A host response to infection by Coxiella burneti was investigated. Infected yolk sacs were harvested from embryonated eggs and assayed for glycolytic activity. Assays of glycolytic enzymes included glucose isomerase, aldolase, phosphofructokinase, fructose-1,6-diphosphatase, glyceraldehyde-3-phosphate dehydrogenase, enolase, and pyruvate kinase. No significant differences in enzymatic activity between normal and infected tissues through the 12th day were observed. From the 13th day through the 16th day, the glycolytic activity of normal tissues decreased. Glycolytic activity of infected tissues did not decrease, but showed a gradual increase during this same time period. Embryos from infected eggs demonstrated a gradual decrease in total weight from the 12th day until death occurred on the 16th day.

BackgroundProlonged sedentary behaviour is associated with a higher risk of cardiometabolic diseases. This longitudinal study examined changes in daily total, prolonged (≥30 min) and highly prolonged (≥60 min) sedentary time across the transition to retirement by gender and occupational status.MethodsWe included 689 aging workers (mean (SD) age before retirement 63.2 (1.6) years, 85% women) from the Finnish Retirement and Aging Study (FIREA). Sedentary time was measured annually using a wrist-worn triaxial ActiGraph accelerometer before and after retirement with on average 3.4 (range 2–4) measurement points.ResultsWomen increased daily total sedentary time by 22 min (95% CI 13 to 31), prolonged sedentary time by 34 min (95% CI 27 to 42) and highly prolonged sedentary time by 15 min (95% CI 11 to 20) in the transition to retirement, and remained at the higher level of sedentary time years after retirement. The highest increase in total and prolonged sedentary time was observed among women retiring from manual occupations. Men had more total and prolonged sedentary time compared with women before and after retirement. Although no changes in men’s sedentary time were observed during the retirement transition, there was a gradual increase of 33 min (95% CI 6 to 60) in prolonged sedentary time from pre-retirement years to post-retirement years.ConclusionThe transition to retirement was accompanied by an abrupt increase in prolonged sedentary time in women but a more gradual increase in men. The retirement transition may be a suitable time period for interventions aiming to decrease sedentary behaviour.

THE MEASUREMENT of enzyme activity of serum as an indicator of disease has a long history in medicine. In the past, it has been the aim of the designers of these methods to make them as specific as possible for assay of an enzyme characteristic of a particular system or group of similar organs. Examples of these venerable tests are those for amylase, acid phosphatase, alkaline phosphatase and choline esterase in the serum. Warburg made the first departure from this specificity by demonstrating that the activity of triosephosphate dehydrogenase in the serum of animals with cancer was much greater than that of controls. This test was partially specific, for as Warburg had earlier shown, the glycolytic activity of tumors is much greater than that of normal tissues. The non-specific approach became extreme with the introduction of the measurement of the glutamic-oxalacetic transaminase reaction in the diagnosis of acute coronary disease.

Enhancement in the production of enzyme by utilizing different strains of microbe is one of the main prospects in biotechnology. In the present work, ethyl methane sulfonate (EMF) was selected as the chemical mutagen for inducing mutagenesis in fungi. It is a cheap method to induce random mutation as compared to other methods of recombinant technologies. Strain improvement was done by incubating the fungal spore suspension at variable concentrations of EMS i.e. 4% (v/v) and 10% (v/v) for the time period of 60, 90, and 120 min respectively. The set of control was treated with distilled water only. The fungal colonies were found to be maximum in control plate as compared to the EMF exposed plates. The number of fungal colonies was reduced as we raised the exposure time of EMF. Specific activity and the lipase activity of wild strain and hyperproducer were evaluated under the submerged (SmF) and solid-state fermentation (SSF). The wild strain denoted the 3.2 U/ml/min of enzymatic activity under SmF and 15.87 U/g/min of activity under SSF. In contrast, the best enzymatic activity was represented by S2St1 at 10% of EMS after the time period of 60 min i.e. 11.7 U/ml/min under SmF and 99.35 U/g/min under SSF after the time period of 72 hrs. Statistical analysis by using one-way ANOVA determined that the value of F calculated was lower than the F tabulated. So, there was a significant relation between the EMS percentage and time of exposure among the mutated strains. In conclusion, this soil fungal strain can be utilized to produce lipase enzyme for numerous industrial applications.

ABSTRACTAllosteric regulation is central to the role of the glycolytic enzyme pyruvate kinase M2 (PKM2) in cellular metabolism. Multiple activating and inhibitory allosteric ligands regulate PKM2 activity by controlling the equilibrium between high activity tetramers and low activity dimers and monomers. However, it remains elusive how allosteric inputs upon simultaneous binding of different ligands are integrated to regulate PKM2 activity. Here, we show that, in the presence of the allosteric inhibitor L-phenylalanine (Phe), the activator fructose 1,6-bisphosphate (FBP) can induce PKM2 tetramerisation, but fails to maximally increase enzymatic activity. Guided by a new computational framework we developed to identify residues that mediate FBP-induced allostery, we generated two PKM2 mutants, A327S and C358A, in which activation by FBP remains intact but cannot be attenuated by Phe. Our findings demonstrate a role for residues involved in FBP-induced allostery in enabling the integration of allosteric input from Phe and reveal a mechanism that underlies the co-ordinate regulation of PKM2 activity by multiple allosteric ligands.

The article provides a review of the literature on the effectiveness of glucocorticoids in viral infections, including the new coronavirus infection COVID-19. The results of our research of the dynamics of laboratory parameters in patients who recovered and those who died are presented. The average age of patients who received 8 mg of dexamethasone and recovered was less than that of the deceased. The average day on which the patients were hospitalized in the ICU and who received 8 mg of dexamethasone in both groups was the same (on average on day 7). It is noteworthy that in patients who recovered, the febrile temperature was more often recorded before hospitalization, while in those who died it was more often subfebrile. The temperature during hospitalization in all categories of patients was on average at subfebrile numbers. The respiratory rate on admission in all categories of patients did not differ significantly and on average was about 19/min (up to 20/min was in 50 % of patients who recovered and 58.3 % of those who died). Those who recovered were more likely to have a normal heart rate on admission, but tachycardia was less common than within those who died. In patients who subsequently died, lower saturation levels were more often recorded upon admission. In the first 3 days after hospitalization, in patients who recovered and received 8 mg of dexamethasone, leukocytosis and granulocytosis were recorded more often; there was an increased number of stab neutrophils and the number of patients with it. The patients who subsequently died more often developed leukopenia, more pronounced lymphopenia with an increased quantity of patients with it; they presented more pronounced thrombocytopenia (the number of patients with it did not differ from those who recovered), higher erythrocyte sedation rate. In patients who subsequently died, during the observation period, there was a gradual increase in the number of leukocytes, but a gradual decrease in the number of lymphocytes, the creatine phosphokinase level increased from the 7th–9th days of hospitalization; on days 4–6 of hospitalization, the lactate dehydrogenase level significantly increased with its subsequent decline to a level that was greater than this in patients who recovered. Initially. The patients who recovered had an increase in leukocytes with their subsequent gradual decrease, a gradual increase in the level of lymphocytes, a decrease in the level of creatine phosphokinase, lactate dehydrogenase. In all categories of patients, a gradual decrease in the number of stab neutrophils was observed over time, a gradual increase in the number of platelets was also observed over time, but in those who recovered their level was slightly higher; in both groups, an increase in the urea level was observed over time, but in those who died its level from day 7 and later was significantly higher than in those who recovered; in patients in both groups, an increase in the level of creatinine was observed over time, but in those who died, its level from day 7 and later was significantly higher than in those who recovered; both groups showed a decrease in C-reactive protein over time, but those who died from the very beginning of their hospital stay and during all follow-up periods had higher levels than those who recovered.

Based on methodologies currently used in commercial hatcheries, four thermic treatments were compared to induce the gametes expulsion of Argopecten purpuratus. The treatments evaluated included: 1) gradual increase in temperature, 2) gradual decrease, 3) 5 °C thermal-shock, 4) 10 °C thermal-shock and 5) control at constant temperature 16 °C. After induction, the number of fertilized oocytes in each treatment was estimated. The results show that a gradual increase in temperature between 16 and 26 °C causes a significantly higher proportion of fertilized oocytes compared to other treatments.

To describe the disease cycle of the root pathogen Aphanomyces euteiches, enzymatic activity in the mycelium was compared with the development of oospores in pea roots. Plants were inoculated with two zoospore concentrations to achieve different disease levels. Hyphae were stained for fungal alkaline phosphatase activity in the roots. Additionally, enzyme activity was measured after electrophoresis of an A. euteiches-specific glucose-6-phosphate isozyme. Development of oospores in the roots was measured after staining the oospores with trypan blue. In plants inoculated with the higher zoospore concentration, the enzymatic activity of the pathogen mycelium peaked 10 to 14 days after inoculation, when oospore formation was initiated. Oospore formation was associated with a gradual increase in disease symptoms. At the last harvest, plants inoculated with the higher zoospore concentration had died. In these plants, oospores were found in 90% of the root length, while the enzymatic activity of the mycelium was low. This suggests that the pathogen mycelium is only active on living plants and does not grow saprophytically on dead plant material.

The differences in properties of pyruvate kinase (EC 2.1.7.40) from normal tissues and animal or human tumors are described and their significance for various metabolic abnormalities is reviewed. The tumor variant gamma3 from M2 isoenzyme of pyruvate kinase sensitive to L-cysteine inhibition, when over-expressed, can be used as a marker of tumorigenic transformation. It seems that this variant represents a tumor-specific oncoprotein, involved in a novel metabolic strategy of energy generation during increased cell proliferation.
