology of the gamete which could explain the reproductive failure, specially if there is no evident cause of the motility defect or if there has been no response to treatment, or if the assisted reproductive programs have failed repeatedly.

Sperm cells that cannot swim and orient properly compromise male fertility. Such defects are responsible for male infertility regardless of the actual quality of the most important content, the sperm’s DNA. Synthetic micromotors are engineered devices that are able to swim in (body) fluids and microscopic environments, similar to flagellated cells like sperm. Coupled together, a sperm-hybrid micromotor embodies the concept of bringing the sperm cell together with artificial components that assist or replace defective functions of the cell, helping it to pursue its goal without interfering with its health, enabling the process of assisted fertilization and further embryo development all inside the body. Non-invasive, remote-controlled in vivo applicability is the key quality of such hybrid microdevices. Assisted reproduction with the help of micromotors is in the focus of this review, although other biomedical applications that arise from the powerful combination of sperm cell and synthetic enhancement are also discussed and summarized. Details are provided about different fabrication processes and cell-material coupling strategies, and the way from proof-of-concept studies to in vivo experiments in animals is outlined.

AbstractDuring the double fertilization process, pollen tubes deliver two sperm cells to an ovule containing the female gametes. In the pollen tube, the vegetative nucleus and sperm cells move together to the apical region where the vegetative nucleus is thought to play a crucial role in controlling the direction and growth of the pollen tube. Here, we report the generation of pollen tubes in Arabidopsis thaliana whose vegetative nucleus and sperm cells are isolated and sealed by callose plugs in the basal region due to apical transport defects induced by mutations in the WPP domain-interacting tail-anchored proteins (WITs) and sperm cell-specific expression of a dominant mutant of the CALLOSE SYNTHASE 3 protein. Through pollen-tube guidance assays, we show that the physiologically anuclear mutant pollen tubes maintain the ability to grow and enter ovules. Our findings provide insight into the sperm cell delivery mechanism and illustrate the independence of the tip-localized vegetative nucleus from directional growth control of the pollen tube.

Objectives: We explored the safety of Tadalafil once-daily treatment for 12 week and its effects on semen quality in a clinical subpopulation of men with psychogenic erectile dysfunction (ED). Patients and Methods: Twenty-seven men, between 19 and 35 years, unaware of their fertility status, suffering from psychogenic ED were enrolled. The examination of the seminal fluid was performed twice before administration of Tadalafil and twice after three months of continuous daily administration of Tadalafil 5 mg. The volume of the seminal fluid, the concentration of sperm, the concentration of sperm with type “a+b” motility, the concentration of sperm with type “a” motility, the concentration of normal sperm were taken into consideration. The results before treatment with Tadalafil (T0) and after 3 months of treatment (T3m) were compared. Results: The administration of once-daily Tadalafil 5mg, brings to an average increase of the total number of sperm cells, both total and fast motility (type a) and the percentage of nemasperms, and to an average increase of semen volume of only 0.41 ml. These quantitative and qualitative improvements of the seminal fluid resulted statistically significant as regard motility, nemasperm percentage and seminal fluid volume. No unespected safety findings were observed. Conclusions: Tadalafil administration improves the quality of sperm cells and seminal fluid: in particular motility, percentage of nemasperms and volume of seminal fluid. We emphasize the safety of the once-daily treatment with tadalafil 5mg and the positive effects on spermatogenesis.

ABSTRACT: Bovine genital campylobacteriosis (BGC) is a venereal and subclinical disease that affects the fertility of cattle herds, and it is caused by Campylobacter fetus subsp. venerealis . This study selected peptides mimetic to the BGC-causing agent from a phage library. Phage display is a technique that applies bacteriophage libraries that reveal peptides fused to the viral capsid in biological selections against target proteins. Biopannings were performed for biological selection in the phage library using rabbit hyperimmune serum and C. fetus subsp. venerealis protein extract. Five selected heptapeptides were considered mimetic to Cfv-NCTC 10354 based on the results of bioinformatics analysis and assays with hyperimmune serum and cervicovaginal mucus obtained from heifers. ALASLPL and LSYLFPP were the most reactive peptides and considered promising as possible mimetic immunogens for C. fetus subsp. venerealis.

This study aims to knowing influence using cocentration of coconut water in physiology of NaCl dilution toquality of spermatozoa of Puntius javanicus. This research was counducted at Meunasah Krueng Village, Beutong Subdistrict, Nagan Raya District from 18th – 20th May 2015. Thecompletely randomized design with 5 treatmentand 3 replication was used for this study. Sample was saved in refrigerator (3 days) with temperature 4oC and it is oberved sample quality and data was collected in daily. Result of the research show water coconut concentration (7%) or in treatment 4th (P4) giving result for best quality of spermatozoa, where motility index average of spermatozoa is up to 80,55%, viability value of spermatozoa is up to 65,88%. In addtion, coconut water in in physiology of NaCl dilution have been affecting to spermatozoa quality of Puntius javanicus.
