Morphological and molecular characterisation of three known species of Criconematoidea from Iran

Summary In this study, we provide morphological and molecular characterisation of three known species of the Criconematoidea, namely Criconema cylindraceum, Paratylenchus alleni and P. steineri, recovered from Iran. Descriptions, morphometric data, drawings and photomicrographs are provided, and comparisons are made with closely related species. Phylogenetic analysis using the D2-D3 expansion segments of the 28S rRNA gene revealed that C. cylindraceum formed a cluster with Ogma decalineatus, a species with a similar head structure, but with differences in cuticle ornamentation, tail shape and morphometric characters. Moreover, P. alleni grouped with species of Paratylenchus having four lateral lines and a short stylet (< 40 μm), exceptionally including the unresolved P. straeleni, while P. steineri formed a clade with long stylet species, including P. teres and P. wuae. The taxonomic placement of C. cylindraceum in Criconema, rather than in Criconemoides, is justified, and P. teres is molecularly differentiated from P. steineri, the two species differing in having or lacking a stylet in the fourth-stage juvenile.

During recent collections in South Africa, two species of Criconemoides were found. Criconemoides brevistylus is described and illustrated from a golf course in KwaZulu-Natal Province. It is compared with several closely related species, and three previously described species (C. helicus, C. onostris and C. paronostris) are regarded as junior synonyms of C. brevistylus. Criconemoides obtusicaudatus is described and illustrated from a maize field in Limpopo Province. The male of the species is described for the first time. Molecular characterisation of C. brevistylus and C. obtusicaudatus using the D2-D3 expansions segments of 28S rRNA and ITS rRNA gene sequences are provided. Phylogenetic relationships of these species with other Criconemoides are discussed.

Summary Helicotylenchus ciceri n. sp. and H. scoticus are described and illustrated based on morphological, morphometric and molecular characters. The new species is characterised by a conical and truncated lip region with five or six distinct annuli, stylet 32-37 μm long with anteriorly concave knobs, secretory-excretory pore posterior to the pharyngo-intestinal valve, dorsally convex-conoid tail with a terminal projection, phasmids 14 (7-20) annuli anterior to the level of anus, empty spermatheca and absence of males. Intraspecific variation of 16 populations of H. scoticus, collected from chickpea and lentil fields in Kermanshah province, western Iran, is discussed. The results of the phylogenetic analyses based on the sequences of the partial 18S rRNA, D2-D3 expansion segments of 28S rRNA and ITS rRNA genes are provided for the studied species, confirming their differences from each other and determining the position of them and their relationships with closely related species.

Xiphinema granatum n. sp. from pomegranate in Saveh, Markazi province, central Iran, is described and illustrated. The new species belongs to species group 8 of the genus Xiphinema sensu Loof & Luc, 1990. The new species is characterised by a body 3.5-4.1 mm long, 37-49 μm diam., odontostyle and odontophore 118-132 and 65-74 μm long, respectively, presence of sperm in the female genital tracts, uterus devoid of any Z-differentiation or spines, and abundant males with spicules 45-58 μm long. The polytomous identification codes of the new species are: A4, B4, C5a, D5, E56, F4, G23, H2, I3, J4, K2, L2. Beside morphological and morphometric data, molecular analyses of D2-D3 fragments of 28S rRNA gene placed the new species in close relationship with X. vuittenezi and X. italiae and some species belonging to Xiphinema morphospecies group 6, but clearly separated from X. index. In addition, an Iranian population of Longidorus pisi from the same locality is morphologically and molecularly characterised.

Summary Six species of the genus Tylencholaimellus, one new and five known, collected in virgin and cultivated soils of East Azarbaijan province, Iran, are studied. Tylencholaimellus zeinabadensis sp. n. is morphologically characterised by its 1.2-1.7 mm long body, lip region 10-13 μm wide and offset by deep and broad constriction, odontostyle 16-20 μm long, stylet 22-30 μm long, neck 186-234 μm long, pharyngeal bulb 30-43 μm long, anterior uterine sac 55-103 μm long or 0.9-1.7 corresponding body diam. long, V = 34-41, tail short and rounded to conoid (20-30 μm, c = 45-83, c′ = 0.5-1.0), spicules 39-50 μm long, and one or two ventromedian supplements with hiatus. Molecular analyses using the D2-D3 expansion regions of the 28S rRNA gene sequences of the new species confirmed the monophyly of this genus based upon currently available data. A population of T. affinis is described and illustrated, and new data (morphometrics, pictures and distribution) are provided for T. loofi, T. paracinctus, T. polonicus and T. striatus.

During a survey on pin nematodes in western Iran, two populations of Paratylenchus audriellus and Paratylenchus tenuicaudatus were collected and subsequently analysed morphologically and molecularly. Paratylenchus audriellus is characterised by the long stylet (48-61 μm) and the typical female tail with a characteristic claw-like process with sharply pointed terminus. To our knowledge, the Iranian population of P. tenuicaudatus is the first record from Iran. The molecular characterisation of P. audriellus nematodes using the D2-D3 of 28S rRNA and the partial 18S rRNA gene sequences revealed that this species is clearly separated from P. straeleni and should be considered as a valid taxon.

AbstractCaloosia longicaudata is described from Maui, the Hawaiian Islands, for the first time and both sexes are characterised morphologically using light and scanning electron microscopy. Molecular characterisation of C. longicaudata using the D2-D3 domain of 28S rRNA, partial 18S rRNA and ITS rRNA gene sequences is also provided. The phylogenetic relationships of this species with other representatives of the suborder Criconematina are presented and discussed. A diagnostic PCR-ITS-RFLP profile for C. longicaudata is given together with an identification table for eight species of Caloosia. Caloosia langola n. comb. is transferred to the genus and C. shorai is synonymised with H. psidii.

Root-lesion nematodes (Pratylenchus) have a worldwide distribution and cause severe production constraints on numerous important crops. During a survey of the wheat-growing area of Morocco, 17 populations of root-lesion nematode were collected. They were identified on the basis of their morphological and morphometric characters, and by molecular methods. Microscopic observations of females and males demonstrated the occurrence ofP. penetransin 13 of the 17 samples;P. thorneiandP. pseudocoffeaewere detected in four samples from Zaers and a single sample from Settat, respectively. A duplex PCR primer set was used to confirm the presence ofP. penetranswhile the species-specific forward primer PTHO and the common reverse primer D3B were used forP. thornei. For the remaining populations, the D2-D3 expansion segments of the 28S rRNA gene were amplified and the obtained sequences were compared with those ofPratylenchusspecies in the GenBank database. This comparison confirmed the morphological identifications and revealed a population ofP. pinguicaudatus. The study of the phylogenetic relationship of the MoroccanPratylenchuspopulations showed a high similarity (99-100%) between allP. penetranspopulations. The population dynamics of sixPratylenchuspopulations from Morocco were evaluated on carrot disk cultures at 4, 8 and 12 weeks after inoculation, and at 10, 15, 20 and 25°C. The optimum temperature for reproduction of all populations was 20°C. After 8 weeks at this temperature, nematode numbers increased up to 458-fold, 310-fold and 252-fold for the four populations ofP. penetrans, theP. thorneiand theP. pseudocoffeaepopulation, respectively.

The reniform nematodes of the genusRotylenchulusare semi-endoparasites of numerous herbaceous and woody plant roots and are mainly distributed in tropical and subtropical regions. In this study, we provide morphological and molecular characterisation of six out of ten presently known valid species ofRotylenchulus:R. clavicaudatus,R. leptus,R. macrodoratus,R. macrosoma,R. reniformisandR. saccharifrom South Africa, USA, Italy and Spain.Rotylenchulus parvuswas only studied morphologically. A new species,R. macrosomoidessp. n., isolated from soil and roots of sugarcane in South Africa, is described. The phylogeny ofRotylenchulus, as inferred from the analyses of D2-D3 of 28S rRNA, ITS rRNA,coxImtDNA andhsp90gene sequences, is presented. The study revealed thatR. reniformisandR. macrosomahave a sister relationship, but that relationships between otherRotylenchulusspecies remain unresolved. The phylogenetic analysis also confirmed the hypothesis that this genus originated from the Afrotropical zoogeographical region. Our study revealed thatR. reniformisandR. macrosomoidessp. n. have two distinct rRNA gene types andR. macrosomahave three rRNA gene types in their genomes. PCR with species-specific primers was developed for rapid diagnostics ofR. reniformis.

AbstractTwo Hemicycliophora species, H. lutosa and H. typica, found in samples from fallow soil and sugarcane soil in South Africa, were studied morphologically and, for the first time, molecularly. Diagnostic PCR-IT-rRNA-RFLP profiles generated by five restriction enzymes are provided. Study of phylogenetic relationships using D2-D3 expansion segment of 28S rRNA gene sequences revealed that H. lutosa was related to H. poranga. Hemicycliophora lutosa and H. poranga are compared morphologically. SEM photographs are given for H. typica and for H. lutosa for the first time. The male of H. typica represents a first report for South Africa.

Pin nematodes of the genus Paratylenchus are widely distributed across the world and associated with many plant species. Morphological identification of Paratylenchus species is a difficult task because it relies on many characters with a wide range of intraspecific variation. In this study we provide morphological and molecular characterisation of several pin nematodes: Paratylenchus aquaticus, P. dianthus, P. hamatus, P. nanus and P. straeleni, collected in different states of the USA and South Africa. Paratylenchus aquaticus is reported from South Africa and Hawaii and P. nanus is found from South Africa for the first time. Morphological descriptions, morphometrics, light and scanning electron microscopic photos and drawings are given for these species. Molecular characterisation of nematodes using the D2-D3 of 28S rRNA and ITS rRNA gene sequence revealed that samples morphologically identified as P. aquaticus, P. hamatus and P. nanus indeed represent species complexes containing several species. Sequences of the rRNA genes are also provided for several unidentified Paratylenchus. Phylogenetic relationships within the genus Paratylenchus are given as inferred from the analyses of the D2-D3 of 28S rRNA and ITS rRNA gene sequences. We present here the most complete phylogenetic analysis of the genus.
