Effects of food intake and cholecystokinin on plasma trypsinogen levels in dogs

Concentrations of plasma immunoreactive anionic and cationic trypsin(ogen) were monitored in unanesthetized dogs to investigate diurnal variation, response to food intake, and effects of cholecystokinin octapeptide (CCK-8) plus secretin administration. Identical meals were consumed at the beginning and end of a 24-h period. Plasma levels of both trypsin(ogen)s increased significantly within 30 min of feeding. Second-meal responses were much larger than those following the first meal. Plasma cationic trypsin(ogen) was also elevated during the normal feeding period when food was withheld and dogs in adjacent cages were fed. Small, irregular variations in plasma trypsin(ogen)s occurred during the interdigestive period when blood was sampled every 2 h. CCK-8 plus secretin, in doses shown previously to stimulate exocrine secretion of enzymes submaximally, increased plasma levels of both trypsin(ogen)s dose dependently. Most (greater than 80%) of the immunoreactive cationic trypsin(ogen) in plasma before and after feeding and following CCK-8 was in proenzyme form. We conclude that plasma levels of pancreatic trypsinogens are increased by food intake and low doses of CCK-8. Furthermore, cephalic mechanisms appear to play an important role in mediating the response to food intake.

ABSTRACT Marked diurnal variations were found in plasma growth hormone (GH), insulin, acetoacetate (AcAc) and sugar in two herds (U and A) of dairy cows kept at two different levels of feeding. Seven animals from each herd were tested. The main diurnal variations were related to food intake, a significant increase in plasma insulin, and a significant decrease in plasma sugar being found. The acetoacetate level rose significantly during feeding in herd U (moderately underfed), while no significant increase was found in herd A (adequately fed). Plasma growth hormone was found to decrease (P < 0.01) only during feeding in herd U, while no change in the GH level could be detected in herd A. The mean level of GH in herd U was found to be twice the value found in herd A. There were no significant differences between the herds in plasma insulin and sugar. Significant differences in plasma levels of GH, insulin and sugar were found between animals when analysed within the herds. Variations in the levels of insulin and acetoacetate were very small during the night. This is contrary to GH, which shows the least variation during food intake. The correlation coefficient between the plasma components was low, although in many cases significant.

ABSTRACT Intraperitoneal injection of 5 μg cholecystokinin octapeptide (CCK-8) into male rats deprived of food for 48 h produced a transient (less than 15 min) increase in plasma levels of CCK-8 but suppressed food intake for an extended period (45 min). Plasma concentrations of CCK-8 after i.p. injection of CCK-8 were raised to levels which were fairly comparable to those after feeding. Intracerebroventricular (i.c.v.) injection of the CCK antagonist proglumide (100 μg) reversed the effect of CCK-8 on food intake, while i.p. injection of proglumide (100 μg) did not have this effect. Feeding increased the plasma concentrations of somatostatin and gastrin but not of oxytocin, and somatostatin and oxytocin but not gastrin were released in response to i.p. injection of CCK-8. However, neither somatostatin nor oxytocin affected food intake, and their release in response to CCK-8 was unaffected by i.c.v. injection of proglumide. These results support the suggestion that CCK-8 is a physiological 'satiety' peptide, which can affect food intake in rats by mechanisms involving both peripheral and central CCK receptors. Feeding responses to continuous intravenous administration of graded doses of the COOH-terminal octapeptide of cholecystokinin (CCK-8) and pancreatic glucagon, alone and in combination, were determined in dogs fasted 4 h. Low doses of glucagon (50, 500, 5,000, 6,000 pmol.kg-1.h-1) had no effect on food intake, whereas higher doses (12 and 24 nmol.kg-1.h-1) depressed intake by 50-60%. Of the CCK-8 doses administered (50 and 400 pmol.kg-1.h-1), food intake was depressed only at the higher dose (53%). This effect was blocked by glucagon (50-5,000 pmol.kg-1.h-1). Simultaneous administration of 50 or 500 pmol.kg-1.h-1 of glucagon and 50 pmol.kg-1.h-1 of CCK-8, doses currently thought to produce plasma peptide levels similar to those occurring postprandially in dogs, had no effect on food intake. These results suggest that plasma levels of CCK and glucagon after a meal are not sufficient alone or in combination to produce satiety.

AbstractThe diurnal variation of the magnetic field cannot be predicted or modeled and for that reason, it is monitored during the magnetic surveys, usually by a stationary magnetometer. However, marine surveys have a practical issue with diurnal monitoring, owing to the distance between the survey, stationary magnetometers, and magnetic observatories. This work aims to verify the use of nearby magnetic observatories to estimate the diurnal variation correction in different marine surveys and evaluate its effectiveness. In this study, we selected surveys at the continental shelf near Santos city (Survey 1), continental slope next to the first survey location (Survey 2), continental shelf near Ubatuba city (Survey 3), and Mamanguá ria in the Paraty city (Survey 4), all southeast to the Brazilian coast. The crossing points were implemented to compare the magnetic field values at different times and days at the same measurement point, before and after the correction. Afterwards, we measure the Pearson’s Correlation of the raw data and the diurnal corrected data in all crossing points of each survey which showed an improvement after correction by the value approximating to 1, which indicates a very well correlation. The Ubatuba and Mamanguá surveys allowed comparing the observatory correction results with the base magnetometer results that were rather similar. Our analyses indicate a satisfactory diurnal correction using the observatory data and the crossing points approach, which can be used for every marine magnetometric survey worldwide placed near the coast (< 280 km) that do not have a stationary magnetometer available.

ABSTRACT Plasma levels of testosterone (T) and 5α-dihydrotestosterone (DHT) were determined by radioimmunoassay in 29 patients with benign prostatic hypertrophy (BPH) and in 56 control men of various ages. No significant difference was found in T, DHT nor DHT/T ratio between BPH and control subjects of similar age. Plasma DHT was higher in the prostatic than in the peripheral veins in 8/9 patients with BPH during laparotomy, indicating a prostatic secretion of DHT. No difference in the mean T nor the mean DHT was found in peripheral plasma before and after adenomectomy.

Objective:Acute stress in patients with rheumatoid arthritis (RA) should stimulate a strong stress response. After cryotherapy, we expected to observe an increase of hormones of the adrenal gland and the sympathetic nervous system.Methods:A total of 55 patients with RA were recruited for whole-body cryotherapy at −110°C and −60°C, and local cold therapy between −20°C and −30°C for 7 days. We measured plasma levels of steroid hormones, neuropeptide Y (sympathetic marker), and interleukin (IL)6 daily before and after cryotherapy.Results:In both therapy groups with/without glucocorticoids (GC), hormone and IL6 levels at baseline and 5 h after cold stress did not change over 7 days of cryotherapy. In patients without GC, plasma levels of cortisol and androstenedione were highest after −110°C cold stress followed by −60°C or local cold stress. The opposite was found in patients under GC therapy, in whom, unexpectedly, −110°C cold stress elicited the smallest responses. In patients without GC, adrenal cortisol production increased relative to other adrenal steroids, and again the opposite was seen under GC therapy with a loss of cortisol and an increase of dehydroepiandrosterone. Importantly, there was no sympathetic stress response in both groups. Patients without GC and −110°C cold stress demonstrated higher plasma IL6 compared to the other treatment groups (not observed under GC), but they showed the best clinical response.Conclusions:We detected an inadequate stress response in patients with GC. It is further shown that the sympathetic stress response was inadequate in patients with/without GC. Paradoxically, plasma levels of IL6 increased under strong cold stress in patients without GC. These findings confirm dysfunctional stress axes in RA.

The effect of cold exposure (0–3 °C vs. 18–21 °C) on pancreatic exocrine secretion was investigated in sheep with chronically implanted bile duct catheters. At comparable feeding levels, cold exposure enhanced pancreatic juice flow by more than 50%, but its protein content and enzymatic activity were reduced. Only in cold-exposed sheep with increased food intake was there an increased daily output of pancreatic protein and enzymes. Key words: Pancreatic juice, protein, enzymes, cold exposure.

This study aimed to investigate the gender difference in the manifestation of physical stress in a strenuous military training on Amazon jungle, using alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatine kinase (CK) and lactate dehydrogenase (LDH) markers, measured before and after an adaptation to jungle training. The sample consisted of 49 military volunteers, 35 male and 14 female, recently moved to the Amazon region. All plasma levels rose after the training. Serum ALT (male and female) and AST (male and female), although borderline, remained within normal limits. Already plasma levels of CK (both male and female) and LDH (male and female) largely exceeded the normal range. The average of all markers listed in female gender remained below the levels of the male gender. However, significant differences in biomarkers ALT, AST and CK between genders were found. The study points out that, in a jungle environment, biometric markers ALT, AST, CK and LDH are efficient for monitoring chronic physical stress in both genders, when used in combination. The influence of the weather on the occurrence of physical stress in unacclimated people of both genders, and the lower responses in the levels of ALT, AST, LDH and CK in females were discussed basing on the scientific literature.

To examine the mechanism of the satiety-producing effect of cholecystokinin (CCK) in the central nervous system, we compared the potency of intraperitoneally (ip) or intracerebroventricularly (icv) administered CCK-8 and its analogues on food intake in fasted mice. The icv administration of a small dose of CCK-8 (0.03 nmol/brain) or of Suc-(Thr28, Leu29, MePhe33)-CCK-7 (0.001 nmol/brain) suppressed food intake for 20 min, whereas CCK-8 (1 nmol/kg, which is equivalent to 0.03 nmol/brain) or Suc-(Thr28, Leu29, MePhe33)-CCK-7 (1 nmol/kg) had satiety effect after ip administration. Dose-response studies indicated the following rank order of potency: Suc-CCK-7 > or = Suc-(Thr28, Leu29, MePhe33)-CCK-7 > or = CCK-8 > or = (Nle28,31)-CCK-8 >> desulfated CCK-8 = CCK-4 = 0 in the case of ip administration and Suc-(Thr28, Leu29, MePhe33)-CCK-7 >> Suc-CCK-7 > or = CCK-8 > or = (Nle28,31)-CCK-8 >> desulfated CCK-8 = CCK-4 = 0 in the case of icv administration. The selective CCK-A receptor antagonist MK-329 reversed the inhibitory effect of the centrally as well as peripherally administered CCK-8, or of Suc-(Thr28, Leu29, MePhe33)-CCK-7, whereas the selective CCK-B receptor antagonist L-365260 did not. The icv administered CCK-8 did not appear in the peripheral circulation. These findings suggest the participation of CCK-A receptors in the brain in mediating the satiety effect of CCK and the difference in CCK-A receptors in the brain and peripheral tissues.
