Effect of disodiumN,N-dialanyl protoporphyrinate, diarginine protoporphyrinate and diarginineN,N-dialanyl protoporphyrinate on respiratory burst of human neutrophilsin vitro

The effect of three amino acid protoporphyrin IX (PPIX) derivatives evaluated as sensitizers in photodynamic therapy — disodium N,N -dialanyl protoporphyrinate ( PP(Ala)2Na2), diarginine protoporphyrinate (PPArg2) and diarginine N,N -dialanyl protoporphyrinate (PP(Ala)2Arg2) – non-irradiated and pre-irradiated with UV-A, on respiratory burst of non-stimulated and opsonized zymosan stimulated neutrophils was studied. A potential synergistic effect of diazepam (7-chloro-1-methyl-5-phenyl-1,3-dihydro-2H-1,4-benzodiazepin-2-one) was also examined. PP(Ala)2Na2showed strong pro-oxidant effect towards non-stimulated neutrophils, while PPArg2and PP(Ala)2Arg2revealed no significant effect. In the case of stimulated neutrophils all studied porphyrins showed antioxidant effect, although for PP(Ala)2Na2this effect was significantly weaker than that of PPArg2and PP(Ala)2Arg2. After pre-irradiation with UV-A (λ = 365 nm, fluence 2.0 J.cm-2, fluence rate 6.7 mW.cm-2) the antioxidant activity of all studied sensitizers towards non-stimulated granulocytes did not change significantly when compared to effects of non-irradiated porphyrins, while in the case of stimulated cells, only PPArg2caused significant decrease of respiratory burst. Non-irradiated diazepam showed significant antioxidant effect and enhanced antioxidant effect of all studied porphyrins towards stimulated neutrophils, while after UV-A pre-irradiation it revealed no significant antioxidant effect on non-stimulated and stimulated neutrophils, both alone and in combination with porphyrin sensitizers.

Propolis, a precious bee product has been widely used for pharmaceutical purposes for centuries. Chemical components, properties, and activities are differed based on geographic origins, respective plant sources, and bee species. In our research, Mongolian propolis from 2 different places was examined for chemical content and antioxidant effect. The samples were collected from Northern and Central parts of Mongolia and were examined for a total of 8 contents. Crude Mongolian propolis sample from central part of Mongolia displayed higher results in every aspect with total moisture of 2.1%; ash 1.7%; fat 67.8, 69.3%; and 1.05% of protein. The free amino acid in 70% EtOH was higher in sample from northern part with 1.21µg/mg and the total amino acid was quantified by HPLC and highest concentration detected in methionine 1.90mg/g, and 2.62 mg/g. Totally 3 alkaloids and 2 phenolic were detected by TLC and high antioxidant activity was presented in DPPH radical scavenging test. The results suggest that the best solvent for propolis samples is EtOH 70% solution. The test propolis samples contain different types of alkaloids and phenolic, have high amount of methionine and has high antioxidant effect.

Aim: The objective of this study was to investigate the possible synergy between doxycycline and photodynamic therapy against Helicobacter pylori and to evaluate the possible side effects on adenocarcinoma gastric cells with and without protoporphyrin IX. Materials & methods: Three H. pylori strains (ATCC 700392, 43504 and 49503) were grown on solid medium either with, or without, doxycycline at subinhibitory concentrations, and irradiated for 10, 20 and 30 minutes with a 400 nm-peaked light source. The phototoxicity tests on AGS cells were evaluated by MTT assay. Results: The photodynamic therapy and doxycycline combination showed an antibacterial synergistic effect with no significant toxicities. Conclusion: The synergistic treatment could be considered as an interesting therapeutic option.
