Indicator Plant and PCR-RAPD for Biotype Determination of Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae).B. tabaci has been known world wide as a major pest and virus vector of horticulture. In Indonesia the presence of B.tabaci was reported since 1980 and its role as virus vector in tomato and chilli pepper has becoming more importantrecently. Genetic diversity of B. tabaci has been well recognized, but very little information available for diversity of B.tabaci in Indonesia. This research was conducted in Bogor, West Java from May 2004 to June 2005. The aim of thisresearch was to initiate basic information regarding genetic diversity of B. tabaci in Indonesia, particularly in Java Island.Whiteflies population collected from different crops, i.e. tomato, broccoli, chill pepper, eggplant, cucumber, soybean, andedamame, was evaluated using silverleaf-induction test, and RAPD-PCR. It was evidenced that only B. tabaci populationfrom broccoli was able to induce silverleaf. Two genetic types of B. tabaci, i.e. biotype B and non B, were identified basedon polymorphism character of DNA. Population from broccoli was belong to biotype B, whereas other populations fromtomato, chill pepper, eggplant, cucumber, soybean, and edamame were belong to biotype non B.

The flight activities of adult sweetpotato whitefly, Bemisia tabaci, and leafhoppers were monitored by plastic cup traps at Boğalı, Taşçı, Hacıali, Doğankent and Balcalı in the Çukurova Plain, Turkey, in 2001 and 2003. Activity of B. tabaci, expressed as numbers of adults caught in traps, was low from May to early July in both years. Numbers of B. tabaci caught at Tasçı were higher than at Boğalı from 10 July and 21 August in 2001. Its numbers were also higher at Doğankent than at Hacıali and Balcalı during August of 2003. The numbers of leafhopper adults caught fluctuated greatly in both years. Numbers remained low until late June, followed by gradual increases in July and August at Boğalı and Taşçı in 2001. In contrast, numbers of adults caught were higher at Hacıali, Doğankent and Balcalı from May through July in 2003, followed by lower catches during the remainder of the season. Daily minimum temperatures in July and August were positively correlated with higher trap catches of both B. tabaci and leafhoppers.

The assessment of the genetic diversity of cultivated varieties is a very important element of breeding programs. This allows the determination of the level of genetic differentiation of cultivated varieties, their genetic distinctiveness, and is also of great importance in the selection of parental components for crossbreeding. The aim of the present study was to determine the level of genetic diversity of oat varieties currently grown in Central Europe based on two marker systems: ISSR and SCoT. The research conducted showed that both these types of markers were suitable for conducting analyses relating to the assessment of genetic diversity. The calculated coefficients showed that the analyzed cultivars were characterized by a high genetic similarity. However, the UPGMA and PCoA analyses clearly indicated the distinctiveness of the breeding programs conducted in Central European countries. The high genetic similarity of the analyzed forms allow us to conclude that it is necessary to expand the genetic pool of oat varieties. Numerous studies show that landraces may be the donor of genetic variation.

The aim of this study was to assess genetic structure of Slovak Pinzgau populationbased on polymorphism at molecular markers using statistical methods. Femaleoffspring of 12 most frequently used bulls in Slovak Pinzgau breeding programmewere investigated. Pinzgau cattle were found to have a high level of diversity,supported by the number of alleles observed across loci (average 5.31, range 2-11)and by the high within-breed expected heterozygosity (average 0.66, range 0.64-0.73). The state of genetic diversity is satisfying and standard for local populations.Detection of 12 possible subpopulation structures provided us with detailedinformation of the genetic structure. The Bayesian approach was applied, detectingthree, as the most probable number of clusters. The similarity of eachsubpopulation using microsatellites was confirmed also by high-throughputmolecular data. The observed inbreeding (FROH=2.3%) was higher than thatexpected based on pedigree data (FPED=0.4%) due to the limited number ofavailable generations in pedigree data. One of the most important steps indevelopment of efficient autochthonous breed protection programs ischaracterization of genetic variability and assessment of the population structure.The chosen set of microsatellites confirmed the suitability in determination of thesubpopulations of Pinzgau cattle in Slovakia. The state of genetic diversity at moredetailed level was successfully performed using bovineSNP50 BeadChip.

Background: Agricultural research chiefly focuses on the ways to increase productivity of staple food crops like wheat and rice, but still there are crops where research focus is meagre like nutritionally important mungbean crop grown by marginal farmers in crop rotation systems. Mungbean is leguminous crop which is high in protein content thus it offers health benefits at cheaper rates. The present work emphasizes on finding genetic diversity in mungbean germplasm on the basis of chemical and molecular analysis for micronutrients variation (iron and zinc). The identified molecular markers having linkage with high iron and zinc concentrations in the seeds can prove helpful in expansion of biofortification programme.Methods: Fifty-one green gram genotypes viz. varieties released from CCS Haryana Agricultural University (HAU), Hisar, Punjab Agricultural University (PAU), Ludhiana, Indian Institute of Pulse Research (IIPR), Kanpur and some advanced breeding lines were included in the study. Acid digested samples were used for determination of Fe and Zn by Atomic Absorption Spectrophotometer. Young seedlings leaves were used for isolation of genomic DNA using 2% CTAB (cetyltrimethyl ammonium bromide). Result: Total of fifty-one mungbean genotypes were tested using fifty simple sequence repeat (SSR) primers. Out of fifty primers screened, 16 primers generated 35 bands. Iron (Fe) and zinc (Zn) in mungbean lines was 36.90 to 107.1 mg/kg and 14.2 to 53.8 mg/kg respectively. The molecular studies based on SSR markers also indicates existence of ample genetic diversity at molecular level.

Understanding the genetic diversity and population structure of fish species is crucial for the sustainable use and protection of fish germplasm resources. Hemibagrus guttatus (Bagridae, Siluriformes) is widely distributed in the large subtropical Pearl River (China) and is commercially important. It's population have been declining. The genetic diversity of wild H. guttatus is not clear, despite its important ecological significance. In this paper, genes mitochondrial cytochrome c oxidase subunit I (COI) and cytochrome b (Cyt b) were used to analyze the genetic structure of H. guttatus population collected from six geographical populations in the main streams of the Pearl River. The results showed that the nucleotide diversity (π) and haplotype diversity (Hd) of wild H. guttatus was low (π < 0.005; Hd < 0.5). In addition, H. guttatus haplotypes did not cluster into clades according to geographical distribution, as revealed by neighbor-joining tree analysis. Analysis of molecular variance analysis (AMOVA) and F-statistics (Fst) values showed high homogeneity among wild H. guttatus populations. Our results suggest that there is degradation in germplasm resources of H. guttatus that could destabilize the sustainable use of this species and there was an urgent need for conservation of this species in South China.

Rice false smut caused by Ustilaginoidea virens is currently one of the most devastating fungal diseases of rice panicles worldwide. In this study, two novel molecular markers derived from SNP-rich genomic DNA fragments and a previously reported molecular marker were used for analyzing the genetic diversity and population structure of 167 U. virens isolates collected from nine areas in Sichuan-Chongqing region, China. A total of 62 haplotypes were identified, and a few haplotypes with high frequency were found and distributed in two to three areas, suggesting gene flow among different geographical populations. All isolates were divided into six genetic groups. The groups Ⅰ and Ⅵ were the largest including 61 and 48 isolates, respectively. The pairwise FST values showed significant genetic differentiation among all compared geographical populations. AMOVA showed that intergroup genetic variation accounted for 40.17% of the total genetic variation, while 59.83% of genetic variation came from intragroup. The UPGMA dendrogram and population structure revealed that the genetic composition of isolates collected from ST (Santai), NC (Nanchong), YC (Yongchuan), and WS (Wansheng) dominated by the same genetic subgroup was different from those collected from other areas. In addition, genetic recombination was found in a few isolates. These findings will help to improve the strategies for rice false smut management and resistance breeding, such as evaluating breeding lines with different isolates or haplotypes at different elevations and landforms.
