Sea urchins can be used in medicine, has potential as a new type of antibiotic to developed in the pharmaceutical field, which is rich in bioactive compounds as a steroid, triterpenoids, saponins, and antioxidant properties. Conventional extraction generally takes a long time, is less environmentally friendly, and potentially triggers bioactive compound damage, so it needs alternative methods such as Ultrasound-Assisted Extraction (UAE). The extracting technology, including ultrasonic-assisted extraction (UAE) and solvent variation (ethyl acetate and methanol) of bioactive compounds from sea urchin (Diadema setosum) were optimized and compared. The purpose of this study was to study the application of UAE and solvent variation methods for sea urchin extraction from the Barrang Lompo Island in South Sulawesi. Comparing the maceration and ultrasonic assisted extraction methods. Optimization of extraction with UAE was carried out on the variable of extraction duration and type of solvent. Gas Chromatography-Mass Spectrometry results show that ultrasound-assisted extraction generally produces compounds of CHOLEST-5-EN-3-OL (3. BETA.), palmitic acid, 9-Octadecenoic acid (Z) -, methyl ester, stearic acid, oleic acid, flavonoids, phenols, pentadecanoic acid and batilol and streoid, which has a function as an antioxidant, anti-inflammatory, anti-tumor, anti-cancer agents and antibacterial. The results showed the best results using ultrasound-assisted extraction with a duration of 30 minutes and using solvent ethyl acetate. These results implied that extracts obtained by sonication showed the highest bioactive compounds and antioxidant activity, thus proving that this activity depends directly on the antibacterial properties.

Abstract The objective of this work was to evaluate the efficacy of ethyl acetate as a solvent in the extraction of macauba kernel oil (MKO) using ultrasonic-assisted extraction (UAE). It was shown that more MKO oil could be extracted with the use of larger amounts of solvent, higher temperatures and longer extraction times. Thus the maximum oil yield (40.61%) was obtained by UAE at 60 °C for 45 min, using a solvent to kernel ratio of 12 (mL g-1), obtaining a higher yield than that obtained with n-hexane under the same experimental conditions. UAE was favorable for this oil extraction (p < 0.05), presenting a yield close to that reported for classical extraction but with a shorter extraction time and smaller solvent volume. Lauric acid corresponded to ~44% of the MKO composition. The oils presented low free fatty acid contents (<0.80% wt), and the phytosterols, campesterol and β-sitosterol, were identified in the MKO with higher levels in the oil obtained by UAE.

Kersen (Muntinga calabura L) adalah pohon yang memiliki buah kecil dan manis. Tumbuhan ini dimanfaatkan antara lain sebagai  obat penurun panas, mengobati pembengkakan kelenjar prostat dan mengobati penyakit asam urat, selain itu juga dapat dimanfaatkan sebagai antiseptic, antioksidan, antimikroba, dan anti inflamasi. Berbagai macam metode ekstraksi telah dikembangkan dari yang konvensional ke metode modern, dimana salah satunya yaitu Microwave Assisted Extraction (MAE) dan Ultrasound Assisted Extraction (UAE). Penelitian ini bertujuan untuk menganalisa pengaruh metode ekstraksi yang dilakukan terhadap rendemen, menganalisa pengaruh rasio solid berbanding liquid serta waktu terhadap rendemen, dan menguji kadar flavonoid yang dihasilkan dari kedua metode tersebut. Percobaan dilakukan dengan variabel rasio-pelarut 1:5 – 1:25, sedangkan variabel waktu 5 menit – 25 menit. Hasil percobaan menunjukan bahwa ke dua variabel berpengaruh terhadap konsentrasi flavonoid hasil ekstraksi. Secara umum meningkat seiring kenaikan variabel sampai maksimum di titik tertentu, kemudian turun. Konsentrasi maksimum diperoleh pada variabel umpan-pelarut MAE sebesar 1:25 dengan kadar flavonoid 132,41 mg/ml dan rendemen sebesar 39%. Adapun waktu terbaik adalah 5 menit dengan kadar flavonoid 91,669 mg/ml dan rendemen sebesar 22,7%. Sedangkan pada proses UAE (Ultrasonic Assisted Extraction) dengan rasio umpan-pelarut maksimal sebesar 1:10 dengan kadar flavonoid 47,5899 mg/ml dan rendemen 26%. Sedangkan waktu terbaik  10 menit dengan kadar flavonoid 56,7769 mg/ml dan rendemen sebesar 19,65%. AbstracKersen  (Muntinga calabura L) is a tree with small and sweet fruit. The fruit is used, among others, as a febrifuge, treating swelling of the prostate gland and treating gout. Besides the fruit, its leaves also has potential as an antiseptic, antioxidant, antimicrobial, and anti-inflammatory. Various extraction methods have been developed from conventional to modern methods, one of which is Microwave Assisted Extraction (MAE) and Ultrasound Assisted Extraction (UAE). The research will extract the kersen leaves and aim to analyze the effect of the extraction method on the yield, the effect of the solid to liquid ratio and time on the yield, and the flavonoid content produced by both methods. The experiment was carried out with a solvent-ratio variable of 1:5 – 1:25, while the time variable was 5 – 25 minutes. The results of the experiment showed that the two variables had an effect on the concentration of flavonoids extracted. In general, it increases as the variables increase to a maximum point, then decreases. The maximum concentration obtained in the feed-solvent variable MAE was 1:25 with a flavonoid content of 132.41 mg/ml, with a yield of 39% and the best time was 5 minutes with a flavonoid content of 91.669 mg/ml, with a yield of 22.7%. While in the UAE Ultrasonic Assisted Extraction process with a maximum feed-solvent ratio of 1:10 with a flavonoid content 47.5899 mg/ml and yield  26%, while the best time of process was 10 minutes with  flavonoid content 56.7769 mg/ml and yield 19, 65%.Kata kunci: Flavonoid, Ekstrak Kersen, MAE, UA

Valorization of an artichoke by-product, rich in bioactive compounds, by ultrasound-assisted extraction, is proposed. The extraction yield curves of total phenolic content (TPC) and chlorogenic acid content (CAC) in 20% ethanol (v/v) with agitation (100 rpm) and ultrasound (200 and 335 W/L) were determined at 25, 40, and 60 °C. A mathematical model considering simultaneous diffusion and convection is proposed to simulate the extraction curves and to quantify both temperature and ultrasound power density effects in terms of the model parameters variation. The effective diffusion coefficient exhibited temperature dependence (72% increase for TPC from 25 °C to 60 °C), whereas the external mass transfer coefficient and the equilibrium extraction yield depended on both temperature (72% and 90% increases for TPC from 25 to 60 °C) and ultrasound power density (26 and 51% increases for TPC from 0 (agitation) to 335 W/L). The model allowed the accurate curves simulation, the average mean relative error being 5.3 ± 2.6%. Thus, the need of considering two resistances in series to satisfactorily simulate the extraction yield curves could be related to the diffusion of the bioactive compound from inside the vegetable cells toward the intercellular volume and from there, to the liquid phase.

AbstractThe grape pomace is a by-product from the industrial processing of grape juice, which can be used as a source of bioactive compounds. The aim of this study was to separate the phenolic compounds from grape pomace using an acidic aqueous solution with 2 % citric acid as a solvent, using both ultrasound-assisted extraction, with powers of 250, 350 and 450 W and times of 5, 10 and 15 min, and microwave-assisted extraction using powers of 600, 800 and 1,000 W and times of 5, 7 and 10 min. The results showed that for both methods of extraction, the contents of total phenolic compounds and antioxidant activity by ABTS and DPPH increased with time, and microwave at 1,000 W for 10 min corresponded to the best extraction condition. However, the contents of phenolic compounds and antioxidant activity were lower than exhaustive extraction using acidified methanol solution.

Blueberry and raspberry pomace are a rich source of bioactive compounds that have not been commercially utilized yet, and ultrasound-assisted technology can efficiently extract these compounds. Also, the use of water as a solvent added to the ultrasound-assisted technology improves this eco-friendly process. Therefore, an aqueous eco-friendly extraction, including extraction time and ultrasound presence or absence (conventional extraction) was performed in order to extract bioactive compounds from blueberry and raspberry pomace. Response parameters included levels of anthocyanins, phenolic compounds, and flavonoids, and antioxidant activity determined by DPPH, ABTS, and FRAP methods. Analysis of variance results indicated that ultrasound-assisted extraction for 45 min. was feasible to extract the bioactive compounds. The antioxidant content of the extract obtained by the ultrasound-assisted process was 1.4 times higher on average and the total phenolic concentration was 1.6 times higher (for blueberry 5.02 and for raspberry 2.53 mg gallic acid equivalent/g) compared with those obtained by the conventional process. Thus, the ultrasound-assisted extraction method can be a profitable alternative to extract bioactive compounds from blueberry and raspberry pomace, as it is energy efficient, requires fewer chemicals, and produces less effluent. This eco-friendly technology is therefore viable for food, nutraceutical, and cosmetic industries, and also for reducing food waste.

Two optimized methods for ultrasound-assisted extraction were evaluated for the extraction of two types of acai bioactive compounds: Total anthocyanins (TAs) and total phenolic compounds (TPCs). For the extraction optimization, a Box Behnken factorial design of different variables in the following intervals was used: Methanol-water (25%–75%) for solvent composition, temperatures between 10 and 70 °C, amplitude in the range between 30% and 70% of the maximum amplitude −200 W), extraction solvent pH (2–7), the ratio for sample-solvent (0.5 g:10 mL–0.5 g:20 mL), and cycle between 0.2 and 0.7 s. The extraction kinetics were studied using different periods between 5 and 30 min. TA and TPC were analyzed by UHPLC and the Folin–Ciocalteu method, respectively. Optimized conditions for TA were: 51% MeOH in water, 31 °C temperature, pH 6.38, cycle 0.7 s, 65% amplitude, and 0.5 g:10 mL of sample-solvent ratio. Optimized conditions for the TPC were: 49% MeOH in water, 41 °C temperature, pH 6.98, cycle 0.2 s, 30% amplitude, and 0.5 g:10 mL of sample-solvent ratio. Both methods presented a relative standard deviation below 5% in the precision study. The suitability of the methods was tested in real samples. It was confirmed that these methods are feasible for the extraction of the studied bioactive compounds from different açai matrices.

Mediterranean plants, such as fig and olive leaves, are well-known to exert beneficial effects in humans because of the presence of a wide range of bioactive compounds. However, scarce information regarding the impact of extraction methods, such as ultrasound and types of solvents, on their profile of antioxidant and anti-inflammatory compounds is provided. In addition, no information is available on the effects of extraction methods and solvents on the inhibition of pathogenic bacteria or promoting probiotic growth. In this scenario, this study was aimed to study the effects of ultrasound-assisted extraction (UAE) and solvent on the phenolic profile (Triple TOF-LC-MS/MS), antioxidant and anti-inflammatory compounds of olive and fig leaves. Results showed that UAE extracted more carotenoids compared to conventional extraction, while the conventional extraction impacted on higher flavonoids (olive leaves) and total phenolics (fig leaves). The antioxidant capacity of aqueous extract of fig leaves was three times higher than the extract obtained with ethanol for conventional extraction and four times higher for UAE. In general terms, hydroethanolic extracts presented the highest bacterial growth inhibition, and showed the highest anti-inflammatory activity. In conclusion, these side streams can be used as sources of bioactive compounds for further development of high-added-value products.
