Ovate family proteins (OFPs) are a family of plant growth regulators that play diverse roles in many aspects of physiological processes. OFPs have been characterized in various plant species including tomato, Arabidopsis, and rice. However, little is known about OFPs in woody species. Here, a total of 30 PtOFP genes were identified from the genome of Populus trichocarpa and were further grouped into four subfamilies based on their sequence similarities. Gene expression analysis indicated that some members of the PtOFP gene family displayed tissue/organ-specific patterns. Analysis of cis-acting elements in the promoter as well as gene expression by hormone treatment revealed putative involvement of PtOFPs in hormonal response. Furthermore, PtOFP1 (Potri.006G107700) was further experimentally demonstrated to act as a transcriptional repressor. Yeast two-hybrid assay showed physical interactions of PtOFP1 with other proteins, which suggests that they might function in various cellular processes by forming protein complexes. In addition, overexpression of PtOFP1 in Arabidopsis conferred enhanced tolerance to PEG-induced drought stress at seedling stage, as well as a higher survival rate than the wild type at mature stage. These results provide a systematic analysis of the Populus OFP gene family and lay a foundation for functional characterization of this gene family.

Functional characterization of plant agrichemical transporters provided an opportunity to discover molecules that have a high mobility in plants and have the potential to increase the amount of pesticides reaching damage sites. Agrobacterium-mediated transient expression in tobacco is simple and fast, and its protein expression efficiency is high; this system is generally used to mediate heterologous gene expression. In this article, transient expression of tobacco nicotine uptake permease (NtNUP1) and rice polyamine uptake transporter 1 (OsPUT1) in Nicotiana benthamiana was performed to investigate whether this system is useful as a platform for studying the interactions between plant transporters and pesticides. The results showed that NtNUP1 increases nicotine uptake in N. benthamiana foliar discs and protoplasts, indicating that this transient gene expression system is feasible for studying gene function. Moreover, yeast expression of OsPUT1 apparently increases methomyl uptake. Overall, this method of constructing a transient gene expression system is useful for improving the efficiency of analyzing the functions of plant heterologous transporter-encoding genes and revealed that this system can be further used to study the functions of transporters and pesticides, especially their interactions.

The stability and value of transgenic pest resistance for promoting tree growth are poorly understood. These data are essential for determining if such trees could be beneficial to commercial growers in the face of substantial regulatory and marketing costs. We investigated growth and insect resistance in hybrid poplar expressing the cry3Aa transgene in two field trials. An initial screening of 502 trees comprising 51 transgenic gene insertion events in four clonal backgrounds (Populus trichocarpa × Populus deltoides, clones 24-305, 50-197, and 198-434; and P. deltoides × Populus nigra, clone OP-367) resulted in transgenic trees with greatly reduced insect damage. A large-scale study of 402 trees from nine insertion events in clone OP-367, conducted over two growing seasons, demonstrated reduced tree damage and significantly increased volume growth (mean 14%). Quantification of Cry3Aa protein indicated high levels of expression, which continued after 14 years of annual or biannual coppice in a clone bank. With integrated management, the cry3Aa gene appears to be a highly effective tool for protecting against leaf beetle damage and improving yields from poplar plantations.
