Biodiversitas 22: 4261-4265. Captive breeding of parrots has been carried out in Indonesia to fulfill market demand for pets and support conservation efforts. However, there is limited information about Infectious Bronchitis Virus (IBV) in captive parrots. Therefore, this study examines IBV virus in psittacines and observes the pathological changes in inoculated chicken embryos. Cloacal swabs were obtained from 31 psittacine birds consisting of 15 sulfur-crested cockatoos (Cacatua sulphurea), 10 palm cockatoos (Probosciger aterrimus), two blue and yellow macaws (Ara ararauna), two green-winged macaws (Ara chloropterus), and two scarlet macaws (Ara macao). Swabs were also injected for 9 days to obtain specific pathogen-free (SPF) embryonated chicken eggs, and allantoic fluids were collected after 7 days. The residual cloacal swabs and allantoic fluid samples were tested for IBV by reverse transcription polymerase chain reaction (RT-PCR) targeting spike 1 glycoprotein (S1) and nucleoprotein (N) gene by primer set XCE2+/XCE2- and IBVN+/IBVN-, while the harvested inoculated embryos were examined for histopathological lesions by hematoxylin-eosin staining. Three and one allantoic fluid of C. sulphurea and P. aterrimus respectively were found positive for IBV by primer set IBVN+IBVN-. However, there was no significant difference in the inoculated embryo's gross anatomy, and the slight histopathological changes observed were not caused by the virus. Therefore, it is unclear whether the IB-like virus causes clinical disease manifestation in both Psittacine species or the chicken embryo. This is the first study carried out on IB-like viruses for both species.

In the last 5 years, frequent outbreaks of infectious bronchitis virus (IBV) are observed in both broiler and layer chicken flocks in the Kingdom of Saudi Arabia (KSA) in spite of extensive usage of vaccines. The IBV is a widespread avian coronavirus affecting both vaccinated and unvaccinated chicken flocks and is attributed to significant economic losses, around the globe. In the present study, 58 (n = 58) samples were collected from four different commercial poultry flocks from 8 KSA districts during 2019. A total of nine positive isolates (9/58; 15.5%), based on real-time reverse transcriptase PCR targeting nucleocapsid (N) gene, were used for further genetic characterization and evolutionary analysis. Genetic characterization of the partial spike (S1) gene revealed the clustering of the reported isolates into three different genotypes, whereas four additional isolates were grouped within 4/91 genotype, two isolates within IS/885 genotype, one isolate was closely related to IS/1494/06, and two isolates were grouped within classic serotype (vaccine-like strains). Phylodynamic revealed clustering of four isolated viruses within GI-13 lineage, three isolates within GI-23 lineage, and two isolates within GI-1 lineage. Results indicate that there are high evolutionary distances between the newly identified IBV strains in this study and the commercially used vaccines (GI-1), suggesting that IBV strains circulating in the KSA are under constant evolutionary pressures. Selective pressure biostatistics analyses consistently demonstrate the presence of a higher positive score which highlights the role of natural selection, a mechanism of virus evolution on sites located on the protein surface, within or nearby domains involved in viral attachment or related functions. Recombination analysis revealed emergence of two isolates through recombination events resulting in new recombinant viruses. Taken together, these finding demonstrate the genetic and evolutionary insights into the currently circulating IBV genotypes in KSA, which could help to better understand the origin, spread, and evolution of infectious bronchitis viruses, and to ascertain the importance of disease monitoring as well as re-evaluation for the currently used vaccines and vaccination programs.

Coronavirus sub-genomic mRNA (sgmRNA) synthesis occurs via a process of discontinuous transcription involving complementary transcription regulatory sequences (TRSs), one (TRS-L) encompassing the leader sequence of the 5′ untranslated region (UTR), and the other upstream of each structural and accessory gene (TRS-B). Several coronaviruses have an ORF located between the N gene and the 3′-UTR, an area previously thought to be non-coding in the Gammacoronavirus infectious bronchitis virus (IBV) due to a lack of a canonical TRS-B. Here, we identify a non-canonical TRS-B allowing for a novel sgmRNA relating to this ORF to be produced in several strains of IBV: Beaudette, CR88, H120, D1466, Italy-02 and QX. Interestingly, the potential protein produced by this ORF is prematurely truncated in the Beaudette strain. A single nucleotide deletion was made in the Beaudette strain allowing for the generation of a recombinant IBV (rIBV) that had the potential to express a full-length protein. Assessment of this rIBV in vitro demonstrated that restoration of the full-length potential protein had no effect on viral replication. Further assessment of the Beaudette-derived RNA identified a second non-canonically transcribed sgmRNA located within gene 2. Deep sequencing analysis of allantoic fluid from Beaudette-infected embryonated eggs confirmed the presence of both the newly identified non-canonically transcribed sgmRNAs and highlighted the potential for further yet unidentified sgmRNAs. This HiSeq data, alongside the confirmation of non-canonically transcribed sgmRNAs, indicates the potential of the coronavirus genome to encode a larger repertoire of genes than has currently been identified.

Background and Aim: Avian coronavirus has a wide range of hosts, from chickens and turkeys to wild birds. This virus causes an economically and, possibly, environmentally, important loss in the poultry industry. Therefore, research into the avian coronavirus in various species of birds is required. The Eclectus parrot (Eclectus roratus) is an endemic bird to Indonesia and Northern Australia and often kept as pets. At present, there has been limited information about avian coronavirus infection among birds. This study aimed to determine the presence of and to characterize avian coronavirus isolated from Eclectus parrots in Indonesia. Materials and Methods: Cloacal swab samples were taken from 10 healthy Eclectus parrots (E. roratus). Each isolate was propagated into specific pathogen-free embryonated chicken eggs. The presence of avian coronavirus was determined using three sets of primers targeting the 3' untranslated region (3'-UTR) of avian coronavirus (UTR41+/11–), the N gene of the infectious bronchitis virus (IBVN+/–), and the S1 gene of the IBV (XCE2+/XCE2–). The infectious bronchitis vaccine strain H120 was used as a positive control. Resulting positive bands were sequenced for the S1 gene. Results: None of the isolates was positive for the 3'-UTR, four isolates were positive for the N gene of infectious bronchitis, and two isolates were positive for the S1 gene of the IBV. However, only one isolate (parrot/Indonesia/BX9/16) was sequenced for the partial S1 gene with primers XCE2+/XCE2–. The partial nucleotide sequence of this isolate showed 100% homology with the IBV GI-13 lineage, specifically with a field isolate of the 4/91 variant 1 Israel and the 4/91 vaccine on the hypervariable region 3 site of the S1 gene. Conclusion: An IB-like avian coronavirus was isolated from healthy Eclectus parrots. Our results indicate that IBV has a wide range of hosts, which prompt the need to understand the interspecies connection of this virus better.

Abstract Background Cattle are intermediate hosts of six Sarcocystis species, among which Sarcocystis hominis and Sarcocystis heydorni can infect humans through the consumption of raw or undercooked meat. In addition to the zoonotic potential, there is increasing interest in these protozoa because of the evidence supporting the role of Sarcocystis spp. in the occurrence of bovine eosinophilic myositis (BEM), a specific inflammatory myopathy which leads to carcass condemnation and considerable economic losses. Actually, all the prevalence studies carried out on cattle in Italy have been based on either morphological or 18S rDNA-based molecular techniques, most likely leading to misidentification of closely related species. Therefore, there is a strong need for new data on the prevalence of the different Sarcocystis spp. in cattle in Italy and their association with bovine eosinophilic myositis. Methods To reach our aim, individual striated muscle samples from BEM condemned carcasses (N = 54) and diaphragm muscle samples from randomly sampled carcasses (N = 59) were obtained from Northwest Italy slaughterhouses. Genomic DNA was extracted and analyzed by multiplex-PCR targeting 18S rDNA and cox1 genes. PCR products amplified using the genus-specific primer set in absence of the specific fragment for S. hirsuta, S. cruzi, S. hominis or S. bovifelis were sequenced to achieve species identification. Results Sarcocystis DNA was detected in 67.8% of the samples from slaughter cattle and in 90.7% of the samples from BEM condemned carcasses. S. cruzi was identified as the most prevalent species in slaughter cattle (61%), followed by S. bovifelis (10.2%), S. hominis (8.5%) and S. hirsuta (1.7%). Notably, among the different Sarcocystis spp. detected, the presence of S. bovifelis and S. hominis was significantly higher in samples isolated from BEM condemned carcasses (46.3% and 40.7% respectively), while there was no statistically significant difference between the presence of S. cruzi or S. hirsuta in BEM condemned carcasses (42.6% and 1.8%, respectively) and randomly sampled carcasses. Furthermore, DNA sequence analysis revealed the presence of a putative new species in two carcasses. Conclusions Our study contributes to updating the data on the prevalence of the different Sarcocystis spp. in cattle in Italy, highlighting the presence of three Sarcocystis spp., S. cruzi, S. hominis and S. bovifelis, in BEM lesions and allowing us to speculate on the possible role of S. hominis and S. bovifelis as the major sarcosporidian species involved in bovine eosinophilic myositis. The desire to shore up the shortfall in protein supply, the increased awareness of the importance of cholesterol free animal protein, like guineafowl, and the need to conserve their wild genes, have necessitated studies on improved captive breeding of feral helmeted guineafowl. Hence, this study was aimed at determining the effects of ecological zones and storage systems on the guineafowl eggs. Guineafowl eggs (n=214), were collected from identified and monitored-nests within the Kainji Lake National Park (KLNP) and Old Oyo National Park (OONP). Out of 91 eggs collected from KLNP, 32 and 38 were stored at room temperature (RT-21-25°C) and refrigerator (RF-17-20°C), respectively for five days prior to incubation while 21 eggs were not stored (NS-27-29°C), and out of 123 collected from OONP, 70 and 19 were stored for five days prior to incubation at (RT-21-25°C) and (RF-17-20°C), respectively while 34 eggs were (NS- 27-29°C). Prior to incubation, eggs were weighed, the height and width were measured. The process was repeated after incubation for unhatched eggs. Candling was done three days before hatching at day 29 of egg incubation. Embryo status of unhatched eggs was determined by cracking the eggs. Descriptive and inferential statistics were used to analyse the data. The NS eggs from Kainji (7.60%) and Oyo (2.01%) had the lowest percentage shrinkage in weight across the three storage systems. The order was reversed in height with RF eggs from Kainji (0.96%) and Oyo (0.46%) having lowest. The least shrinkage in width of eggs from Oyo was recorded in the RF eggs (0.00%) and in NS (0.59%) from Kainji. Eggs candling showed that presumed fertile (opaque) was highest (69.10%) in NS eggs followed by RT (45.00%). There was no significant difference (p>0.05) between the hatchability of eggs from the two parks though hatchability (7.60%) of Kainji eggs was higher than those of Oyo (7.40%). Further check on fertilization after incubation showed that RT (37.50%) eggs from Kainji were fertilized but were unable to hatch alive so also was RF eggs (21.10%). The study showed that the eggs sizes vary with ecozones while size of the eggs and storage systems affects hatchability and survival of feral helmeted guineafowl eggs in captivity. Background: National Family Health Survey (NFHS-4 2015-2016) documented the prevalence of anaemia as overall more than three-quarters (76 percent) of children. Anaemia is the most common Haematological disease of the paediatric age group. Anaemia is the highest prevalence in developing countries. The population differences in the prevalence of anaemia are explained by environmental factors affecting nutrition, chief among these are economic status, ethnic customs & geographic considerations. Furthermore, there is very limited information on prevalence of Iron and B12 deficiencies among children belonging to different communities with culturally defined eating habits. In the present study carried out to compare the Serum Iron & Vitamin B12 in children of different communities in the South Indian state of Telangana. Material & Methods: In this population based cross sectional observational study was conducted on the department of paediatrics in the Chalmeda Anandrao Institute of Medical Sciences, Karimnagar, Telangana, during the period from 1st January 2020 to till reached the sample size. The study was conducted with the approval from the institutional review and ethical committees. In this study includes children were of the age between 5 to 18 years with the 3 different communities like, Hindu, Muslim & others community. Results: In the above table we shows that the Others community of age is 12.85 ± 2.65 years, Hindu community of age is 12.76 ± 3.46 years & Muslim community of age is 14.96 ± 2.00 years. In our study, the prevalence of Serum Iron was found to be 21.7% (26 out of 120) & prevalence of Vitamin B12 was found to be 50.0% (60 out of 120). Conclusion: The overall prevalence of anaemia (low Haemoglobin) was found to be 43.33%. There was no significant difference between the prevalence of anaemia in 3 different communities. Background: Currently anatomy training and assessment is seriously disrupted in COVID-19 Pandemic lockdown. New challenges of anatomy curriculum and assessment method development are being attempted based upon resources, expertise and technical skills of educators. This study aims at creating a standard operation procedure (SOP) for conducting gross anatomy assessment, finding statistical relevance and discusses viewpoints of students reflected in feedback. Materials and methods: a retrospective cross sectional study was conducted on 250 first year students at GSVM Medical College Kanpur India. Students were briefed about SOP and were given choice to opt for either of three interview modalities of assessment tool [Viz. Telephonic Interview (GROUP T), Whatsapp© video call (GROUP W) and Zoom© meetings (GROUP Z)] and submit subjective feedbacks to assessor regarding new assessment format. The marks obtained as response distribution were subjected to descriptive statistics. One way ANOVA was applied to test any difference in means at (P<0.05) followed by post-hoc pair wise test amongst groups corrected to Bonferroni adjusted alpha level of [P<0.0167] per test. Results: Among 97.2% respondent ANOVA test result pointed that the types of interview pattern preferred had no significant impact on obtained score [F (2,240) = 2.80, p = .062], post-hoc test further confirmed that there was no significant difference between groups. Conclusion: the assessor and students both need to evolve and adapt to paradigm changes. Student’s feedback will serve to tailor the assessment tool by refining validity, consistency, reliability and interaction dynamics of assessment designs. Aim: We report the diagnostic evaluation of a confirmatory reverse transcription-PCR (RT-PCR) kit targeting the Middle East respiratory syndrome coronavirus (MERS-CoV) N gene. Material & methods: 33 patient samples from two collections sites in Riyadh, Saudi Arabia, which were pre-characterized via real-time RT-PCR targeting MERS-CoV orf1a and upE, and were tested using the MERS-CoV N gene, as a confirmatory assay. This diagnostic procedure follows a two-step diagnostics scheme, recommended by the WHO. Results: 18/33 samples tested positive, 11/33 tested negative for MERS-CoV RNA and 2/33 showed uncertain results. Conclusion: The results suggest, that the RealStar® MERS-CoV (N gene) RT-PCR kit 1.0 can be considered a suitable and reliable confirmatory assay in combination with the RealStar MERS-CoV RT-PCR kit 1.0 according to the diagnostic scheme recommended by WHO.

BackgroundDifferent approaches are used to treat resectable tumours in patients having adenocarcinoma at the oesophagogastrointestinal junction (EGJ) or in the stomach. However, there is limited information about treatment efficacy for patients at metastatic stage. A recent molecular analysis of upper gastrointestinal tract adenocarcinoma revealed that the anatomical location can influence the molecular backgrounds of tumours. This study sought to elucidate whether different therapeutic approaches should be used for EGJ tumours relative to those in the stomach.MethodsThis retrospective cohort study was conducted at a single institute in Japan. Patients having metastatic or recurrent adenocarcinoma in the EGJ or stomach who underwent platinum doublet chemotherapy between January 2007 and August 2014 were enrolled. Patients in the EGJ tumour group had tumours having an epicentre within 2 cm proximal or 5 cm distal to the estimated anatomical EGJ and cardia.ResultsAmong 378 consecutively enrolled patients, 61 were grouped into the EGJ group and the remainder comprised the stomach group. The EGJ group had more men and lower incidence of diffuse type and Borrmann type IV tumours and peritoneum metastasis compared with the stomach group. The median overall survival of patients in the EGJ and stomach groups was similar (17.3 months (95% CI 13.5 to 23.2) vs 14.5 months (95% CI 13.3 to 16.4)). No statistically significant difference was observed in progression-free survival. Although the overall postprogression survival differed significantly between the EGJ and stomach groups (8.2 months (95% CI 5.7 to 12.7) vs 7.1 months (95% CI 6.1 to 7.8)), on grouping patients by histological type, the two groups exhibited similar postprogression survival. Multivariate analysis demonstrated that diffuse-type histology, higher serum CA19-9 levels and neutrophil to lymphocyte ratios were independent poor prognostic factors.ConclusionsDifferent clinicopathological features of EGJ adenocarcinoma were not associated with clinical outcomes of platinum doublet chemotherapy. Histological subtype rather than anatomical location has more significance for treatment decisions for advanced gastric cancers.
