Objective. To evaluate the phytochemical content and antioxidant potential of the acetone, aqueous, and methanol extracts of the fruit, leaf, and root of Cucumis africanus L.f. Methods. Total phenolic, flavonoid, and proanthocyanidin contents were evaluated using spectrophotometric methods. The free radical scavenging activity of the acetone, methanol, and aqueous extracts of the fruit, leaf, and root were evaluated against DPPH+, ABTS+, and phosphomolybdenum. Alkaloid and saponin contents were also determined. Results. The acetone extract of the fruit revealed the highest polyphenol content when compared with the other solvent extracts analyzed. Total phenol content of the parts tested ranged from 3.66 ± 0.17 to 44.98 ± 3.41, flavonoid content ranged from 4.63 ± 3.33 to 401.33 ± 7.89, and proanthocyanidin content ranged from 8.84 ± 2.65 to 504 ± 36.6. Significant amount of alkaloids present was observed in the fruits, leaf, and root (10.68 ± 0.68, 14.12 ± 1.67, and 12.15 ± 4.74), respectively, while saponin content was 33.33 ± 11.55, 26.67 ± 11.55, and 20.00 ± 0.00 for the fruit, leaf, and root, respectively. Solvent extracts showed significant antioxidant activity, with acetone showing highest antioxidant ability in correlation with the polyphenol contents. Based on the IC50 values, acetone extract of the root revealed the best DPPH radical scavenging ability, the leaf aqueous extract had the highest IC50 value for ABTS, and the methanol extract of the leaf was best for phosphomolybdenum assays. Conclusion. This study suggests that fruit, leaf, and root of Cucumis africanus could be a potential source of natural antioxidant and justifies its use in ethnomedicine.

Abstract Ginger (Zingiber officinale) is a popular spice which used for the treatment of different gastrointestinal and inflammatory discomfort. In the present study, the total phenolic content (TPC) and antioxidant activity of ginger extract using four solvents (ethanol, methanol, acetone and ethyl acetate) were determined. Among the four solvents, methanol extract showed that the maximum phenolic (1183.813 mg GAE/100 g at Ayikel and 1022.409 mg GAE/100 g at Mandura) and the least were found in acetone extract (748.865 mg GAE/100 g at Ayikel and 690.152 mg GAE/100 g at Mandura). In addition, the highest DPPH radical scavenging activity (84.868% at Ayikel and 82.883% at Mandura) was observed in methanol. However, acetone showed the least DPPH radical scavenging activity (73.864% at Ayikel and 70.597% at Mandura). Antioxidant activities of ginger extracts were also expressed as IC50 values and acetone extract has maximum IC50 value (0.654 and 0.812 mg/mL) followed by ethyl acetate and ethanol, while the lowest for methanol extracts (0.481 and 0.525 mg/mL). The result of this study showed that extraction solvents significantly affected the total phenolic content and antioxidant activities of ginger. Thus, ginger can be regarded as promising candidates for natural sources of antioxidants with high value of phenolic contents.

This study was designed to determine the polyphenolic contents of Eucommia ulmoicles Oliv leaf extracts and evaluate their antioxidant activities in vitro. All the extracts were evaluated for their radical scavenging activity by the DPPH method and their oxidative-retarding efficiency in vegetable oils. It was observed that the highest values were found in 70% MeOH extracts for total phenolic content as gallic acid. A good correlation between antioxidant activity and total phenol content in the extracts was found. The extraction yield, total phenol content and radical scavenging activity were increased and peroxide formation in vegetable oils were decreased with the graded solvent concentration of MeOH. The phenolic compound yield ranged from 2.45 to 11.35% and the total phenol content increased from 34.51 mg/g to 47.68 mg/g. DPPH scavenging activity increased from 56.32% to 90.37% as MeOH concentration increased. Compared to the control (70.45 meq/kg), 70% MeOH fraction with 47.68 mg/g phenolic compounds shows much stronger antioxidant ability (30.12 meq/kg) in vegetable oil system.

The effects of puffing on ginsenosides content and antioxidant activities of American and Canadian ginsengs, Panax quinquefolius, were investigated. American and Canadian ginsengs puffed at different pressures were extracted using 70% ethanol. Puffing formed a porous structure, inducing the efficient elution of internal compounds that resulted in significant increases in extraction yields and crude saponin content. The content of minor ginsenosides (Rg2, Rg3, compound K) increased with increasing puffing pressure, whereas that of major ginsenosides (Rg1, Re, Rf, Rb1, Rc, Rd) decreased, possibly due to their deglycosylation and pyrolysis. Furthermore, 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activity, total phenolic content, total flavonoid content, amount of Maillard reaction products, and acidic polysaccharides content increased with increasing puffing pressure, but 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity did not. There was no substantial difference in the results between puffed American and Canadian ginsengs. Consequently, these results suggest that puffing can be a promising novel technology for processing P. quinquefolius to achieve higher levels of minor ginsenosides and obtain value-added products.

Albizia myriophylla (ABZ) is a plant used in Thailand and Peninsular Malaysia for the treatment of diabetes mellitus type II. The antioxidant activities of ABZ have been suggested as one of the mechanisms for the observed beneficial effects. Hence, this study examined the phenolic, flavonoid, and saponin contents and antioxidant activity from methanol extract (ME) and its derived fractions hexane (HE), chloroform (CE), ethyl acetate (EAE), butanol (BE), and aqueous fraction of the bark of ABZ (AE). Amongst the extracts, EAE showed the highest total phenolic content of about 0.77 mg of gallic acid equivalent/g of extract (mg GAE/mg). However, the highest flavonoid content was detected in HE at 1.04 µg retinol equivalent ((RE)/g extract), while the saponin content was highest in CE at 1.1 µg diosgenin equivalent ((DE)/g extract). In 1,1-diphenyl-2-picrylhidrazyl (DPPH) radical scavenging test, EAE at 100 µg/mL had the highest percentage of inhibition of about 72.55%. 2-2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS radical scavenging test) showed BE had the highest percentage of inhibition at 100 µg/mL at 82.91%. Ferric reducing antioxidant power elucidated BE as having the highest percentage of inhibition which was 86.04% followed closely by ME at 85.90%. Thus, the different extracts of ABZ displayed various antioxidant capacity with probable free radical scavenging activity, which may be useful for the treatment of chronic inflammatory related metabolic diseases, such as diabetes and obesity.

Oenothera laciniata Hill is a perennial herb traditionally used to alleviate inflammatory complications. This study investigated the antioxidant and anti-melanogenic activities of O. laciniata. The methanolic extract (OLM) of O. laciniata and its different fractions, including ethyl acetate (OLEF), n-butanol (OLBF), and water (OLWF) fractions, were prepared. Antioxidant activities were evaluated by total phenolic content, the radical-scavenging effect on 2,2-diphenyl-1-picrylhydrazyl (DPPH•), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+•), and superoxide anion (O2−•), reducing capacity, and metal chelating ability. OLM and its fractions exhibited potent antioxidant activity in these in vitro assays, with a correlation between radical-scavenging activity and total phenolic content. OLM and its fractions inhibited the mushroom tyrosinase activity superior to the reference control, ascorbic acid. In B16-F10 melanoma cells, OLM and its fractions significantly decreased melanin production and tyrosinase activity. Mechanistic investigations revealed that OLM and its fractions inhibited tyrosinase and TRP-2 expressions via downregulating MITF and phosphorylated CREB and differentially inducing ERK or JNK phosphorylation. Additionally, OLM and its fractions caused no significant cytotoxicity towards B16-F10 or skin fibroblast cells at concentrations used in these cellular assays. These findings demonstrated the potential of O. laciniata extracts as the ideal skin protective agent with dual antioxidant and anti-melanogenic activities.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

Phenolic acid profiles and antioxidant activities of outer bran, coarse bran, and shorts from blue, black, and purple wheat were analyzed. Phenolic acids were mainly in the bound form in pigmented wheat bran fractions. Phenolic acid content decreased in the order of outer bran, coarse bran, and shorts for the three pigmented wheat varieties. HPLC analysis of phenolic extracts demonstrated that the bound form of phenolic acids contained more ferulic, isoferulic, and p-coumaric acids compared to their free counterparts. Among the three pigmented wheat varieties, the bran fractions from blue wheat contained higher bound phenolic acids than the other two pigmented wheat bran fractions, except for purple coarse bran. The blue wheat outer bran had the highest total bound phenolic acid of 3458.71 μg/g while the purple wheat shorts had the lowest of 1730.71 μg/g. The contribution of bound phenolic acids to the total phenolic content and antioxidant activity was significantly higher than that of free phenolic acids. Blue wheat bran fractions had the highest radical scavenging activity against DPPH∙ while those of purple wheat gained the highest ABTS∙+ scavenging activity. High correlations were observed between TPC and radical scavenging capacities for DPPH and ABTS (R2>0.85, P<0.05).

Phytochemical contents and antioxidant activities of fern (Asplenium ceterach L.) distributed in different altitudes (22 stations) were compared and the relationships between altitude and plant chemical contents were studied. The highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity was found in 22nd station (IC50 = 47.91 μg/ml) and the highest total phenolic content was found in 9th station (110.62 μgGAE/ml) whereas the maximum total flavonoid content was found in 20th station (232.67 μgCE/ml). High performance liquid chromatography (HPLC) analysis indicates that the maximum pterosin b (0.235 μg/ml), catechin (2.756 μg/ml) and quercetin (0.207 μg/ml) values were found in 21th station whereas the maximum chlorogenic acid (17.718 μg/ml) was obtained in 9th and caffeic acid (6,598 μg/ml) in 13th stations. It was observed that altitude is not potent alone, but it can be a factor in the occurrence of other ecological factors like soil properties, water, humidity, light and temperature.

In the honey industry, heat treatments are usually applied to maintain honey’s quality and shelf life. Heat treatment is used to avoid crystallisation and allow the easy use of honey, but treatment with heat might affect the antioxidant and antibacterial activities, which are the immediate health effects of honey. This study will determine the effect of heat treatment on Malaysian and Australian stingless bee honey (SBH) produced by the common bee species in both countries. Eighteen honey samples were subjected to heat at 45 °C, 55 °C and 65 °C for one hour and subsequently analysed for their total phenolic content (TPC), total flavonoid content (TFC), DPPH radical scavenging activity, ferric reducing antioxidant power (FRAP) and minimum inhibitory concentration (MIC). The results show that all samples had high TPC, TFC and antioxidant activities before the treatment. The heat treatments did not affect (p < 0.05) the TPC, TFC and antioxidant activities in most samples, but did inhibit the antibacterial activities consistently in most of the samples, regardless of the bee species and country of origin. This study also confirms a strong correlation between TPC and TFC with FRAP activities for the non-heated and heated honey samples (p < 0.05). Other heat-sensitive bioactive compounds in SBH should be measured to control the antibacterial properties present.
