Isolation and identification of rumen bacteria capable of anaerobic rutin degradation

Fifteen strains of bacteria capable of degrading rutin anaerobically were isolated from bovine rumen contents and identified by morphological and biochemical evidence as strains of Butyrivibrio sp. Three cultures from a laboratory collection of 53 strains of rumen bacteria also used rutin anaerobically. Two, Butyrivibrio fibrisolvens D1 and Selenomonas ruminantium GA192, cleaved the glycosidic bond of rutin and fermented the sugar but did not degrade the insoluble aglycone produced; the third strain, Peptostreptococcus sp. B178, degraded the substrate to soluble products. Butyrivibrio sp. C3 degraded rutin, quercitrin, and naringin to water-soluble products, showing that the organism cleaved the heterocyclic ring of these compounds. Butyrivibrio sp. C3 fermented the sugar moiety of hesperidin but did not cleave the heterocyclic ring. It did not attack quercetin, taxifolin, protocatechuic acid, or phloroglucinol. In a medium containing rumen fluid, Butyrivibrio sp. C3 degraded rutin more than twice as fast as it did in a medium containing enzymatic casein hydrolyzate, volatile fatty acids, yeast extract, and hemin in place of rumen fluid.The observations reported in this paper are believed to represent the first recorded demonstration of degradation of the heterocyclic ring structure of rutin and other bioflavonoids in pure cultures of anaerobic bacteria.

Three groups of five rumen and duodenum cannulated fauna-free sheep were used in a 28d experiment. One group remained fauna-free, whereas the second (EN) and third (PP) groups, respectively, were inoculated intraruminally with the protozoan species Entodinium caudatum and Polyplastron multivesiculatum. Rumen fluid, duodenal digesta and faecal samples were collected during the last 12d. The flow of digesta to the duodenum was determined using Yb and Co as dual-phase markers. 15Nitrogen and phosphatidylcholine were used as markers to calculate the duodenal flow of bacterial and protozoal N, respectively. Results showed an increase (P<0·1) in the rumen concentration of NH3-N and total volatile fatty acids, and a decrease (P<0·05) in the duodenal flow of non-NH3-N and bacterial N in sheep with EN and PP monofaunas, compared with fauna-free sheep. There were no differences (P>0·05) in these variables between the two monofauna groups. Protozoal N accounted for 8% of the duodenal non-NH3-N flow in the EN-monofaunated sheep, whereas no such flow was detected in the PP-monofaunated sheep. Apparent rumen digestibility of organic matter, neutral detergent fibre and acid detergent fibre were similar (P>0·05) in the monofaunated groups of sheep, but rumen acid detergent fibre digestibility was higher (P<0·05) in the monofaunated than in the fauna-free groups. Experimental results suggested that, unlike EN, the PP monofauna might not contribute to the duodenal flow of microbial protein, whereas both monofaunas showed a virtually equal degree of predation on rumen bacteria.

The aim of this study was to investigate the prevalence and therapeutic management of indigestion in milch cows in and around Bhubaneswar, Odisha. Crossbred milch cows (n=257) with the history of anorexia presented to Teaching Veterinary Clinical Complex, College of Veterinary Science and Animal Husbandry, OUAT, Bhubaneswar and in private herds in and around Bhubaneswar were examined for indigestion following standard procedures to identify the type of indigestion leading to decrease in milk yield. Clinical examination, determination of pH, study of rumen protozoa, rumen bacteria were carried out in three different types of indigestion. Amongst the affected animals, 48.33 % (n=58), 21.67 % (n=26) and 30.00 % (n=36) were detected to be affected with simple, acid and alkaline indigestion, respectively. Simple indigestion was higher (p < 0.05) as compared to acid and alkaline and alkaline indigestion (30 %) was found to be the next to simple indigestion. Ruminal movement varied from 1-4 per 5 min and pH of the rumen fluid ranged from 7.5-8.6 and the idophillic activity of the rumen protozoa was moderate type (+++). Average total count of protozoa was estimated to be 3.5 - 4.6 x 105 per ml with an average of 4.1 x 105 per ml of rumen liquor and drugs used in the study were found efficacious to treat the different types of indigestion.. Rumen bacteria have played role resulting in production of volatile fatty acids and lactic acid thereby decreasing / disturbing the rumen pH to non-physiological levels, reducing efficiency of rumen microflora.

Eight strains of rumen bacteria capable of degrading phloroglucinol (1,3,5-trihydroxybenzene) under anaerobic conditions were isolated from enrichment cultures of the bovine rumen microflora established in a prereduced medium containing 0.02 M phloroglucinol. Five of the strains were facultatively anaerobic Gram-positive streptococci which were identified as Streptococcus bovis. Three strains of obligately anaerobic Gram-positive cocci were assigned to the genus Coprococcus. Anaerobic cultures of the Streptococcus bovis strains in a 40% rumen fluid medium initially containing 0.02 M phloroglucinol degraded 50–80% of the substrate within 2 days, whereas cultures of the Coprococcus strains degraded more than 80% of the substrate under the same conditions. The Streptococcus bovis strains were incapable of degrading phloroglucinol in brain heart infusion or in the medium of de Man, Rogosa, and Sharpe (MRS broth) incubated aerobically.

Four sheep, fed a basal diet which included 44% cereal grain and 50% hay and which was supplemented with KNO3, were progressively acclimated to a KNO3 intake of 2.5 g∙kg body wt−1∙day−1. Nitrate and nitrite reducing activity in strained rumen fluid (SRF) collected 2 h after feeding showed maximum values of 45.3 μg N∙mL−1∙h−1 and 39.4 μg N∙mL−1∙h−1, respectively, at an intake of 1.5 g∙kg body wt−1∙day−1. The rate of nitrate reduction was threefold higher (P < 0.01), and that of nitrite reduction fivefold higher (P < 0.01), than in SRF from sheep not receiving KNO3. When the KNO3 supplement was withdrawn the reducing activities fell to their initial levels within 3 wk. In a second experiment, nitrate, nitrite and volatile fatty acids in SRF, and methemoglobin in peripheral blood, were estimated at 30-min intervals after feeding in four sheep, two of which received 1.5 g KNO3∙kg body wt−1∙day−1. In the animals fed KNO3 the peak concentration of nitrate in SRF (13.30 μg NO3−–N∙mL−1) was reached 30 min after feeding, and that of nitrite (2.90 μg NO2−–N∙mL−1) 60 min after feeding; the presence of nitrate in SRF was associated with an increase in the molar proportion of acetate (P < 0.01) and a decrease in the proportion of n-butyrate (P < 0.01). The blood methemoglobin concentration did not exceed 2% (wt/wt) of total hemoglobin at any sampling time. In these animals 30–35 g KNO3 cleared the rumen within 3 h after feeding with no symptoms of nitrate toxicity. Enumeration of rumen bacteria by a direct isolation procedure indicated that the proportion of nitrate reducers in SRF was threefold higher (P < 0.01) in acclimated animals than in control animals. It was concluded that safe acclimation of sheep to high levels of dietary nitrate involved an increase in the rates of nitrate and nitrite reduction in the rumen, a narrowing of the ratio of these activities, and an increase in the proportion of nitrate reducing rumen bacteria. The residual effect of nitrate on the fermentation following clearance from the rumen was short-lived. 1. A pepsin + pancreatin method was used to assess the digestibility of pure cultures of rumen bacteria and mixed bacteria prepared from rumen fluid.2. Individual species of Gram-negative rumen bacteria were highly digestible, whereas Gram-positive species, especially cocci, were more resistant to digestion.3. A similar difference was observed microscopically with mixed rumen bacteria, but the influence of the relative proportions of Gram-positive and Gram-negative bacteria on the digestibility of bacterial protein in rumen fluid was small.

The rumen contains abundant microorganisms that aid in the digestion of lignocellulosic feed and are associated with host phenotype traits. Cows with extremely high milk protein and fat percentages (HPF; n = 3) and low milk protein and fat percentages (LPF; n = 3) were selected from 4000 lactating Holstein cows under the same nutritional and management conditions. We found that the total concentration of volatile fatty acids, acetate, butyrate, and propionate in the rumen fluid was significantly higher in the HPF group than in the LPF group. Moreover, we identified 38 most abundant species displaying differential richness between the two groups, in which Prevotella accounted for 68.8% of the species, with the highest abundance in the HPF group. Functional annotation based on the Kyoto Encyclopedia of Gene and Genome (KEGG), evolutionary genealogy of genes: Non-supervised Orthologous Groups (eggNOG), and Carbohydrate-Active enzymes (CAZy) databases showed that the significantly more abundant species in the HPF group are enriched in carbohydrate, amino acid, pyruvate, insulin, and lipid metabolism and transportation. Furthermore, Spearman’s rank correlation analysis revealed that specific microbial taxa (mainly the Prevotella species and Neocallimastix californiae) are positively correlated with total volatile fatty acids (VFA). Collectively, we found that the HPF group was enriched with several Prevotella species related to the total VFA, acetate, and amino acid synthesis. Thereby, these fulfilled the host’s needs for energy, fat, and rumen microbial protein, which can be used for increased biosynthesis of milk fat and milk protein. Our findings provide novel information for elucidation of the regulatory mechanism of the rumen in the formation of milk composition.

Melilotus officinalis is known to contain several types of secondary metabolites. In contrast, the carotenoid composition of this medicinal plant has not been investigated, although it may also contribute to the biological activities of the drug, such as anti-inflammatory effects. Therefore, this study focuses on the isolation and identification of carotenoids from Meliloti herba and on the effect of isolated (all-E)-lutein 5,6-epoxide on primary sensory neurons and macrophages involved in nociception, as well as neurogenic and non-neurogenic inflammatory processes. The composition of the plant extracts was analyzed by high performance liquid chromatography (HPLC). The main carotenoid was isolated by column liquid chromatography (CLC) and identified by MS and NMR. The effect of water-soluble lutein 5,6-epoxide-RAMEB (randomly methylated-β-cyclodextrin) was investigated on Ca2+-influx in rat primary sensory neurons induced by the activation of the transient receptor potential ankyrin 1 receptor agonist to mustard-oil and on endotoxin-induced IL-1β release from isolated mouse peritoneal macrophages. (all-E)-Lutein 5,6-epoxide significantly decreased the percent of responsive primary sensory neurons compared to the vehicle-treated stimulated control. Furthermore, endotoxin-evoked IL-1β release from macrophages was significantly decreased by 100 µM lutein 5,6-epoxide compared to the vehicle-treated control. The water-soluble form of lutein 5,6-epoxide-RAMEB decreases the activation of primary sensory neurons and macrophages, which opens perspectives for its analgesic and anti-inflammatory applications.

Glutamate, succinate, propylene glycol, or glycerol were added to a basic concentrate at 3.3% of air-dry feed. Each concentrate was fed both ad libitum and in restricted amounts to four cows in early lactation. Dietary intake, milk yield and composition, molar proportions of rumen volatile fatty acids and blood glucose, ketones and plasma free fatty acids were used as criteria of effect of these supplements. Propylene glycol in the diet resulted in a lower intake of concentrate compared with glycerol (11.44 versus 14.30 kg/day) and significantly decreased (P < 0.05) rumen butyrate and plasma beta-hydroxybutyrate. Glutamate supplementation prevented the fall in milk fat content which occurred when the other three supplemented concentrates were fed ad libitum, and this effect may have been related to the constancy in the ratio of acetate to propionate in the rumen fluid.
