Antioxidants are the radical scavengers that inhibit peroxidation and other free-radical processes, which in return safeguard different organisms from various diseases attributed to radical reactions. Synthetic antioxidants inhibit free radicals, but they also have harmful side effects. However, mycochemicals of natural fungal origin are safe and best substitutes for harmful synthetic chemical antioxidants. The prime objectives of the study include appropriate qualitative and quantitative mycochemical screening, antioxidant potential, and chemical composition of Snef1216 (Penicillium chrysogenum). The study has used aluminium chloride colourimetric method, Folin–Ciocalteu reagent assay, and DPPH (1,1-diphenyl-1-picrylhydrazyl) for analysis of total flavonoid content and phenol content and antioxidant activity, respectively. However, the presence of biologically active compounds was screened through gas chromatography-mass spectrometry (GC-MS). Quantitative analysis demonstrated the existence of flavonoids, glycosides, flavones, saponins, phenols, and catecholic tannins excluding alkaloids, terpenoids, steroids, and gallic tannins. The outcomes exposed total flavonoid content and phenolic content in P. chrysogenum were 85.31 ± 1.23 mg·QE/g and 135.77 ± 1.14 mg·GAE/g, respectively. Snef1216 (P. chrysogenum) displayed the highest free-radical scavenging activity with 63.86% inhibition of DPPH. The analysis confirms that Snef1216 (P. chrysogenum) is an alternative source of natural antioxidants. The obtained data have provided the foundation for its use in agricultural, environmental, and pharmaceutical industries.

Litsea glutinosa (Lour) leaves are deliberated as worthy traditional medicine. The aim of this study was to screen the phytochemicals, to evaluate the total flavonoid contents as well as antioxidant activity of various extract of Litsea glutinosa (Lour). The phytochemical analysis of each solvent extract was also carried out. Total flavonoid content was determined by aluminium chloride colorimetric assay. Antioxidant activity was determined using 2, 2-diphenyl-1-picrylhydrazyl (DPPH), Ferric reducing ability of plasma (FRAP) and Hydrogen Peroxide (H2O2) free radical scavenger methods. Phytochemical screening of various extract revealed the presence of Alkaloids, Flavonoids, Diterpenes, Proteins, Carbohydrate and Saponins. The total flavonoid content was found 1.89, 5.01 and 3.16 mg/100mg of dry weight of pet ether, ethanol and aqueous extract respectively, expressed as Quercetin equivalents. Antioxidant activity was performed using three methods DPPH, Ferric reducing ability of plasma (FRAP) assay and Hydrogen Peroxide scavenging activity.  Our current results emerged that Litsea glutinosa (Lour) act as an antioxidant agent due to its free radical scavenging. Dendrobium amoenum has biologically active phytoconstituents valued for traditional Chinese or folk medicine as tonic. Total phenolic and flavonoid contents of its stem extract was estimated by using Folin-Ciocalteu’s reagent and Aluminium chloride methods respectively. Antioxidant activity was determined by DPPH free radical scavenging method. Total phenolic content found in acetone (134.34 μg GAE/mg extract) and chloroform (101.55 μg GAE/mg extract) extract was significantly higher than other solvent extracts. Similarly, these two extracts had significantly high flavonoid content (acetone: 115.73 μg QE/mg extract, and chloroform: 84.16 μg QE/mg extract). Presence of high phenolic and flavonoid contents in these extracts showed the highest antioxidant activity. Highest antioxidant activity of these extracts was determined by their lowest IC50 value (acetone: 53.19 μg/ml and chloroform: 36.48 μg/ml). Significant negative relationship was found between phenolic content and IC50 (R2 = 0.209, p < 0.01) and flavonoid content and IC50 (R2 = 0.389, p < 0.01), which indicates high antioxidant activity due to high phenolic and flavonoid contents. This result revealed that D. amoenum act as an antioxidant agent due to its free radical scavenging activity which plays a crucial role in the development of new chemotherapeutic agents.

Objective: The present study evaluates the antioxidant and α-amylase inhibitory activity of leaf and bark extracts of Phyllanthus indofischeri with methanol and water as solvents. In addition to this, the total phenolic content and total flavonoid content was determined.Methods: The total phenolic and total flavonoid content of the extracts was determined by folin ciocaletus reagent method and aluminium chloride colorimetric method respectively. The antioxidant and α-amylase inhibitory activity were measured by various assays, including α, α-diphenyl-ẞ-dipicryl-hydrazyl (DPPH) free radical scavenging, 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radical scavenging, superoxide radical scavenging, total antioxidant capacity by phosphomolybdate method and porcine pancreatic α-amylase inhibitory assay. The IC50 values were calculated and compared with standards such as gallic acid, ascorbic acid and α-acarbose.Results: The results illustrated that all the extracts of Phyllanthus indofischeri exhibit significant antioxidant and α-amylase inhibitory activity. Among the extracts, methanolic leaf extract showed high levels of activity followed by bark water extract.Conclusion: Phyllanthus indofischeri extracts had shown antioxidant and α-amylase inhibitory activity. On the basis of these results, Phyllanthus indofischeri can be used as a natural antioxidant and hypoglycemic agent against various disorders related to oxidative stress; and the isolation of bioactive compounds was warranted.

  The focus of the present study was to determine the association of the anti-oxidant activity of Costus speciosus (Koen Ex. Retz.) Sm. with the total phe-nolic content and the total flavonoid content. The antioxidant activity of the extracts was quantified by diphenylpicrylhydrazyl (DPPH) assay. The total phenolic content was determined by the Folin-Ciocalteau method and the total flavonoid content was determined by the aluminium chloride method. Among the four extracts of the plant, the ethanol extract exhibited the high-est antioxidant activity and also the highest total phenolic and flavonoid con-tents. A strong correlation was observed between the antioxidant activity and the total phenolic and flavonoid contents. A statistical model was de-rived to explain this dependence and a non-linear association was observed between the antioxidant activity and the total flavonoid content.

3 monovarietal wines obtained in two consecutive years in Jidvei Winery Romania were investigated in respect of total polyphenols, total flavonoids and antioxidant activity before and after blending for sparkling wine obtaining. The three varieties were Chardonnay, Pinot Noir and Royal Maiden (Feteasca Regala). Total polyphenols were determined with Folin-Ciocalteu reagent, total flavonoid content with sodium nitrite, aluminum chloride and sodium hydroxide and antioxidant activity using Radical Scavenging Activity and Ferric Reducing Antioxidant Power methods. All mentioned parameters were determined by UV-VIS absorption spectrometry. The determined total polyphenolic content were within the interval 307.21-727.43 mgGAE/l wine for simple varieties and within 405.44-472.74 mgGAE/l for blends and final product (sparkling wine). Total flavonoid content ranged between 16.74 and 242.46 mgQE/l wine in simple varieties and between 38.48 and 80.18 mgQE/l for blends and sparkling wine. Antioxidant activity was correlated with total polyphenols and total flavonoids.

Garlic (Allium sativum L.) is one of the most commonly grown vegetables and thanks to its sensory properties, it has an important place in numerous world cuisines. Garlic is also known for its health-promoting properties, which are attributed to its chemical composition. The health benefits of garlic depending on the content of biologically active compounds, which vary between cultivars and geographical areas. Seven cultivars of garlic, namely Sukoradsky, Zahorsky, Germidour, Mojmir, Karel IV., Arkus, Makoi, were analyzed in this study. This study aimed to determine the total polyphenol content, total flavonoid content, and antioxidant activity of garlic. Studied characteristics were analyzed by the UV-VIS spectrometry method. Total polyphenol content ranged from 430.26 to 640.04 mg GAE.kg-1 FW. Total flavonoid content ranged from 10.29 to 60.49 mg CE.kg-1 FW. Antioxidant activity measured by ABTS assay ranged from 1.098 to 1.955 mmol TE.kg-1 FW. Antioxidant activity measured by FRAP assay ranged from 0.63 to 1.467 mmol.kg-1 FW. Highest TPC, TFC, and AA were determined in the cultivar Mojmir. The lowest TPC and TFC were determined in the cultivar Zahorsky. The lowest AA was determined in the cultivar Sukoradsky. High positive correlations were determined between individual parameters.

Objective: The present study qualitative phytoconstituents examine the total phenol, total flavonoid content (TFC), and antioxidant efficiencies traditionally used plant, Meyna spinosa. Methods: Chemical profiling, estimation of total phenolic content (TPC), TFC, and antioxidant activity of ethanol extracts of M. spinosa have performed by applying standard protocols. Antioxidant activity of leaf and stem was evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay. TPC and TFC of the plant were assessed using Folin–Ciocalteu colorimetric and aluminum colorimetric assay, respectively. Results: The findings of the study exhibit that ethanol extract of M. spinosa is proved to be the presence of phytoconstituents (7/9) such as alkaloids, terpenoids, saponins, tannins, phytosterols, flavonoids, and phenolic compounds, while carbohydrate, fixed oils, and fats are unavailable. In addition, phenolic compositions of ethanol extract of leaf and stem; 93.21±2.93 and 54.33±0.69 mg gallic acid equivalents/g extract, respectively; TFCs of leaf and stem have recorded as 61.55±1.21 and 37.55±1.28 mg quercetin equivalents/g extract, respectively. Antioxidant efficiency of both leaf and stem is tested using DPPH radical scavenging assay as IC50 20.68±0.32 and 50.99±0.56 μg/ml, respectively. Conclusion: From the above results, it has concluded that the ethanol extract of the M. spinosa leaves and stems seizes rich phytoconstituents which can be applied in food technology, drug industries, ethnopharmacological fields, etc., for the development of healthiness and to battle against negative health consequences.

Telah dilakukan penelitian untuk menentukan kandungan fitokimia dan aktifitas antioksidan dari ekstrak etanol daun yantan (Blumea chinensis DC). Kandungan fitokimia yang diuji adalah total fenolik dan total flavonoid. Total fenolik menggunakan metode Folin-Ciocalteu  (Yadaf dan Agarwala, 2010), lalu untuk mengetahui total flavonoid menggunakan aluminium klorida (Sultana et al., 2011), dan pengujian aktifitas antioksidan menggunakan 1,1-difenil-2-pikrilhidrazil (DPPH) (Subedi et al., 2011). Hasil penelitian menunjukan kandungan fenolik total sebesar 26,75 mg/L dan kandungan flavonoidnya sebesar 12,56 mg/L,untuk pengujian aktifitas antioksidan didapatkan nilai IC50 sebesar 240,19 mg/L.Research have been done to determining the phytochemical compound and antioxidant activity of yantan (Blumea chinensis DC) leaves. The phytochemical compound which tested is phenolic total and flavonoid total. Phenolic total using Folin-Ciocalteu  method (Yadaf dan Agarwala, 2010), then to determining the total flavonoid using aluminium chloride (Sultana et al., 2011), and test of the antioxidant activity using 1,1-difenil-2-pikrilhidrazil DPPH (Subedi et al., 2011).The result of the total phenolic content was showed 26,75 mg/L and the total flavonoid content is 12,56 mg/L, for antioxidant activity test obtained the value IC50 is 240,19 mg/L.

Abstract Free radicals can cause various degenerative diseases such as cancer and cardiovascular disease. Flavonoids have antioxidant abilities that inhibit damage from free radicals. The roselle antioxidant mechanism is associated with high flavonoid content. Red and purple roselles are two varieties of roselle (Hibiscus sabdariffa L.) cultivated in Indonesia. The aim of this research was to analyze the differences in flavonoid content and antioxidant activity in the calyces of two varieties of roselle (Hibiscus sabdariffa L.), red and purple. Total flavonoid analysis and antioxidant activity testing were done by spectrophotometry in the visible region. Total flavonoid content was analyzed by pretreatment with aluminium chloride and sodium acetate. Antioxidant activity was tested with 2,2-diphenyl-1-picrylhydrazyl. The total flavonoids for both red and purple roselle calyces ethanolic extract were 27.55 and 39.18 mg quercetin equivalent per gram. The half maximal inhibitory concentration values obtained for both red and purple roselle calyces ethanolic extract a were 51.59 and 42.17 μg/mL. The total flavonoid content and antioxidant activity of purple roselle calyces ethanolic extract was higher and stronger than red roselle calyces ethanolic extract.

The flowers of Rhododendron arboreum Smith is a source of polyphenolic compounds. A flavonol, quercetin, was isolated from ethyl acetate fraction of the methanol extract of flower petals of R. arboreum by repeated Sephadex LH-20 column chromatography. Isolated quercetin was characterized by comparing melting point, Rf values, UV and IR spectra with authentic quercetin. The isolated quercetin was used as a standard for the estimation of total flavonoids.  Total phenolic and total flavonoid content in different parts of R. arboreum was carried out spectrophotometrically using Folin-Ciocalteu reagent and Aluminium chloride reagent respectively. Gallic acid and quercetin were used as standard for the construction of calibration curve of phenolic and flavonoid respectively. The results showed that the highest total phenolic content was detected in the 70% acetone extract of the flowers (600 mg GAE/g extract) and petals (600 mg GAE/g extract) and the lowest amount was detected in methanol extract of stem (188 mg GAE/g extract). Similarly, the highest total flavonoid was detected in the 70% acetone extract of the twigs (170 mg QE/g extract) and the lowest amount was detected in the methanol extract of stem (45 mg QE/g extract).  The antioxidant activity of the methanol extracts obtained from different parts of R. arboreum was determined by 2, 2- diphenyl-1-picryl hydrazyl (DPPH) assay and the radical scavenging activity (IC50) was calculated. The highest free radical scavenging effect was observed in leaves with IC50=8.34 ?g/ml and lowest was observed in stem with IC50=67.83 ?g/ml. The IC50 values, total phenolic and total flavonoid content (correlation coefficient R2= 0.923 for phenolic, R2= 0.965 for flavonoid) were correlated which showed strong correlation indicating that the major components responsible for antioxidant activity is phenolics. The highest the phenolic content, the lowest the IC50 value observed. The result indicated that R. arboreum is a rich source of high value polyphenols as natural antioxidant to use in preventive medicine as well as in food and pharmaceutical industry.Scientific World, Vol. 12, No. 12, September 2014, page 34-40
