Histopathological findings in the uteri and ovaries of clinically healthy cats presented for routine spaying

Objectives The aim of this study was to assess the histopathological findings in the uteri and ovaries from clinically healthy queens presented for elective spaying. Methods Ovaries and distal uterine horns or complete uteri from 106 female cats were evaluated for pathological alterations. Results Pathological alterations of the uterus and/or ovaries were evident in 29 cats; of these, corpora lutea were present on the ovaries of 15 cats. Ovarian cysts were found in 15 cats and were classified as cysts of the Wolffian or Muellerian ducts (n = 4), follicular cysts (n = 4), luteal cysts (n = 1), cystic rete ovarii (n = 2), combinations of different cysts (n = 2) and non-classifiable cysts (n = 2). In 21/106 cats, cystic endometrial hyperplasia (CEH) was present. The incidence of CEH increased with the age of the cat. Six cats had purulent endometritis with or without distension of the uterine lumen. Hyperplastic lesions of the endometrium were detected in two cats. In one cat, a uterine horn malformation with duplication of one uterine horn lumen was diagnosed. Conclusions and relevance Whereas the majority of ovarian cysts and slight-to-moderate CEH are unlikely to interfere with an animal’s wellbeing, endometritis must be considered a serious health problem that requires veterinary attention.

SUMMARY Blastocysts were studied on the 5th and 8th day of pregnancy in lactating mice, in the fresh state, flushed from the uterus, in squash preparations and in serial sections. At the earlier period some mitosis was observed. Tritiated thymidine incorporation studies gave some evidence of DNA synthesis on the 5th and 6th days of pregnancy. By the 8th day the blastocysts were longer, contained more cells, and mitosis had ceased. They were located at the anti-mesometrial end of the uterine lumen, closely apposed to the uterine epithelium, and with their long axes parallel to the long axis of the uterine horn. Implantation could be induced, either by the removal of the litter, or by the injection of an appropriate dose of oestrogen on the 5th or 7th (but not the 4th) day of pregnancy. Both treatments were followed by the appearance of W-bodies in the neighbourhood of the blastocysts, the disappearance of the shed zonae, and the appearance of Pontamine Blue reactivity, oedema of the uterine stroma and formation of the primary decidual zone, in that order.

Objective: Spinal pain with or without referred pain is a major and costly health problem that can arise from many anatomical structures. Sophisticated diagnostic imaging devices cannot show some of these structures, and frequently imaging provides only a shadow of the truth. This review illustrates how symptoms may well have an organic cause that is not detectable by current methods of examination, including imaging. Method: This study reviews some histopathological findings that can be associated with spinal pain with or without referred pain but cannot be seen on imaging. Result: Some histopathological changes illustrate imaging device limitations. Conclusion: Awareness of the considerable limitations of even sophisticated imaging devices is necessary when managing patients with acute or chronic spinal pain with or without referred pain. Symptoms may well be genuine and not of psychogenic origin: a diagnosis of malingering, therefore, should not be made lightly.

SUMMARY In mated guinea-pigs one uterine horn was rendered sterile by ligation of the oviduct 2 or 3 days after finding spermatozoa in the vaginal smear. Two glass beads were inserted into the sterile horn on each of days 3–12 and on day 14 in experimental animals but not in controls. At autopsy on day 20 large corpora lutea were present in both ovaries of the control animals. The presence of beads that had been introduced on days 3 and 4 and on days 10–14 resulted in marked regression of the corpora lutea in the adjacent ovary, in the absence of a decidual reaction in the uterus, while luteal enlargement typical of pregnancy occurred in the contralateral ovary. Beads inserted on days 5–8 caused decidualization in the sterile horn but did not induce premature luteal regression in the ipsilateral ovary.

Bovine uterine horns with both ovaries containing a corpus luteum (CL) were compared for progesterone (P4) and oxytocin (OT) concentrations during the luteal phase of the estrous cycle. Uterine tissue samples from five Holstein cows with bilateral CL obtained from the slaughterhouse were used for this study. No significant difference was observed in P4 and OT levels in the right and left horns with corpora lutea in both ovaries. The data clearly indicate that both sides of the uterine horn having a functional CL are exposed to similar levels of P4 and OT, supporting the hypothesis that luteal products are delivered locally to the uterus. SUMMARY The numbers of corpora lutea and the numbers and sizes of 6-day blastocysts have been studied in rabbits, widely varying in ages and weights, that had been subjected to a superovulation treatment. For all does, the average value of each characteristic on the left sides of the animals was similar to that on the right, the average left-to-right difference being not significant. An average of thirty-one corpora lutea and thirteen blastocysts per doe was obtained. Maximum numbers per doe were one hundred and fifty-three corpora lutea and seventy-three blastocysts. Average blastocyst diameter was 2·203 mm, the average volume being 6·780 mm3. There was a fairly large variation from one doe to another in the relative numbers of corpora lutea (or in the relative numbers of blastocysts) on the two sides of the animal. The number of blastocysts per doe was positively related to the number of corpora lutea. The number of corpora lutea per doe was inversely related to the age of the doe. An inverse relation between the number of blastocysts and the age of the doe was presumably indirect, being mediated by the number of corpora lutea. All these relationships were highly significant. There was no demonstrable relationship between the number of corpora lutea and the weight of the doe. No firm evidence was found for a direct relationship (independent of weight and number of corpora lutea) between the number of blastocysts and age, or for any relationship with weight. Diameter of blastocysts seemed more appropriate than volume as a measure of size suitable for analysis. Transformations tending to give linear relationships between variables have been studied. Size differences between blastocysts within the same uterine horn formed a large source of variation, probably not attributable to the superovulation procedure. Differences in blastocyst size between uterine horns within does were significant but introduced little extra variation. Mean blastocyst size varied greatly from one doe to another, but none of this variation was related significantly to known factors such as the weight of the dam or sire, or the age of the dam. The experiment appeared to be fairly sensitive in detecting these relationships. The relationships found between the number of corpora lutea and the age or weight of the doe can be reconciled with the previous literature. The lack of demonstrable connexion between blastocyst size and the weight of dam or sire is in conflict with the results of other authors, and no specific reconciliation can be made. It is suggested that uniformity in age is more important than uniformity in weight when test animals are selected for assay of follicle stimulating hormone under conditions comparable with those of the present work.

Induction of ovulation post partum is associated with a high incidence of prematurely regressing corpora lutea. However, inadequate luteal function is not the sole reason for pregnancy failure, because ewes with normal corpus luteum function and successful fertilization also fail to establish pregnancies. The effects of suckling status and the interval from post partum to rebreeding on corpus luteum and endometrial function were examined in vivo and in vitro. Ewes were weaned early or allowed to lactate, induced to ovulate using a progesterone-impregnated controlled internal drug release device and an intramuscular injection of pregnant mare serum gonadotrophin, and inseminated (intrauterine) at either 21 or 35 days post partum (n = 10 per group). A further 10 standard ewes whose interval from parturition was in excess of 150 days were included for comparative purposes. On Day 10 after insemination the pregnancy rate was determined in four ewes from each of the post-partum groups and five standard ewes. These ewes were then ovariectomized and hysterectomized for studies in vitro. The incidence of premature luteal regression, as assessed by progesterone concentrations in peripheral blood was independent of the suckling stimulus but dependent on stage post partum (21 days post partum, 6 of 19 ewes; 35 days post partum, 0 of 19 ewes; P less than 0.05). Luteal function was normal in all standard ewes. Ovulation rate, corpus luteum weight, corpus luteum progesterone content and basal progesterone production in vitro were significantly less in 21-day than in 35-day post-partum ewes. Pregnancy rates as determined on Day 10 or at term were low in all post-partum groups (7 out of the 38 ewes inseminated) compared with standard ewes (8 of 10). Uterine function was assessed by culturing endometrial tissue from the tip and body of each uterine horn in the presence of [3H]leucine for 30 h at 37 degrees C. Incorporation of radiolabel into non-dialysable proteins synthesized and secreted by the endometrium in vitro was independent of uterine horn location and suckling status but was significantly lower (P less than 0.001) in media from 21-day than from 35-day post-partum ewes. Irrespective of treatment group, incorporation of radiolabel was positively correlated with mean plasma progesterone concentrations on Days 2-10 after insemination and with basal progesterone production in vitro. Introduction. The aim of this study was to analyze and compare the histopathological findings in curettage and hysterectomy specimens, to evaluate the accuracy of histopathological diagnosis in curettage specimens, and to determine the frequency of coexisting endometrial carcinoma in patients with histopathological diagnosis of endometrial hyperplasia. Material and methods. Curettage and hysterectomy specimens of 135 female patients with initially diagnosed endometrial hyperplasia were retrospectively analyzed and compared. Results. Simple hyperplasia was found in 49 patients (36.3%), complex hyperplasia in 14 (10.4%), simple atypical hyperplasia in 24 (17.8%), and complex atypical hyperplasia in 48 (35.5%) patients. After hysterectomy, 59 (43.7%) patients were found to have simple hyperplasia, 12 (8.9%) complex hyperplasia, 15 (11.1%) simple atypical hyperplasia, 18 (20.7%) complex atypical hyperplasia, and 21 (15.5%) endometrial carcinoma. The accuracy of histopathological diagnosis of endometrial hyperplasia in curettage specimens was 82.2-89.6% and dependent on the types of hyperplasia. The frequency of coexisting endometrial carcinoma was significantly higher (p<0.001) in patients with atypical hyperplasia than in patients with hyperplasia without cytological atypia. After hysterectomy, coexisting endometrial carcinoma was found in 27.8% of patients with histopathological diagnosis of atypical hyperplasia in curettage specimens. In contrast to simple atypical hyperplasia, the frequency of coexisting endometrial carcinoma was significantly higher (p<0.05) in complex atypical hyperplasia. Conclusion. The frequency of coexisting endometrial carcinoma in hysterectomy specimens in patients with histopathological diagnosis of atypical hyperplasia in curettage specimens was relatively high and it should be taken into account when planning therapy. .

SUMMARY The presence of beads in the guinea-pig uterus caused premature regression of the corpora lutea, and a corresponding decrease in utero-ovarian venous plasma levels of progesterone, by days 9–10 of the oestrous cycle. If the beads were placed in one horn only, premature luteal regression occurred only in the adjacent ovary. Induced luteal regression was preceded by an increased release of prostaglandin F2α (PGF2α) from the uterine horns which contained the beads. Oestradiol benzoate treatment of guinea-pigs carrying beads in one uterine horn overcame the unilateral effect and resulted in premature luteal regression in both ovaries. This was also associated with the early release of PGF2α from both uterine horns, though a difference in the timing of the release was apparent between the bead-containing (experimental) and control horns.

Embryo implantation begins when the blastocyst both assumes a fixed position in the uterus and establishes a more intimate relationship with the endometrium. Successful implantation depends upon hormonal synchronization and development and the receptivity of the endometrium. CD44 is a cell surface molecule that has been implicated in the initial attachment of the embryo. The aim of this work was to study the hormonal levels of P4 in three groups of animals which have a normal pregnancy or an induced reduction in the number of implants. Twelve adult New Zealand does (n = 12) were naturally inseminated with a buck of proven fertility. Blood samples were obtained daily during 20 days of pregnancy. Hormonal determinations were performed by enzyme immunoassay. Animals were divided into three groups: group A (n = 4): control animals; group B (n = 4): endometriosis was surgically induced in the right horn a month before the animal was mated; and group C (n = 4): animals received an injection of 20 micrograms of anti-CD44 in the right horn via mid-ventral laparotomy on Day 6.5 post-coitum (0.5 mL each, from the ovarian end to the cervix). Each animal served as her own control with the left uterine horn receiving 0.5 mL of saline. Statistical analysis was performed using ANOVA.The number of corpora lutea was similar in all treatments. No statistical differences were found comparing CLs in the right/left ovary. In group A, a mean of 3.4 ± 0.47 (mean ± SE) implants was found in the right horn while the mean in the left side was 4.6 ± 0.81. In group B, a marked reduction in implantation sites was found, with 1.8 ± 0.60 and 4.66 ± 0.84 on the right and left horns, respectively. With anti-CD44 injected into the uterine horn (Group C), a mean of 0.12 implant was present in the right uterine horn compared with 3.6 implants on the left side (P < 0.001). Progesterone levels from Days 1 to 10 are shown in the following table (mean ± SE). Comparisons in day values are not statistically significant P > 0.05. After Day 10 the levels of progesterone were similar in all groups. The results showed an increase of progesterone levels in group B; this could be due to endometriosis and not to the number of implants. The results in the CD44 group reveal that progesterone profiles were similar to those in the control group, and we can conclude that the reduced number of implants found in group C did not affect the progesterone levels.
