Essential Oil Composition, Antimicrobial and Cytotoxic Activities of Two Endemic Stachys Cretica Subspecies (Lamiaceae) from Turkey

The chemical compositions of the water-distilled essential oil of Stachys cretica ssp. lesbiaca Rech. fil. and S. cretica ssp. trapezuntica Rech. fil. were determined by GC and GC–MS. Altogether 63 compounds were identified. The sesquiterpene hydrocarbon, germacrene D (20.3% and 12.9% respectively) was the main component identified in both oils. Furthermore, ethanol, light petroleum, dichloromethane, ethyl acetate and n-butanol extracts prepared from the aerial parts of the plants were tested for their antimicrobial activities against six bacterial strains and the yeast Candida albicans. The extracts exhibited no antibacterial activity, but the light petroleum and n-butanolic fractions showed low antifungal activities. Crude ethanolic extracts of the two subspecies were tested for their ability to inhibit the growth of HL-60 and Ishikawa human tumor cell lines. The IC50 values were 100 μg/mL for the HL-60 cell line and 200 μg/mL for the Ishikawa cell line.

The aim of this study was to determine the chemical variability of wormwood extracts as affected by the growing region. Antioxidant and antimicrobial activities were also investigated. The essential oil composition variability ofA. absinthiumL. aerial parts collected from four different Tunisian regions was assessed by gas chromatography (GC/FID) and by gas chromatography mass spectrometry (GC/MS). In addition, total polyphenols, flavonoids, and condensed tannins as well as antioxidant, antibacterial, and antifungal activities of methanolic extract and essential oils were undertaken. Chromatographic analysis of wormwood essential oils showed the predominance of monoterpene hydrocarbons represented mainly by chamazulene. RP-HPLC analysis of wormwood methanolic extract revealed the predominance of phenolic acids. Antiradical activity was region-dependant and the methanolic extract of Bou Salem region has the strongest activity (CI50=9.38±0.82 µg/mL). Concerning the reducing power, the methanolic extract of Bou Salem, Jérissa, and Boukornine regions was more active than the positive control. Obtained results of antimicrobial activities showed that wormwood essential oil is endowed with important antibacterial activity which was strongly related to the organoleptic quality of oil which appeared strongly region-dependant.A. absinthiumL. EOs investigated are quite interesting from a pharmaceutical standpoint because of their biological activities.

AbstractThe present study investigates the chemical compositions of three Satureja montana L. ssp montana essential oils and correlates chemical variability with biological activities. GC/MS analysis showed that with an increase in altitude (100–500–800 m), a higher content of linalool, terpinen-4-ol and cis-sabinene hydrate was found, while the percentage of phenolic compounds, thymol and carvacrol decreased. Antimicrobial activity of the essential oils was tested against 7 fungal and 23 bacterial strains. The essential oil characterized by the highest content of phenols and alcohols exhibited the highest antimicrobial potential. The correlation analysis showed that the major carriers of the obtained antioxidant activity are oxygenated monoterpenes. All essential oils inhibited human serum cholinesterase activity. High antimicrobial potential, together with moderate antioxidant capacity and strong inhibition of human serum cholinesterase, classifies S. montana essential oil as a natural source of compounds that can be used in the treatment of foodborne and neurological diseases, wound and other infections, as well as for general health improvement.

Background: Antibiotic resistance is today one of the most serious threats to global health, food security and development. Due to the growing number of infections, treatment becomes more difficult, if not impossible, because of the loss of antibiotic efficacy. Objective: In the present investigation, the chemical composition of essential oils of Ammoides verticillata and Satureja candidissima and their synergistic effects on antimicrobial activities were investigated. Methods: The chemical composition of the essential oil was analyzed by Gas Chromatography (GC) and Gas Chromatography-Mass Spectroscopy (GC/MS). The antimicrobial activity of the essential oils was investigated using dilution-agar method against nine bacterial strains three Gram-negative Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853) and Salmonella typhi (ATCC 14028), and six Gram-positive: Staphylococcus aureus (ATCC 43300,) Clostridium sporogenes (ATCC 19404), Bacillus subtilis (ATCC 6633), Enterococcus faecalis (ATCC 7314), Lactobacillus rhamnosus (ATCC 53103) and Bacillus cereus (ATCC 14579). Results: The essential oil of A. verticillata was characterized principally by carvacrol (44,3%), Limonene (19,3%) and p-cymene (19,2%). The constituents identified of S. candidissima essential oil were principally oxygenated monoterpenes represented by pulegone (70,4%). The essential oil of A. verticillata had a good antimicrobial activity against four bacterial strains (Escherichia coli, Salmonella typhi, Lactobacillus rhamnosus and Bacillus cereus) with MIC and MBC values between 0.2-0.4 µl/ml and 0.2-6.2 µl/ml, respectively. While, S. candidissima essential oil had moderate antimicrobial activities against all strains with MIC and MBC values between 1.5-6.2 µl/ml and 6.2-12.5 µl/ml, respectively. The results of antimicrobial activity of essential oils blend presented higher antimicrobial activity against all tested bacteria with MIC and MBC values between 0.3-1.5 µl/ml and 0.4-6.2 µl/ml, respectively. Conclusion: The essential oils blend presented high antimicrobial activity compared to virgin oils. This activity can be due to the association of active compounds such as carvacrol and pulegone. These findings provide a new source of drugs that may help in therapy to lead to the development of a new treatment based on a combination of these essential oils against gram-negative and gram-positive bacteria that continue to pose a threat to public health.

The leaf essential oils of five species of Beilschmiedia from Monteverde, Costa Rica (Beilschmiedia alloiophylla, B. brenesii, B. costaricensis, B. tilaranensis, and an undescribed Beilschmiedia species “chancho blanco”) have been obtained by hydrodistillation and analyzed by GC-MS in order to discern the differences and similarities between the volatile chemical compositions of these species. The principal constituents of B. alloiophylla leaf oil were germacrene D (18.9%), cis- and trans-β-ocimene (18.8% and 9.3%, respectively), α-pinene (11.8%), and bicyclogermacrene (9.1%). The leaf oil of B. brenesii was composed largely of the sesquiterpenes germacrene D (19.3%), β-caryophyllene (13.4%), α-copaene (9.0%), α-humulene (8.1%), and δ-cadinene (5.8%), and the carbonyl compounds 2-undecanone (12.8%), trans-2-hexenal (8.8%), and 2-tridecanone (3.8%). α-Bisabolol (72.1%) dominated the leaf oil of B. costaricensis, while B. tilaranensis had germacrene D (54.9%), β-caryophyllene (14.8%), and δ-cadinene (5.1%) as major components. Beilschmiedia “chancho blanco” leaf oil was composed largely of β-caryophyllene (16.6%), bicyclogermacrene (14.1%), and α-pinene (12.1%).

The chemical composition of the essential oil obtained from the aerial parts of Cleome serrata by hydrodistillation was analyzed by employing GC-FID, GC-MS and RI. Fourteen compounds comprising 90.4% of the total oil composition were characterized. The main components identified were ( Z)-phytol (53.0%) and di(2-ethylhexyl)-phthalate (DEHP) (14.7%). The oil was evaluated for its in vitro antimicrobial activities against nine pathogenic microorganisms using the filter paper disc diffusion method. Moderate antimicrobial activity was observed against five of the pathogens assayed. In addition, the essential oil was tested against the sweet potato weevil, Cylas formicarius elegantulus. Strong knockdown insecticidal activity was observed.

The composition and antimicrobial activity of the essential oil of Heracleum sibiricum L. (Apiaceae) was studied. The aerial part of plant was hydro-distilled and chemical composition of the essential oil was analyzed by GC and GC-MS. Forty-six compounds, corresponding to 95.12% of the total oil, were identified. Esters represented the major chemical class (69.55%) while the main constituents were octyl butanoate (36.82%), hexyl butanoate (16.08%), 1-octanol (13.62%) and octyl hexanoate (8.10%). Antibacterial activity of the essential oil and reference antibiotics against nine bacterial strains was tested by the broth microdilution method. The results of the bioassays showed that essential oil had slight antimicrobial activities against all tested microorganisms (MIC and MBC values were in the range of 2431.2 to 9724.8 μg/mL). Reference antibiotics were active in concentrations between 0.5 and 16.0 μg/mL. The results confirm that Gram-positive bacteria were more susceptible to the essential oil of H. sibiricum, in comparison with Gram-negative bacteria.

The composition of the essential oil of the fruits, leaves and stems of Daucus glaber Forssk has been studied by GC/MS. It was found that, the essential oil of the fruits consists of monoterpene hydrocarbons (limonene and sylvestrene are the majors) and phenylpropanoids (elemicin is the major). Sylvestrene has never been reported before in the essential oil of any Daucus species. The study of the essential oil of the leaves revealed the presence of monoterpene hydrocarbons; limonene and γ-terpinene are the majors and a small amount of sylvestrene. The essential oil of stems consists of monoterpene hydrocarbons (γ-terpinene is the major), terpene alcohols (mainly 4-terpineol) and phenylpropanoids (myristicin and elemicin are the majors). It is interesting that, the essential oil of the fruits is free from any oxygenated terpenes while that of the stems is free from limonene and sylvestrene which are present in the essential oil of the fruits and leaves in fairly large amounts. The essential oil of the fruits, leaves and stems shows broad antimicrobial activities against both gram positive and gram negative bacteria. In addition, the volatile oil of the stem, particularly, show activi­ties against Candida albicans (yeast). Also, the prepared oils have variable cytotoxic activities with LC50 21.52, 36.01 and 42.34 μrg/ml. respectively.

The aerial parts of Satureja spicigera were collected at full flowering stage at Gazvin, Iran. The essential oil was isolated by hydrodistillation and analyzed by a combination of capillary GC and GC-MS. Fourteen compounds were identified, of which carvacrol (53.74%) and thymol (36.03%) were the main constituents, representing 99.12% of the total oil. The in vitro antibacterial activity of the essential oil was determined against six ATCC standard bacterial strains (Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa) using disc diffusion as well as measurement of minimum inhibitory concentrations. The disc diffusion results and MIC values indicated high inhibitory activity against the test bacteria. The most susceptible organisms were the Gram-positive B. subtilis and S. aureus followed by E. faecalis, usually resistant to most common antibiotics. Among the Gram-negative bacteria, E. coli and K. pneumoniae were highly sensitive to the different oil concentrations in the disc diffusion method. Finally, P. aeruginosa, a highly resistant organism to most antibiotics, showed moderate susceptibility to Satureja spicigera essential oil.
