In this study, the antioxidant mechanism of Astragalus membranaceus fermented by Lactiplantibacillusplantarum MG5276 (MG5276F-AM) was evaluated in HepG2 cells and in an animal model. HPLC analysis was performed to confirm the bioconversion of the bioactive compounds in A. membranaceus by fermentation. Calycosin and formononetin, which were not detected before fermentation (NF-AM), were detected after fermentation (MG5276F-AM), and its glycoside was not observed in MG5276F-AM. In HepG2 cells, MG5276F-AM alleviated H2O2-induced oxidative stress by mediating lipid peroxidation and glutathione levels, and upregulated antioxidant enzymes including catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx). In the tBHP-injected mouse model, administration of MG5276F-AM reduced hepatic aspartate transaminase, alanine transaminase, and lipid peroxidation. MG5276F-AM also modulated antioxidant enzymes as well as HepG2 cells. Thus, fermentation of A. membranaceus with L. plantarum MG5276 elevated the isoflavonoid aglycone by hydrolysis of its glycosides, and this bioconversion enhanced antioxidant activity both in vitro and in vivo.

The escalating problem of obesity has become a cause of great concern in the world today as it leads to adverse effects on human health, including cardiovascular diseases, cancer etc. The major causes of obesity may be attributed to sedentary lifestyle and bad food habits. Conventional modalities to tackle obesity are not free from side-effects. Urgency of a novel, nontoxic means needs to be developed to control obesity. In this study we aim to screen the phytochemical compounds of Camellia Sinensis and evaluate its antiobesity and antioxidant effects. The methanolic extract of Camellia Sinensis was analyzed for its phytochemical screening and assayed for its in-vitro activity against pancreatic lipase, its antioxidant potential and quantitative estimation of flavonoids and phenolics were done. The methanolic extract of Camellia Sinensis strongly inhibited pancreatic lipase by 63% and it also possesses a strong antioxidant effect and there was a significant positive correlation between phenolics, flavonoids and with alkaloid contents. From these results, it could be concluded that methanolic extracts of Camellia Sinensis possesses antipancreatic lipase compounds. It also possesses antioxidant effect. It is suggested that the phytochemical compounds from there plants may be applied for the prevention and treatment of obesity or hyperlipidemia. The chemical composition of the Cynophalla flexuosa hydroethanolic extract (CFHEE) was identified and its antioxidant, antifungal and antipleomorphic activities against C. albicans and C. tropicalis strains were evaluated. Phytochemical prospecting evidenced polyphenolic and triterpenoid compounds while UPLC-MS analysis revealed the presence of Isopropyl/n-propyl-GLS; Methylpropyl-GLS/butyl-GLS; Methylbutyl-GLS; Quercetin O-di-hexoside; Quercetin-pentosyl-hexoside; Rutin; Quercetin O-glucoside; Kaempferol O-rhamnosyl-hexoside; Kaempferol O-pentosyl-hexoside and Lariciresinol hexoside. The CFHEE inhibited the effect of the DPPH● free radical, both when evaluated individually and in combination with the antifungal fluconazole. When associated with 256 μg/mL fluconazole, the extract, at concentrations from 128 μg/mL, reduced the effect of the DPPH● free radical with values ranging from 37.7% to 95.8%. The extract’s antifungal effect was considered clinically irrelevant and its combination with the antifungal triggered an antagonistic effect against the two strains, thus indicating the popular use of bravo beans in the form of teas or infusions should not be combined with the intake of the drug Fluconazole as this may lead to a reduction of its clinical effect. The extract, however, inhibited morphological changes (pleomorphism) in Candida species, preventing the development of hyphae. The CFHEE possesses an antioxidant effect and a potential pharmacological activity for the inhibition of one of the Candida spp. virulence factors.

Antioxidants are substances counteracting the effects of free radicals. Antioxidants are formed in several forms, encompassing vitamins, minerals, and phytochemicals. Genjer or yellow velvet leaves (Limnocharis Flava) is an aquatic plant potentially beneficial alternative to natural antioxidants. This study aims to measure the antioxidant effect from the yellow velvetleaf ethanol extract on malondialdehyde (MDA) activities in livers of diabetic rats (Rattus novergicus) induced by alloxan in three different doses. The tested animals were randomly divided into five groups comprising six white rats previously induced with alloxan by intra-peritoneal injection. Group 1 was the negative control provided 0.5% CMC Na while group 2 was a positive control administered 0.45 mg glibenclamide. On the other hand, groups 3, 4, and 5 were the experimental groups that provided the extract of yellow velvetleaf as many as 32.5 mg/kg body weight (BW) ethanol, 65 mg/kg BW ethanol extract, and 130 mg/kg BW, respectively. Furthermore, the extracts were administered once on the 7th, 14th, and 21st days; the parameters observed were MDA activities in the rats’ livers. The Thiobarbituric Acid Reactive Substances (TBARs) method was then employed to investigate these actions. The one-way ANOVA was administered to examine the acquired data from the examination, followed by a post hoc test. The average points of the MDA level from group 1 to group 5 were 10.78 0.33, 3.71 + 0.19, 7.25 + 0.26, 5.76 + 0.25, and 4.01 + 0.22, correspondingly, according to the results. The one-way ANOVA test revealed that MDA levels in each test group were significantly different (p 0.05). Based on these findings, it may be concluded that feeding ethanol extract from yellow velvet leaves to diabetic rats reduces MDA levels in their livers, therefore preserving cells from harm.

Introduction. The effect of laser irradiation on the body can lead to local tissue damage, which will be accompanied by changes in biochemical processes that occur in the kidneys. The direction of these changes is completely unclear, so the study of free radical oxidation of lipids and proteins as the main markers of oxidative stress in the body and their correction by ω-3 polyunsaturated fatty acids (PUFA) remains relevant. Aim.  To evaluate the intensity of lipid peroxidation and oxidative modification of proteins in the postnuclear fraction of rat kidneys under the action of laser irradiation and additional administration of ω-3 PUFA. Methods. The studies used white outbred rats, which were irradiated daily for 4 minutes with a laser diode with a wavelength of 650 nm, power of 50 mW in the abdominal cavity at a distance of 10 cm from the skin surface. The state of lipid peroxidation (LPO) was determined by the level of primary, secondary and final products. The intensity of oxidative modification of proteins (OMP) was evaluated by the level of carbonyl derivatives and protein SH-groups. To correct the prooxidant state, the animals were additionally injected with ω-3 PUFA. Results. Low-intensity laser irradiation has a destructive effect on the cell membranes of the kidneys, which is expressed by an increase in primary, secondary and final products of LPO in phospholipid extracts and an increase in the intensity of OMP. The introduction of ω-3 PUFAs reduces free radical processes in the kidneys of irradiated rats, but this effect depends on the scheme of their introduction. ω-3 PUFAs do not show antioxidant effect under the conditions of their introduction after laser irradiation. Daily administration of ω-3 PUFA two hours before irradiation shows a slight antioxidant effect only in the initial stages of irradiation. Preliminary seven-day administration of ω-3 PUFA before laser irradiation is the most effective, as it helps to reduce free radical processes. Conclusion. It is established that ω-3 PUFAs are able to have a corrective effect on the action of low-intensity laser irradiation, which depends on the scheme of their introduction. The highest antioxidant effect is observed in groups of animals to which ω-3 PUFA was previously administered before the action of laser irradiation.

Topicality. Modern manufacturers of foodstuff and beverages are faced with the task of creating the new generation nutritive – sanative and functional, including drinks, that can have a positive impact on consumers’ health condition. Functional beverages are intended for systematic usage in the diet compound for all healthy population age groups; they are able to reduce the risk of diseases development, caused by nutrition, maintain and improve health state due to the presence of physiologically functional ingredients in their complement (vitamins, phenolic compounds, dietary fibers, carotenoids, organic acids, minerals). Aim and methods. The aim of this article is to substantiate the rowan juice choice for the functional meaning drink creation, to determine its composition and physico-chemical indicators, the selection of effective formula components with antioxidant effect, and the working of the non-alcoholic drink with sanative qualities technology. In this work standard organoleptic, physico-chemical and calculated research methods are used. Results. It was proved that the rowan juice should be used as the basis for the juice-containing beverage of functional meaning. For maximal juice extraction, it is necessary to pre-freeze the raw material, followed by its grinding and enzymatic preparation machining. Green tea and sage extracts, which are sources of antioxidant effect substances, were chosen as a supplement to the rowan juice. Discussion and conclusions. It is established that the non-alcoholic juice-containing beverage belongs to the category of functional foodstuff, as during its using the level of daily needs providing of ascorbic acid, β-carotene, pectin, bioflavonoids, catechins is within 10...50%. The scientific novelty of this research lies in the technology development of the juice-containing functional beverage on the basis of rowan juice with vegetative extracts use. The practical significance of the received results is in their usage at enterprises, producing non-alcoholic beverages, in restaurant industry and sanatorium-resort catering establishments.

Objective: This study aimed to investigate the effects of the antioxidant potential of Arrabideae chica (crajiru) extract on oxidative stress in diabetic rats. Methods: Adult Wistar rats (Rattus norvegicus), weighing 238±12g were divided into three groups of six rats each: CN normal untreated control; DIAB+NS diabetic rats treated with normal saline; and diabetic rats treated with crajiru extract, DIAB+CR. The CN and DIAB+NS groups (control groups) received normal saline solution (NS) orally (gavage); rats in the DIAB+CR group received crajiru extract (300 mg/kg) once a day by gavage for 6 weeks. Measurements of urea and creatinine in serum, and kidney tissue catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx) were performed. The variables were assessed using the Tukey test, significance p<0.05. Results: All animals survived the experiments. In the CN group, compared with the DIAB+NS group, there was significant difference between the levels of glycemia on the second day of dosing and on the 10th day (p<0.05). No difference was observed on glycemia comparing the 2th and 10th day on the rats of group C+NS (p>0.05). Diabetic animals from DIAB+CR group had a significant reduction in glycemia on 10th day of treatment, comparing the 2nd day (p<0.05). There was a significant reduction in glycemia in the DIAB+CR group, comparing with the DIAB+NS group (p<0.05). There was an increase in urea and creatinine levels in rats DIAB+SN when compared to controls, C+SN (p<0.001). Rats from the DIAB+CR group had a significant reduction in urea and creatinine, compared to the DIAB+NS group (p<0.001). There were no significant differences in urea and creatinine comparing the C+NS and DIAB+CR groups. The rats from the DIAB+NS group had significantly lower levels of CAT, GSH-px and SOD when compared to the normal control rats (p<0.001). In animals from the DIAB+SN group, the levels of these antioxidant enzymes were significantly reduced (p<0.001). The treatment of diabetics with crajiru extract caused a significant increase (p<0.001) in the levels of CAT, GSH-px and SOD, when compared to rats in the BIAB+SN group. Conclusion: The data of the present study confirms that the crajiru extract positively influenced the control of hyperglycemia in diabetic rats. More research is needed to provide a better understanding of the mechanisms of diabetes treatment using crajiru extract and its flavonoids.

Mulberry (Morus alba L.), and above all the extract from the leaves of this plant, is a natural medicine that has been used in traditional medicine for hundreds of years. Mulberry leaves contains polyphenol compounds: flavonoids, coumarins, numerous phenolic acids, as well as terpenes and steroids. The antioxidant effect of these compounds may be beneficial to the fat fraction of meat products, thereby increasing their functional qualities. The aim of the study was to evaluate the effectiveness of the use of mulberry water leaf extract, as an additive limiting adverse fat changes and affecting the functionality in model liver pâtés. Pork pâtés were prepared by replacing 20% of animal fat with rapeseed oil (RO), and water extract of mulberry leaves was added in the proportion of 0.2%, 0.6% and 1.0%. It has been shown that the addition of mulberry leaf extract delayed the appearance of primary and secondary fat oxidation products. The most effective antioxidant effect during 15-day storage was observed in the sample with the addition of 0.6% and 1.0% water mulberry leaf extract. These samples also showed inhibiting activity against angiotensin-converting enzymes and cholinesterase’s. During storage, the tested pâtés had a high sensory quality with unchanged microbiological quality. Mulberry leaf extract can be an interesting addition to the production of fat meat products, delaying adverse changes in the lipid fraction and increasing the functionality of products.
