Prostate cancer (PCa) is the second most commonly diagnosed malignant tumor and the fifth leading cause of cancer death in men in the world. The most common types of tumors are adenocarcinomas. Prostate cancer is a slow-growing cancer. The incidence increases with age. Evaluation of proinflammatory factors such as IL-17A, IL-17F, IL-17RA, and IL-17RC expression makes it possible to assess the impact of inflammatory process on progression of PCa. The aim of the study was to retrospectively assess the histological material of PCa divided into few groups using the Gleason score. Studies were carried out on archival tissue material in the form of paraffin blocks of 40 men with PCa after radical prostatectomy. The control group was composed of 10 men with benign prostatic hyperplasia (BPH). The material was obtained by the transurethral resection of the prostate (TURP). Immunohistochemistry was performed on prepared material using specific primary antibodies against IL-17A, IL-17F, IL-17RA, and IL-17RC. Expression of the antibody to be examined using light microscopy and the Remmele-Stegner score (IRS) in cancer staining was then evaluated. Expression of IL-17 RA was not shown in a group of patients with PCa and in the control group. In the group of patients with Gleason score 8 and 9 PCa, the expression of IL-17A was higher compared to that of IL-17F. In addition, in PCa with an increased grade of Gleason scale, a decrease in the expression of the study inflammatory parameters was found. The inflammatory process has an impact on PCa. A study on IL-17 may become a starting point for further research on an attempt to use, for example, immunotherapy in PCa.

e17504 Background: The inflammatory reaction pathways have impact on tumor cells development or anti-tumor responses. Both by the activity of the innate and acquired immune response signalling pathways. A proper acquired immune response is considered as anti-tumor. While inadequate innate or acquired stimulation of the immune system can cause chronic inflammation that can lead to oncogenesis. Numerous reports have revealed a relationship between chronic prostatitis and prostate cancer (PCa). The aim of the study is to retrospectively assess the histological material of PCa with the Gleason grading score. The histological material evaluated the level of inflammatory factors of pathways initiated by IL-17A and IL-17F. The evaluation of the expression of pro-inflammatory factors such as IL-17A, IL-17F, IL-17RA, IL-17RC, AKT1, EBPbeta, TRAF 6 and NF-kB made it possible to assess the influence of the inflammatory process on the progression of PCa. Methods: Studies were carried out on archival tissue material in the form of paraffin blocks of 40 men with PCa after radical prostatectomy. Statistical analysis was performed using the non-parametric Kruskal-Wallis test. Results: In statistical analysis, it was shown that the inflammatory pathway IL17A/IL-17RC/TRAF6/NF-kB occurs in both BPH and PCa. IL-17 RA did not show expression in any group of patients and in the control group. In addition, along with the increase in the grading of Gleason score, a decrease in the expression of the tested inflammatory parameters was demonstrated. Conclusions: The inflammatory process has an impact on the cancer of prostate cancer. There is a correlation between the grades according to Gleason score and the level of expression of inflammation parameters. Activation of the inflammatory pathway through IL17A/IL-17RC/TRAF6/NF-kB cascade could correlate PCa development on the base of BPH. Evaluation of the inflammatory pathway in PCa, the initiated IL-17, may become a starting point for further research on an attempt to use, for example, immunotherapy in PCa.

Body fluids often contain freely circulating nucleic acids, many of which can be exploited as noninvasive tools for the diagnosis of cancer as well as for clinical prognostication. Identifying microRNAs (miRNAs) in subjects’ blood with various malignancies means that they can serve as novel biomarkers for prostate cancer (PCa) diagnosis. This study analyzed serum-circulating miRNAs as a noninvasive biomarker in subjects with PCa and subjects with benign prostatic hyperplasia (BPH). In total, 31 PCa subjects and 31 BPH subjects were included, with the BPH group serving as the control group. RT-qPCR was used to quantify the levels of 10 miRNAs, which included miR-18a, miR-34a, miR-106b, miR-183, miR-200a, miR-301a, miR-141, miR-182, miR-200b, and miR-375 in serum. Statistical tests were used to assess the relationship between the levels of miRNAs and the clinicopathological data. A significant increase was observed in the relative expression ratios of miR-141, miR-182, miR-200b, and miR-375 (1.89-, 2.09-, 2.41-, and 2.27-folds, respectively) in the PCa group when compared to the BPH group. Based on the receiver operating characteristic (ROC) analysis, the largest area under the curve (AUC), 0.923, was associated with the miR-200b group, indicating effective diagnostic properties for this biomarker. A correlation was observed between total prostate-specific antigen (TPSA) and the relative levels of miR-141, miR-182, miR-200b, and miR-375. The Gleason score and the miR-200b expression level were also correlated. These results are consistent with previous studies regarding the possibility of differentiating between PCa subjects and healthy controls based on the detection of miRNA. The findings attest to a distinctive expression profile of miRNA that is detectable in the blood of PCa subjects, thereby confirming the role of miRNAs as diagnostic biomarkers for PCa.

Background: Prostate cancer is the most common cancer in men. Prostate specifi c antigen is a key tumor marker to screen for prostate cancer. However, controversy exits regarding the impact of prostate cancer screening. Materials and Methods: This was a retrospective study carried out in the Department of Histopathology of B and B Hospital during a period of 2 years from 01-05-2064 to 01-05-2066. The study included 26 cases of prostate needle biopsy specimens. Results: The more common age group (43.9%) was 70-79 years followed by the age group above 80 years old (7.5%). The most common benign lesion observed was benign prostatic hyperplasia (5 out of 11, 45.45%), followed by benign prostatic hyperplasia with chronic prostatitis (3 out of 11, 27.27%). The most common malignant types of lesion encountered was prostatic adenocarcinoma with an incidence of 84.62% (11 out of 13 cases), following nodular hyperplasia of prostate with low grade prostatic intraepithelial neoplasm 15.38% (2 out of 13 cases). Among the benign and the malignant cases serum PSA ranged from 0 - ?10.1 ng/ml. Conclusion: Serum PSA value has signifi cant role in the diagnosis of malignant prostatic lesions. However, Histological diagnosis still remains the gold standard for the diagnosis of prostatic cancer. Background: Depletion of cellular antioxidants can result from free radical formation due to normal endogenous reactions and the ingestion of exogenous substances and environmental factors. The levels of reactive oxygen species (ROS) have been shown to be significantly altered in malignant cells and in primary cancer tissues. We undertook the present study to investigate the possible alteration of oxidant/antioxidant status in Algerian patients with benign prostatic hyperplasia (BPH) and prostate cancer (PCa). Methods: In total, 89 subjects made up of 26 patients with non-metastatic prostate cancer, 31 with benign prostatic hyperplasia (BPH), and 32 controls participated in this study. The concentrations of plasmatic malondialdehyde (MDA), erythrocytes catalase activity (CAT), and the plasma glutathione levels (GSH) were estimated using standard procedures. Results: The results showed that MDA concentrations were significantly increased while erythrocyte catalase activity was significantly decreased in the prostate cancer group versus controls (P < 0.01) and BPH group (P < 0.05). GSH levels were lowered in prostate cancer patients versus control group with no significant changes. Conclusions: Our results suggest that an alteration in the lipid peroxidation index with concomitant changes in the antioxidant defense system in prostate cancer patients compared with controls. We hypothesize that an altered pro-oxidant–antioxidant balance may lead to an increase in oxidative damage and consequently may play an important role in prostate carcinogenesis.
