A male mixed breed dog was presented with two large wounds, extending the epidermis, dermis, and fascia: one at the dorsum of the thoracolumbar region and the other at the lumbosacral area. Lesions had extended inconspicuously to the dorsum of thorax affecting a large area, which showed regions with necrotic and crepitating foci after shaving. Based on histopathological and bacterial culture examinations, polymicrobial necrotizing fasciitis (NF) was diagnosed. Using the Bruker MALDI Biotyper identification technique, Macrococcus caseolyticus, Proteus mirabilis, and Escherichia coli were identified. Hitherto, there is no report on these bacteria linking them simultaneously to NF in a dog. In addition, the authors highlight other microbes associated with NF in humans and animals.

Aim: To find out the frequency of different bacterial species and their antibiogram among the patients of chronic suppurative otitis media. Study Design: Descriptivecross-sectional. Place and Duration of Study: The study was conducted at the ENT department of Bahawal Victoria hospital between July to December 2020. Methodology: About 50 patients presented with unilateral or bilateral ear discharge and diagnosed as a case of chronic suppurative otitis media were included in the study. The disk diffusion method was used on Mueller-Hinton Agar to detect antimicrobial susceptibility. Multipledrugs were tested for checking antimicrobial susceptibility. The data were analyzed by using Statistical Package for the Social Sciences (SPSS) version 20. Results: The mean age of the study participants was 13.89 ± 12.37 years. The majority of participants were froma younger age group i.e. less than10 years of age with female predominance. It had been found that the majority of cases (51%) were having Staphylococcus aureus infestation followed by Pseudomonas aeruginosa, Escherichia coli and very few with Proteus mirabilis. On the other hand, multiple drugs were tested to find out the antimicrobial susceptibility among the cases of chronic otitis media and the results reported that all the bacterial species were susceptible to Ceftriaxone while the other antibiotics were havinga variable response for different strains of bacteria. Conclusion: Results concluded that the Staphylococcus aureus was the most common organism followed by Pseudomonas aeruginosa, Escherichia coli and very few with Proteus mirabilis. It is important to identify the pathogenic organism and its sensitivity pattern before prescribing any antibiotic.

The first step in the infection of a bacterium by a virus consists of a collision between cell and bacteriophage. The presence of virus-specific receptors on the cell surface will trigger a number of events leading eventually to release of the phage nucleic acid. The execution of the various "steps" in the infection process varies from one virus-type to the other, depending on the anatomy of the virus. Small viruses like ØX 174 and MS2 adsorb directly with their capsid to the bacterial receptors, while other phages possess attachment organelles of varying complexity. In bacteriophages T3 (Fig. 1) and T7 the small conical processes of their heads point toward the adsorption site; a welldefined baseplate is attached to the head of P22; heads without baseplates are not infective.

Freeze-fracturing splits membranes into two helves, thus allowing an examination of the membrane interior. The 5-10 rm particles visible on both monolayers are widely assumed to be proteinaceous in nature. Most membranes do not reveal impressions complementary to particles on the opposite fracture face, if the membranes are fractured under conditions without etching. Even if it is considered that shadowing, contamination or fracturing itself might obscure complementary pits', there is no satisfactory explanation why under similar physical circimstances matching halves of other membranes can be visualized. A prominent example of uncomplementarity is found in the erythrocyte manbrane. It is wall established that band 3 protein and possibly glycophorin represents these nonccmplanentary particles. On the other hand a number of membrane types show pits opposite the particles. Scme well known examples are the ";gap junction',"; tight junction, the luminal membrane of the bladder epithelial cells and the outer membrane of Escherichia coli.

ABSTRACT Recently it has been demonstrated that L-form cells ofProteus mirabilis (L VI), which lack a periplasmic compartment, can be efficiently used in the production and secretion of heterologous proteins. In search of novel expression systems for recombinant antibodies, we compared levels of single-chain variable-fragment (scFv) production in Escherichia coliJM109 and P. mirabilis L VI, which express four distinct scFvs of potential clinical interest that show differences in levels of expression and in their tendencies to form aggregates upon periplasmic expression. Production of all analyzed scFvs in E. coli was limited by the severe toxic effect of the heterologous product as indicated by inhibition of culture growth and the formation of insoluble aggregates in the periplasmic space, limiting the yield of active product. In contrast, the L-form cells exhibited nearly unlimited growth under the tested production conditions for all scFvs examined. Moreover, expression experiments with P. mirabilis L VI led to scFv concentrations in the range of 40 to 200 mg per liter of culture medium (corresponding to volume yields 33- to 160-fold higher than those with E. coli JM109), depending on the expressed antibody. In a translocation inhibition experiment the secretion of the scFv constructs was shown to be an active transport coupled to the signal cleavage. We suppose that this direct release of the newly synthesized product into a large volume of the growth medium favors folding into the native active structure. The limited aggregation of scFv observed in the P. mirabilis L VI supernatant (occurring in a first-order-kinetics manner) was found to be due to intrinsic features of the scFv and not related to the expression process of the host cells. The P. mirabilis L VI supernatant was found to be advantageous for scFv purification. A two-step chromatography procedure led to homogeneous scFv with high antigen binding activity as revealed from binding experiments with eukaryotic cells.

In Veliz-Cuba and Stigler 2011, Boolean models were proposed for the lac operon in Escherichia coli capable of reproducing the operon being OFF, ON and bistable for three (low, medium and high) and two (low and high) parameters, representing the concentration ranges of lactose and glucose, respectively. Of these 6 possible combinations of parameters, 5 produce results that match with the biological experiments of Ozbudak et al., 2004. In the remaining one, the models predict the operon being OFF while biological experiments show a bistable behavior. In this paper, we first explore the robustness of two such models in the sense of how much its attractors change against any deterministic update schedule. We prove mathematically that, in cases where there is no bistability, all the dynamics in both models lack limit cycles while, when bistability appears, one model presents 30% of its dynamics with limit cycles while the other only 23%. Secondly, we propose two alternative improvements consisting of biologically supported modifications; one in which both models match with Ozbudak et al., 2004 in all 6 combinations of parameters and, the other one, where we increase the number of parameters to 9, matching in all these cases with the biological experiments of Ozbudak et al., 2004.

Abstract The purpose of this study was to evaluate the behavior of E. coli O157:H7 during lactose hydrolysis and fermentation of traditional and low lactose yogurt. It also aimed to verify E. coli O157:H7 survival after 12 h of storage at 4 ºC ±1 ºC. Two different types of yogurts were prepared, two with whole milk and two with pre-hydrolyzed whole milk; in both groups one yogurt was inoculated with E. coli O157:H7 and the other one was not inoculated. The survival of E. coli and pH of yogurt were determined during fermentation and after 12-h refrigeration. The results showed that E. coli O157:H7 was able to grow during the fermentation period (from 4.34 log CFU.mL-1 to 6.13 log CFU.mL-1 in traditional yogurt and 4.34 log CFU.mL-1 to 6.16 log CFU.mL-1 in low lactose yogurt). The samples with E. coli O157:H7 showed gas formation and syneresis. Thus, E. coli O157:H7 was able to survive and grow during fermentation of traditional and low lactose yogurts affecting the manufacture technology. Moreover, milk contamination by E. coli before LAB addition reduces the growth of L. bulgaricus and S. thermophilus especially when associated with reduction of lactose content.

Bidens pilosa is less widespread than the other composites commonly found in Singapore as it is animal-dispersed, as opposed to wind-dispersed like most of the Asteraceae. However, this species has been observed to spread over a large area within a year in suitable habitats of sandy or rocky moist substrate with adequate sunlight, such as Jalan Chichau and Jalan Lam Sam. Judging by its invasive success in other tropical and subtropical countries, it should do well in Singapore. More studies can be done on this species, and the populations in the abovementioned localities should be closely monitored to determine the success, and the spread of the species.

1. A rumen bacterial culture containing specifically labelled nucleic acids was prepared using [8-14C]adenine.2. The labelled preparation was given in a liquid diet to two preruminant lambs and via a rumen tube to two ruminant lambs. The radioactivity excreted in exhaled gases, faeces and urine and that incorporated into tissues was determined.3 The preruminant lambs absorbed 58.3% of the total radioactivity measured after 24 h and the ruminant lambs 66.6% of the total activity measured after 48 h.4. Of the total radioactivity absorbed the preruminant lambs exhaled 38%, excreted 34% in urine and retained 29% in tissues. The corresponding values for the ruminant lambs were 12,41 and 47% respectively.5. There was a close relationship between total nucleic acid content and radioactivity per g of tissues of both preruminant and ruminant lambs.6. Of the radioactivity in the urine, the ruminant and one preruminant lamb excreted most in the fraction containing allantoin, while the other lamb excreted most activity in the uric acid fraction.7. The salvaging of the breakdown products of bacterial nucleic acids to make tissue nucleic acids appears to be an important synthesis in preruminant and ruminant lambs and of the likely precursors the purine base may be more important than the nucleoside.
