To address which mitochondria-related nuclear differentially expressed genes (DEGs) and related pathways are altered during human oocyte maturation, single-cell analysis was performed in three oocyte states: in vivo matured (M-IVO), in vitro matured (M-IVT), and failed to mature in vitro (IM-IVT). There were 691 DEGs and 16 mitochondria-related DEGs in the comparison of M-IVT vs. IM-IVT oocytes, and 2281 DEGs and 160 mitochondria-related DEGs in the comparison of M-IVT vs. M-IVO oocytes, respectively. The GO and KEGG analyses showed that most of them were involved in pathways such as oxidative phosphorylation, pyruvate metabolism, peroxisome, and amino acid metabolism, i.e., valine, leucine, isoleucine, glycine, serine, and threonine metabolism or degradation. During the progress of oocyte maturation, the metabolic pathway, which derives the main source of ATP, shifted from glucose metabolism to pyruvate and fatty acid oxidation in order to maintain a low level of damaging reactive oxygen species (ROS) production. Although the immature oocytes could be cultured to a mature stage by an in vitro technique (IVM), there were still some differences in mitochondria-related regulations, which showed that the mitochondria were regulated by nuclear genes to compensate for their developmental needs. Meanwhile, the results indicated that the current IVM culture medium should be optimized to compensate for the special need for further development according to this disclosure, as it was a latent strategy to improve the effectiveness of the IVM procedure.

Soil microbial communities are essential to phosphorus (P) cycling, especially in the process of insoluble phosphorus solubilization for plant P uptake. Phosphate-solubilizing microorganisms (PSM) are the dominant driving forces. The PSM mediated soil P cycling is easily affected by water condition changes due to extreme hydrological events. Previous studies basically focused on the effects of droughts, floods, or drying-rewetting on P cycling, while few focused on drought-flood abrupt alternation (DFAA), especially through microbial activities. This study explored the DFAA effects on P cycling mediated by PSM and P metabolism-related genes in summer maize field soil. Field control experiments were conducted to simulate two levels of DFAA (light drought-moderate flood, moderate drought-moderate flood) during two summer maize growing periods (seeding-jointing stage, tasseling-grain filling stage). Results showed that the relative abundance of phosphate-solubilizing bacteria (PSB) and phosphate-solubilizing fungi (PSF) increased after DFAA compared to the control system (CS), and PSF has lower resistance but higher resilience to DFAA than PSB. Significant differences can be found on the genera Pseudomonas, Arthrobacter, and Penicillium, and the P metabolism-related gene K21195 under DFAA. The DFAA also led to unstable and dispersed structure of the farmland ecosystem network related to P cycling, with persistent influences until the mature stage of summer maize. This study provides references for understanding the micro process on P cycling under DFAA in topsoil, which could further guide the DFAA regulations.

The HAFS model is an effort under the NGGPS and UFS initiatives to create the next generation of hurricane prediction and analysis system based on FV3-GFS. It has been validated extensively using traditional verification indicators such as tracker error and biases, intensity error and biases, and the radii of gale, damaging and hurricane strength winds. While satellite images have been used to verify hurricane model forecasts, they have not been used on HAFS. The community radiative transfer model CRTM is used to generate model synthetic satellite images from HAFS model forecast state variables. The 24 forecast snapshots in the mature stage of hurricane Dorian in 2019 are used to generate a composite model synthetic GOES-R infrared brightness image. The composite synthetic image is compared to the corresponding composite image generated from the observed GOES-R data, to evaluate the model forecast TC vortex intensity, size, and asymmetric structure. Results show that the HAFS forecast TC Dorian agrees reasonably well with the observation, but the forecast intensity is weaker, its overall vortex size smaller, and the radii of its eye and maximum winds larger than the observed. The evaluation results can be used to further improve the model. While these results are consistent with those obtained by traditional verification methods, evaluations based on composite satellite images provide an additional benefit with richer information because they have near-real-times spatially and temporally continuous high-resolution data with global coverage. Composite satellite infrared images could be used routinely to supplement traditional verification methods in the HAFS and other hurricane model evaluations. Note since this study only evaluated one hurricane, the above conclusions are only applicable to the model behavior of the mature stage of hurricane Dorian in 2019, and caution is needed to extend these conclusions to expect model biases in predicting other TCs. Nevertheless, the consistency between the evaluation using composite satellite images and the traditional metrics, of hurricane Dorian, shows that this method has the potential to be applied to other storms in future studies.

In this paper, we propose a deep learning-based model to detect extratropical cyclones (ETCs) of the northern hemisphere, while developing a novel workflow of processing images and generating labels for ETCs. We first labeled the cyclone center by adapting an approach from Bonfanti et al. in 2017 and set up criteria of labeling ETCs of three categories: developing, mature, and declining stages. We then gave a framework of labeling and preprocessing the images in our dataset. Once the images and labels were ready to serve as inputs, an object detection model was built with Single Shot Detector (SSD) and adjusted to fit the format of the dataset. We trained and evaluated our model with our labeled dataset on two settings (binary and multiclass classifications), while keeping a record of the results. We found that the model achieves relatively high performance with detecting ETCs of mature stage (mean Average Precision is 86.64%), and an acceptable result for detecting ETCs of all three categories (mean Average Precision 79.34%). The single-shot detector model can succeed in detecting ETCs of different stages, and it has demonstrated great potential in the future applications of ETC detection in other relevant settings.

Air pollution in China has become a matter of increasing public concern. In this paper, we attempted to build a theoretical model to explore the impact of the dynamics of agglomeration externalities on haze pollution in urban China, where agglomeration is differentiated by regional specialization and geographical concentration. Based on China’s panel data for 289 cities during the period of 1998–2018, the empirical result shows that the relationship between industrial agglomeration and urban haze pollution is not simply linear or of an inversed U-type but turns out to be dynamically N-shaped. To be specific, the increase in local haze pollution can be explained by agglomeration externalities in the beginning stage, whereas the reducing effect only occurs during the mature stage. The heterogeneity test indicated that the effect of the type of agglomeration on haze pollution seems to be mixed in different groups of cities, but is still consistent with the hypothesis of the dynamic change of agglomeration externalities. The results are found to be quite robust and consistent after replacing variables and using other regression methods. This paper provides answers to the question of how to coordinate the relationship between developing industry parks and air pollution in terms of the life cycle of agglomeration as well as the types of city.

Artemisinin resistance in Plasmodium falciparum has emerged and spread widely in the Greater Mekong Subregion threatening current first line artemisinin combination treatments. New antimalarial drugs are needed urgently. Cipargamin (KAE609) and ganaplacide (KAF156) are promising novel antimalarial compounds in advanced stages of development. Both compounds have potent asexual blood stage activities, inhibit P. falciparum gametocytogenesis and reduce oocyst development in anopheline mosquitoes. In this study, we compared the asexual and sexual stage activities of cipargamin, ganaplacide and artesunate in artemisinin resistant P. falciparum isolates (N=7, K13 mutation; C580Y, G449A and R539T) from Thailand and Cambodia. Asexual blood stage antimalarial activity was evaluated in a SYBR-green I based 72h in vitro assay, and the effects on male and female mature stage V gametocytes were assessed in the P. falciparum dual gamete formation assay. Ganaplacide had higher activities when compared to cipargamin and artesunate, with a mean (SD) IC50 against asexual stages of 5.5 (1.1) nM, 7.8 (3.9) nM for male gametocytes and 57.9 (59.6) nM for female gametocytes. Cipargamin had a similar potency against male and female gametocytes, with a mean (SD) IC50 of 123.1 (80.2) nM for male gametocytes, 88.5 (52.7) nM for female gametocytes and 2.4 (0.6) nM for asexual stages. Both cipargamin and ganaplacide showed significant transmission-blocking activities against artemisinin resistant P. falciparum in vitro .

It is an important goal of China’s 2025 Strategy to build an environmental governance system with the government as the leading role, enterprises as the main body, and social organizations and the public all participate actively. In this context, whether and how small and micro enterprises, as a force of rejuvenating the country through science and technology, participate in the construction of environmental governance system is discussed in this paper. This study uses the 12th China small and micro enterprise survey database to deeply analyze the direct and indirect effects between green innovation and small and micro enterprise performance, and finds that: in the direct effect analysis, the direct promotion effect of green innovation on the financial performance of small and micro enterprises is related to the development stage of small and micro enterprises, which is significant in the initial stage and mature stage, but not significant in the growth stage; in the indirect effect analysis, green innovation can indirectly improve the overall performance of small and micro enterprises by improving their environmental and social responsibility performance. At the same time, green innovation can amplify the role of other factors in promoting the overall performance. This study not only enriches the research scope of green innovation theory from the subdivision dimension, provides a new perspective for the integration of green innovation and social responsibility theory, it also provides theoretical reference and guidance for the green innovation practice of small and micro enterprises.

Empirical findings from the impact of innovation investment volatility on enterprise technological innovation are mixed. Based on the punctuated equilibrium theory, this study explores the impact of innovation investment volatility on enterprise technological innovation in different life cycles and whether innovation subsidy has expected effects on enterprises’ technological innovation. By using the 205 Chinese listed enterprises in strategic emerging industries from 2010 to 2019 as the research sample, the results show that the innovation investment volatility has a positive impact on technological innovation of enterprise in the growing stage, while it has no significant effect on enterprise technological innovation in the mature and declining stages. In addition, the negative moderating effect of innovation subsidy on the relationship between innovation investment volatility and technological innovation is the most significant for enterprises in the growing stage, weakly significant for enterprises in the mature stage, and insignificant for enterprises in the declining stage.

PurposeThe paper analyses the dramatic global slowdown of tourism caused by the pandemic and how it has provided an exceptional opportunity for the tourism sector to reassess both its priorities and practices, especially in relation to our planet and society.Design/methodology/approachThis paper provides an analysis of the European Travel Commission's position regarding the development of a sustainable tourism ecosystem and the definition of new metrics to measure this transition, thus allowing European National Tourism Organisations (NTOs) to share knowledge and learn from each other.FindingsNTOs across Europe are at different stages in their journey towards a sustainable tourism development model. Some destinations are still focussing on attracting more travellers from international markets while others at a more mature stage, are switching their work towards the management of tourism flows. These latter examples support the approach that developing a tourism strategy should start with the needs of the destination, and the local people should be directly and fully involved in the tourism strategy. Initiatives such as this can support tourism decision-making based on data and firm evidence and help to address doubts and common misunderstandings around how residents feel.Originality/valueThis paper highlights how the European Travel Commission, together with its members, are working towards a new world benchmark for responsible, sustainable and innovative tourism in response to exponential growth of demand, the reality of the ecological transition and the emergence of a new digital era.
