An experimental dentifrice containing nano-silver fluoride (NSF) and a sodium fluoride (NaF) toothpaste were testedin vitro, againstS. mutans, to evaluate the minimal inhibitory concentration (MIC), minimal bactericidal concentration (MBC), antiadherence, antiacid, enamel microhardness, and OCT. The microdilution technique was used to determine the MIC and MBC. Fragments of deciduous enamel were treated with dentifrice slurries, containing bacterial suspension and PBS-treated saliva. The quantification of the microorganisms that adhered to the enamel was determined after 24 hours of incubation, and media pH readings were performed after 2 hours and 24 hours. Deciduous teeth were evaluated for microhardness and OCT during 14 days of pH cycling. Data were statistically analyzed using Student’st-test, Mann–WhitneyUtest, ANOVA, and Tukey tests at 5% of significance. Dentifrices containing NSF presented a lower MIC and higher statistically significant results compared to NaF dentifrices with respect to preventing bacterial adhesion and pH decreases. NSF and NaF dentifrices showed the same ability to avoid enamel demineralization corroborated by the OCT images. The NSF formulation had a better antibacterial effect compared to NaF dentifrices and similar action on the demineralization of enamel indicating their potential effectiveness to prevent caries.

AbstractBackgroundNanosilver fluoride (NSF) was developed as an alternative in the prevention of dental caries.PurposeThe aim of this study was to test the remineralizing action of NSF on incipient enamel caries and its antimicrobial action on the acid production and adhesion of Streptococcus mutans.MethodsDeciduous enamel fragments were treated with sodium fluoride (NaF), NSF and deionized water. Microhardness, fluorescence spectroscopy and optical coherence tomography imaging were performed on each specimen before chemical caries induction, after caries induction and after 14 days of pH cycling. The treated enamel fragments were also placed into test tubes containing bacterial suspension and saliva. The pH readings and quantification of the adhered microorganisms to the dental enamel were determined. Analysis of variance, Kruskal-Wallis, Mann-Whitney, Tukey and mixed linear regression model were applied.ResultsNSF and NaF were effective in enamel remineralization, with a statistically significant difference (p<0.001) to deionized water, and they had no statistically significant difference between themselves (p>0.005). NSF had greater effectiveness compared to NaF in preventing decreases of pH and adhesion of S. mutans to the enamel surface, with statistically significant (p<0.001) differences.ConclusionNSF may be more effective than conventional fluorides in treating incipient caries lesions due to its remineralization and antibacterial actions.

ABSTRACT: Sausages are highly susceptible to microbial spoilage. Lactic acid bacteria (LAB) is the main group of spoilage bacteria in vacuum packed cooked sausages. To control microbial growth natural antimicrobials have been used as food preservatives. The aim of this study was to identify strains of lactic acid bacteria isolated from spoiled commercial Calabresa sausages and use them in an in vitro challenge with the natural antimicrobials, nisin (NI) and ε-poly-L-lysine (ε-PL). Mass spectrometry identification of LAB isolated from sausages using MALDI-TOF revealed a predominance of L. plantarum in the LAB population. RAPD-PCR of L. plantarum strains showed four different genetic profiles. Minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of NI and ε-PL, alone and in combination, against a pool of different profiles L. plantarum were determined. MIC of NI and ε-PL were 0.468 mg/ L and 75 mg/ L; respectively, whereas MBC of NI and ε-PL were 12.48 mg/L and 150 mg/L, respectively. The combined effect of NI and ε-PL was determined using concentrations at 1/4 and 1/8 of individual MICs. Synergistic effect was confirmed at both concentrations showing a fractional inhibitory concentration index of 0.5 and 0.2, respectively. The combination of NI and ε-PL at a small concentration of 0.05 mg/L and 9.375 mg/L, respectively, showed inhibitory effect towards spoilage L. plantarum Results show the potential of the combined use of NI and ε-PL to control sausage spoilage-associated with lactobacilli.

Objective: This study aimed to evaluate the effect of ferrous sulfate (FS) on demineralized and non-demineralized human deciduous teeth. Additionally, it was evaluated the penetration extent of FS and its remineralizing effect on the enamel of deciduous teeth using Polarized Light Microscopy (PLM). Method: The sample comprised 44 human deciduous teeth. The 44 crowns were divided randomly into four groups: group A (FS after demineralization), group B (FS without demineralization), group C (only demineralization), and group D (control group). FS at 0.45 mol/L-1 was used daily (15 days) and demineralization was done by pH cycling (7 days). Then, three longitudinal slices of the crowns were photographed using PLM. The degree of penetration of the lesion or stain was measured in micrometers, as well as the distance between the external enamel surface and the core of lesion. Results: Group A showed a dark stain on the outer surface of enamel larger than the group B. It is suggested, a remineralizing effect when comparing groups, A and C. The mean depth and standard deviation for groups A, B, and C were 4.27µm (±1.49), 3.72 µm (±1.68) and 5.00 µm (±1.84), respectively. No dark stains were observed in group D. Conclusion: FS stained the demineralized and non-demineralized human deciduous teeth. However, dark stains in the non-demineralized teeth were smaller or absent, than in the demineralized teeth. Therefore, FS may have a protective effect against demineralization.

Background: Methicillin-resistant Staphylococcus aureus (MRSA) causes nosocomial infections, and it has been considered as a worldwide epidemic. The medical system seeks new strategies to fight against MRSA that do not generate resistant strains to antibiotics. Homeopathy has been explored as one of these new strategies, which may play a pivotal role. In this context, we conducted studies on the action of homeopathy on growth of MRSA bacteria in vitro. The results showed a decrease in growth of bacterial strains with homeopathic dilutions of Belladonna and the S. aureus nosode. Now we have proposed to evaluate the minimum inhibitory concentration (MIC) of the antibiotic methicillin or oxacillin on S. aureus MRSA, previously incubated with the homeopathic dilutions of Belladonna or S. aureus nosode. Methods: The Clinical and Laboratory Standards Institute (CLSI 2014) standards were followed according to the determination of the minimum inhibitory concentration (MIC). In 5 mL of cation adjusted Mueller Hinton (CAMH) broth, it was added 420 µl of 30% alcohol or Belladonna and S. aureus’ nosode in the dilutions 6cH, 12cH and 30cH. Then a 20µl of bacterial suspension of MRSA was added to 0.5 McFarland range and diluted to 1/10. The tubes were incubated in an oven at 37⁰C for three hours. The plates were previously prepared with 50µl per well of serial dilutions of the antibiotic oxacillin in concentrations of 128 µg/mL to 0.5 µg/mL in CAMH broth. Then it was added 50 µl per well of bacterial cultures. The plate was incubated in an oven at 37⁰C for 24 hours and the bacterial growth measured in a spectrophotometer 600nm. The point of the MIC of oxacillin for S. aureus is 4 µg/mL, according to CLSI 2014 criteria. Results: We did not observe the total inhibition of bacterial growth when incubated with the homeopathic medicine and oxacillin. In evaluation of the spectrophotometer culture, we observed significant changes in the growth, compared to the control (30% alcohol). Cultures treated with Belladonna 6cH and the antibiotic in the dilution 4 µg/mL showed a decrease of 40% of the growth, while in the 30cH the drop was of 75%. Cultures treated with the S. aureus nosode 30cH and the antibiotic at 4 µg/mL dilution, showed a decrease of 60% in bacterial growth in vitro. Conclusion: The results suggest that bacterial cultures the S. aureus (MRSA) incubated with the homeopathic medicines would be more susceptible to oxacillin’s antimicrobial action.

Streptococcus mutans (S. mutans) is one of the cariogenic microorganisms. The restorative materials which harbor a biofilm with high number of S. mutans can accelerate the occurrence of dental caries. The purpose of this study was to evaluate the adherence of S. mutans to nanoceramic and nanohybrid resin composites. Fifteen discs of each material (Nanohybrid resin composite, Nanoceramic resin composite) were prepared, polished, and sterilized in a gamma radiation chamber. Specimens were exposed to the S. mutans bacterial suspension (0.5 McFarland) and were incubated for 4 hours. Specimens were rinsed and sonicated in normal saline, 10 μl of the obtained suspension was cultured in a sterile blood agar medium. After 24 hours, the number of colony forming units of S. mutans was counted. A sterility test control was considered for each group of materials. The data was analyzed by Independent t test. The means and standard deviations of the logarithmic counts of the colonies on the surfaces of nanohybrid resin composites and nanoceramic resin composite were equal to 3.2±0.87 and 2.8±0.324 respectively. Ceram X Universal did not show any significant difference in the bacterial adhesion compared to Filtek Z350XT. Both composites showed similar behaviour in terms of S. mutans colonization in a simple biofilm formation model.

Background: Streptococcus sanguinis is known as an early agent of formation of bacterial plaque. White tea is one of plant with various biological activities that are beneficial to human health.Objective: to examine the antibacterial activity of white tea cathechin extract against bacteria S. sanguinis.Methods: test material was white tea cathechin extract. In vitro tests were conducted to obtain minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC).Result: The MIC of White tea cathechin extract was 500ppm (0,5 mg/ml) and MBC was 2000ppm (2mg/ml).Conclusion: White tea cathechin extract has antibacterial properties. White tea cathechin extract can be used as herbal moutwash to control dental plaque.

This work aims to verify the in vitro action of antiseptics used in the pre- and post-dipping against multidrug-resistant bacteria from bovine mastitis. Antiseptic solutions in the concentration of use of iodine, chlorhexidine and sodium hypochlorite were tested against Staphylococcus aureus (n=12), Staphylococcus sciuri (n=1), Staphylococcus lentus (n=1), Streptococcus sp. (n=1), Enterococcus faecalis (n=1), Enterococcus casseliflavus (n=1), Kocuria kristinae (n=2), Kocuria varians (n=1). Was verified the Minimal Inhibitory and Bactericidal Concentration. The cytotoxicity test complemented the work. It was found that the Minimal Inhibitory Concentration and Minimal Bactericidal Concentration of the iodine, sodium hypochlorite and chlorhexidine was effective in the values recommended for use, however the presence of organic material reduced the activity of the tested products. Chlorhexidine showed higher efficiency of the other disinfectants.
